Discovering anomalous hybridization kinetics on DNA nanostructures using single-molecule fluorescence microscopy.

DNA nanostructures are finding diverse applications as scaffolds for molecular organization. In general, components such as nucleic acids, proteins, and nanoparticles are attached to addressable DNA nanostructures via hybridization, and there is interest in exploiting hybridization for localized computation on DNA nanostructures. This report details two fluorescence microscopy methods, single-particle fluorescence resonance energy transfer (spFRET) and DNA-PAINT (points accumulation for imaging in nanoscale topography), that have been successfully used to detect anomalies of hybridization reac…

Study of coupling loss on strongly-coupled, ultra compact microring resonators.

Small-radius microring resonators with large free spectral range (FSR) are of great interest for optical communication and optical interconnect applications. The resonator loss of a waveguide-coupled ring resonator, if the gap width between the microring and the bus waveguide is extremely small, can be significantly influenced by the coupling loss which corresponds to the microring operated in a strong coupling regime. This effect is particularly prominent for small radius microrings. We have studied the coupling loss with respect to the gap width on a waveguide-coupled microring both experime

A facile strategy to fabricate thermoresponsive polymer functionalized CdTe/ZnS quantum dots: assemblies and optical properties.

Novel thermoresponsive CdTe/ZnS quantum dots (QDs) decorated with a copolymer ligand (CPL) containing 8-hydroxyquinoline and NIPAM units are … n (517 nm) of the CPL complex formed on the QD surface. For QDs-CPL systems, both Förster resonance energy transfer (FRET) and a high rate of photoinduced electron transfer (PET) are simultaneous, the latter mainly contributing to PL quenching. The thermoresponsive QDs/CPL assemblies also exhibit dual reversible PL properties between the inherent emission of QDs …

Assessing the mechanical energy costs of various tissue reshaping mechanisms.

Early-stage embryos must reshape the tissues of which they are made into organs and other life-sustaining structures; and if non-mammalian embryos fail to complete these tasks before the energy provided by their yolk runs out, they die. The aim of this study is to use a cell-level computational model to investigate the energetic cost of a variety of mechanisms that can drive an in-plane reshaping pattern known as convergent extension--a motif in which a tissue narrows in one in-plane direction and expands in another. Mechanisms considered include oriented lamellipodia, directed mitosis, stress

Impact of increasing current drain in a pacemaker operating during activation of the elective replacement indicator.

The occurrence of a 13.8 s episode of ventricular asystole in a patient whose VVIR pacemaker displayed the elective replacement indicator (ERI) is reported. Increasing battery current drain by VARIO testing and programming of the emergency VVI mode markedly increased battery current drain with a resultant decrease in the battery voltage below the pacing threshold. The prolonged lack of capture occurred because the lowered battery voltage could not return instantaneously to its previous level after the demand for a higher battery current drain had ceased. Rather, the battery voltage increased p

Segment-interaction in sprint start: Analysis of 3D angular velocity and kinetic energy in elite sprinters.

The aim of the present study was to measure during a sprint start the joint angular velocity and the kinetic energy of the different segments in elite sprinters. This was performed using a 3D kinematic analysis of the whole body. Eight elite sprinters (10.30+/-0.14s 100 m time), equipped with 63 passive reflective markers, realised four maximal 10 m sprints start on an indoor track. An opto-electronic Motion Analysis system consisting of 12 digital cameras (250 Hz) was used to collect the 3D marker trajectories. During the pushing phase on the blocks, the 3D angular velocity vector and its nor

Violet ZnSe/ZnS as an alternative to green CdSe/ZnS in nanocrystal-fluorescent protein FRET systems.

Fluorescent proteins from the green fluorescent protein family strongly interact with CdSe/ZnS and ZnSe/ZnS nanocrystals at neutral pH. Green emitting CdSe/ZnS nanocrystals and red emitting fluorescent protein dTomato constitute a 72% efficiency FRET system with the largest alteration of the overall photoluminescence profile, following complex formation, observed so far. The substitution of ZnSe/ZnS for CdSe/ZnS nanocrystals as energy donors enabled the use of a green fluorescent protein, GFP5, as energy acceptor. Violet emitting ZnSe/ZnS nanocrystals and green GFP5 constitute a system with 43

Cytogenetic effects of high-energy iron ions: dependence on shielding thickness and material.

We report results for chromosomal aberrations in human peripheral blood lymphocytes after they were exposed to high-energy iron ions with or without shielding at the HIMAC, AGS and NSRL accelerators. Isolated lymphocytes were exposed to iron ions with energies between 200 and 5000 MeV/nucleon in the 0.1-1-Gy dose range. Shielding materials consisted of polyethylene, lucite (PMMA), carbon, aluminum and lead, with mass thickness ranging from 2 to 30 g/cm2. After exposure, lymphocytes were stimulated to grow in vitro, and chromosomes were prematurely condensed using a phosphatase inhibitor (calyc

Effect of push frequency and strategy variations on economy and perceived exertion during wheelchair propulsion.

Wheelchair locomotion is a cyclical activity and participants are free to select any push frequency-propulsion strategy combination that suits their needs at a given power output. The aim of the study was to examine the physiological effects of varying push frequency and strategy on pushing economy. Twelve male, able-bodied participants completed four, randomly assigned, 5-min bouts of submaximal exercise at 32 W on a wheelchair ergometer. Each bout of exercise combined two different push frequencies (40 and 70 push min(-1)), with one of two different push strategies [synchronous (SYN): both a

The F value cannot be ruled out as a chromosomal fingerprint of radiation quality.

The F value, the ratio of inter- to intrachromosomal interchanges, as a biomarker for densely ionizing radiation which was proposed by Brenner and Sachs (Radiat. Res. 140, 134-142, 1994) has been a matter of repeated discussion. We examined our experimental data on radiation-induced chromosome aberrations in human peripheral blood lymphocytes for the F value as measured by the ratio of dicentrics to centric rings. Because of the rarity of aberrations, the F value showed a considerable variability, with a large error range particularly in the low-dose range of low-LET radiation. However, the da

Chromosome aberrations induced in human lymphocytes by 16.5 MeV protons.

In a track segment irradiation experiment with 16.5 MeV protons the induction of dicentrics was studied in human T-lymphocytes. The dose-response relationship was linear quadratic with estimated parameters alpha = (0.44 +/- 0.07) x 10(-1) Gy-1 and beta = (1.95 +/- 0.30) x 10(-2) Gy-2. With respect to X-rays a limiting RBE of 1.1-1.2 exists. The present findings are compared with data from other laboratories obtained with 4.9 and 8.7 MeV protons. It was found that the data do not fit theoretical predictions on a proportional relationship between alpha values and LET. Using the microdosimetric q

Three-dimensional network of creatine metabolism: From intracellular energy shuttle to systemic metabolic regulatory switch.

BACKGROUND: Creatine serves as an intracellular shuttle for high-energy phosphate bonds, enabling rapid ATP transfer from energy-producing to energy-consuming cellular compartments. In skeletal muscle, creatine coordinates energy distribution among mitochondrial oxidative phosphorylation, glycolysis, and the phosphagen system. Consequently, creatine supplementation acutely enhances muscular performance and is widely utilized as an ergogenic aid in power-based sports. Recent studies demonstrate that enhanced creatine metabolism in adipose tissue promotes brown adipocyte renewal and boosts energ

Intercellular mitochondrial transfer in the brain, a new perspective for targeted treatment of central nervous system diseases.

AIM: Mitochondria is one of the important organelles involved in cell energy metabolism and regulation and also play a key regulatory role in abnormal cell processes such as cell stress, cell damage, and cell canceration. Recent studies have shown that mitochondria can be transferred between cells in different ways and participate in the occurrence and development of many central nervous system diseases. We aim to review the mechanism of mitochondrial transfer in the progress of central nervous system diseases and the possibility of targeted therapy. METHODS: The PubMed databank, the China Na

Molecular assembly of artificial photosynthetic antenna core complex on an amino-terminated ITO electrode.

Bacterial photosynthetic membrane proteins, light-harvesting antenna complex (LH1), reaction center (RC), and their combined 'core' complex (LH1-RC) are functional elements in the primary photosynthetic events, i.e., capturing and transferring light energy and subsequent charge separation. These photosynthetic units (PSUs) isolated from Rhodospirillum rubrum (Rs. rubrum) were assembled onto an ITO electrode modified with 3-aminopropyltriethoxysilane (APS-ITO). The near IR absorption spectra of PSUs on the assembled electrodes were identical to those of solutions, indicating that the LH1 and LH

Direct Z-scheme ZnI2/GeTe van der Waals heterostructure for photocatalytic overall water splitting.

The rising global energy crisis and environmental degradation highlight the urgent demand for efficient photocatalytic materials for solar-driven hydrogen production through water splitting. Two-dimensional (2D) heterostructures are attractive because interfacial coupling can combine efficient charge management with strong redox capability. In this study, first-principles calculations are used to investigate the structural, electronic, optical, and photocatalytic properties of a ZnI2/GeTe van der Waals heterostructure. The optimized heterostructure exhibits indirect band gaps of 1.23 eV at the

Facile Fabrication of Nickel Supported on Reduced Graphene Oxide Composite for Oxygen Reduction Reaction.

Due to the depletion of fossil fuels, the demand for renewable energy has increased, thus stimulating the development of novel materials for energy conversion devices such as fuel cells. In this work, nickel nanoparticles loaded on reduced graphene oxide (Ni/rGO) with small size and good dispersibility were successfully prepared by controlling the pyrolysis temperature of the precursor at 450 °C, assisted by a microwave-assisted hydrothermal method, and exhibited enhanced electrocatalytic activity towards oxygen reduction reaction (ORR). Additionally, the electron enrichment on Ni NPs was due
