Differential sensitivity of human neuroblastoma cell lines to ethanol: correlations with their proliferative responses to mitogenic growth factors and expression of growth factor receptors.

Early ethanol exposure depletes neurons in the developing nervous system, however the effects on neuronal precursors are not homogeneous. Some cells are more susceptible to ethanol toxicity than others. Growth factors are important mitogens for neuronal precursors. We tested the hypothesis that the differential sensitivity of neuronal precursors to ethanol is determined by their responses to growth factors using an in vitro model (SH-SY5Y, SK-N-SH, and IMR32 neuroblastoma cells) of neuronal precursors. The three cell lines were raised in a medium containing 10% or 0% fetal calf serum. Cells we

Comparative In Vitro Evaluation of Early Cell Responses of Porous and Nonporous Polycaprolactone Electrospun Nanofibrous Membrane for Guided Tissue Regeneration.

OBJECTIVE: The production of porous polycaprolactone (PCL) electrospun fibers has already been reported in the literature. However, most of the studies reported the production of porous fibers on a micrometer scale. Hence, the main aim of this study is to (1) fabricate PCL membranes with secondary nanoporous fibers to improve the attachment, spreading, and proliferation of cells, and subsequent extracellular matrix deposition and (2) evaluate its biocompatibility on different cell types in comparison to a nonporous one. MATERIALS AND METHODS: Secondary nanopores were introduced by adjusting h

Early-Onset Preeclampsia-Derived hUC-MSCs Exhibit Accelerated Initial Culture Kinetics Without Sustained Growth Advantage: A Comparative In Vitro Study.

Preeclampsia (PE) is a pregnancy-related disorder associated with significant maternal and fetal morbidity, and intrauterine conditions may influence the biological behavior of human umbilical cord (UC)-derived mesenchymal stem cells (hUC-MSCs). However, comparative data between early-onset PE (EOPE) and late-onset PE (LOPE) remain limited. This study aimed to compare the in vitro growth kinetics of hUC-MSCs derived from pregnancies complicated by EOPE and LOPE. In this comparative in vitro study, UC samples (n = 66) were classified into three groups according to the maternal clinical conditio

Demethylnobiletin ameliorates cerebral ischemia-reperfusion injury in rats through Nrf2/HO-1 signaling pathway.

BACKGROUND: Demethylnobiletin (DN), with a variety of biological activities, is a polymethoxy-flavanone (PMF) found in citrus. In the present study, we explored the biological activities and potential mechanism of DN to improve cerebral ischemia reperfusion injury (CIRI) in rats, and identified DN as a novel neuroprotective agent for patients with ischemic brain injury. METHODS: Rat CIRI models were established via middle cerebral artery occlusion (MCAO). Primary nerve cells were isolated and cultured in fetal rat cerebral cortex in vitro, and oxygen-glucose deprivation/reperfusion (OGD/R) mo

Uptake and transport of pullulan acetate nanoparticles in the BeWo b30 placental barrier cell model.

INTRODUCTION: Nanomedicine has shown a great potential in perinatal medicine because of its characteristics of sustained, controlled release and targeting ability; on the other hand, it may also lead to unexpected toxicities such as embryotoxicity and even malformation after crossing the placental barrier, but data concerning transplacental transport are scarce. Pullulan acetate (PA) nanoparticles (NPs) are a promising nanocarrier derived from natural polysaccharide; however, their transplacental transport ability and mechanism are unknown. MATERIALS AND METHODS: In this study, fluorescein is

Toxoplasma gondii antigens: recovery analysis of tachyzoites cultivated in Vero cell maintained in serum free medium.

Vero cells have been used successfully in Toxoplasma gondii maintenance. Medium supplementation for culture cells with fetal bovine serum is necessary for cellular growth. However, serum in these cultures presents disadvantages, such as the potential to induce hypersensitivity, variability of serum batches, possible presence of contaminants, and the high cost of good quality serum. Culture media formulated without any animal derived components, designed for serum-free growth of cell lines have been used successfully for different virus replication. The advantages of protozoan parasite growth i

Chronic treatment with P2-purinergic receptor agonists induces phenotypic modulation of the HL-60 and U937 human myelogenous leukemia cell lines.

In previous studies we have demonstrated that extracellular ATP (and UTP), acting through P2-purinergic receptors, can stimulate the inositol phospholipid signaling system in neutrophils and monocytes, as well as in neutrophil/monocyte progenitor cells. In this study we have examined the ability of extracellular nucleotides to modulate the phenotype of myelomonocytic progenitor cells. As model systems, we utilized the established HL-60 promyelocytic and U937 promonocytic human cell lines which were cultured in the continuous presence of nucleotides known to be potent agonists for P2-purinergic

Activation of artemisinin and heme degradation in Leishmania tarentolae promastigotes: A possible link.

Endoperoxides (EPs) appear to be promising drug candidates against protozoal diseases, including malaria and leishmaniasis. Previous studies have shown that these drugs need an intracellular activation to exert their pharmacological potential. The efficiency of these drugs is linked to the extensive iron demand of these intracellular protozoal parasites. An essential step of the activation mechanism of these drugs is the formation of radicals in Leishmania. Iron is a known trigger for intracellular radical formation. However, the activation of EPs by low molecular iron or by heme iron may stro

The need for implementation of first trimester screening for preeclampsia and fetal growth restriction in low resource settings.

BACKGROUND: Preeclampsia [PE] and fetal growth restriction [FGR] is a major cause of perinatal morbidity in both developed and developing countries but the disease leaves a severe impact in developing countries, due to the late presentation of cases where prevention and treatment becomes impossible. Routine antenatal ultrasound and health checkups in periphery are usually done in first trimester for dating and viability scan, in midtrimester for anomaly scan and in third trimester for safe confinement. Underlying disorder of deep placentation which is unidentified can lead to increased materna

Complexity of CNC transcription factors as revealed by gene targeting of the Nrf3 locus.

Cap'n'collar (CNC) family basic leucine zipper transcription factors play crucial roles in the regulation of mammalian gene expression and development. To determine the in vivo function of the CNC protein Nrf3 (NF-E2-related factor 3), we generated mice deficient in this transcription factor. We performed targeted disruption of two Nrf3 exons coding for CNC homology, basic DNA-binding, and leucine zipper dimerization domains. Nrf3 null mice developed normally and revealed no obvious phenotypic differences compared to wild-type animals. Nrf3(-/-) mice were fertile, and gross anatomy as well as

The Effect of Maternal Near-Miss on Adverse Perinatal Outcomes Among Women Admitted to Hospitals of Gurage Zone, Southwest Ethiopia: A Prospective Cohort Study.

INTRODUCTION: Perinatal outcomes refer to events affecting a newborn after fetal viability, typically from 28 weeks of gestation through the first seven days of life. Adverse perinatal outcomes include stillbirth, low birth weight, preterm birth, early neonatal death, perinatal mortality, admission to a neonatal intensive care unit, and birth asphyxia. Despite substantial reductions in under-five mortality over recent decades, neonatal mortality rates have shown minimal improvement in sub-Saharan Africa. OBJECTIVE: This study aimed to assess the effect of maternal near-miss on adverse perinat…

Feasibility and safety of cellular therapy for in-utero repair of myelomeningocele (CuRe Trial): a first-in-human, phase 1, single-arm study.

BACKGROUND: The Management of Myelomeningocele Study (MOMS) trial established the benefit of in-utero repair of myelomeningocele, with a decreased need for ventriculoperitoneal shunt placement. However, although there was some improvement of motor function, over half of the patients were unable to ambulate independently. Live placenta-derived mesenchymal stem cells (PMSCs) seeded on an extracellular matrix have shown promise in rescuing neurological function in the fetal ovine model of myelomeningocele. We aimed to evaluate the safety of this novel, living, stem cell product to augment the pre
