Bone metabolic activity in hyperostosis cranialis interna measured with 18F-fluoride PET.

PURPOSE: (18)F-Fluoride PET/CT is a relatively undervalued diagnostic test to measure bone metabolism in bone diseases. Hyperostosis cranialis interna (HCI) is a (hereditary) bone disease characterised by endosteal hyperostosis and osteosclerosis of the skull and the skull base. Bone overgrowth causes entrapment and dysfunction of several cranial nerves. The aim of this study is to compare standardised uptake values (SUVs) at different sites in order to quantify bone metabolism in the affected anatomical regions in HCI patients. METHODS: Nine affected family members, seven non-affected family

Effect of root surface treatment with propolis and fluoride in delayed tooth replantation in rats.

Replantation is an acceptable option for treatment of an avulsed permanent tooth. Nevertheless, an extended extraoral period damages the periodontal ligament and results in external root resorption. The purpose of this study was to assess by histologic and histometric analysis, the influence of propolis 15% (natural resinous substance collected by Apis mellifera bees from various plants) and the fluoride solution used as root surface treatment on the healing process after delayed tooth replantation. Thirty Wistar (Rattus norvegicus albinus) rats were submitted to extraction of their upper righ

Skeletal fluorosis from the point of view of an occupational exposure to fluorides in former Czechoslovakia.

Electrolytic production of aluminium in former Czechoslovakia started in the year 1953 in the Žiar valley in the central Slovakia. However, till 1995 the hygienic conditions for health protection were not met in the factory. It underwent a reconstruction afterwards. The authors demonstrate cases of occupational skeletal fluorosis (currently rare in Europe) in 14 metallurgists which were all disclosed in foundry workers in Žiar nad Hronom as to the year 2005. The occupational disease was diagnosed after 17.7 ± 7.67 years (x±SD) of exposure in the foundry. The authors describe the clinical conditi

Overnight malodor effect with a 0.454% stabilized stannous fluoride sodium hexametaphosphate dentifrice.

Stannous fluoride (SnF2) is a broad-spectrum antimicrobial agent effective against caries, plaque, and gingivitis. Because oral malodor has a microbial etiology, the potential immediate effects of SnF2 on malodor were evaluated in 2 independent, randomized clinical trials. Both studies used a similar double-blind, crossover design, with subjects randomized to a treatment sequence with a 0.454% stabilized SnF2 sodium hexametaphosphate dentifrice or sodium fluoride dentifrice control. Overnight malodor was assessed at baseline in the morning before brushing and 24 hours later, after morning and

Clinical evaluation of the effect of attenuation correction technique on 18F-fluoride PET images.

OBJECTIVE: The purpose of this study is to evaluate effect of attenuation correction technique on 18F-fluoride positron emission tomography (PET). METHODS: We performed PET scans after the injection of 185 MBq 18F-fluoride on 32 patients from October 20th, 2004 to April 13th, 2005. We calculated bone-to-muscle ratios for the images with and without attenuation correction. We placed regions of interest (ROIs) on normal bone accumulation in 22 patients. The exclusion criteria were bone metastasis, Paget's disease, and rheumatoid arthritis. Several regions were chosen for ROI placement: skull, c

Effect of a fluoride-releasing one-step adhesive system on monkey pulp tissue.

PURPOSE: The purpose of this study was to evaluate the pulpal response of a fluoride-releasing one-step adhesive in nonexposed monkey teeth. MATERIALS AND METHODS: Cervical Class V cavities were prepared in monkey teeth. The cavities were divided into three groups of different restorative materials. A one-step adhesive (SI-IB551, Shofu) was applied to the teeth, and the cavities were filled with a resin composite (Beautifil, Shofu). In the other two groups, a two-step adhesive (Imperva FluoroBond, Shofu) together with either a resin composite (Beautifil) or a glass-ionomer cement (Fuji II, GC

The impact of the hyperacid Ijen Crater Lake: risks of excess fluoride to human health.

The Asembagus irrigation area (East Java, Indonesia) receives a high input of fluoride (F) via surface water that partially originates from the hyperacid crater lake of the Ijen volcano. Endemic dental fluorosis among local residents has been ascribed to F in water wells. In this study, the total F intake by children and adults was estimated, based on concentrations in well waters and foods throughout the area. These values were compared with the Lowest Observed Adverse Effect Level (LOAEL) for dental fluorosis among children and skeletal fluorosis among adults. Fluorosis hazard maps were prep

Lethal inhalation exposure during maintenance operation of a hydrogen fluoride liquefying tank.

Calcium sulfate adheres to the inside of liquefying pipes during the production of liquefied hydrogen fluoride. It is regularly washed away with water jets every six months. Two days before the operation, the pipes were experimentally washed down with water and the safety of the operation was confirmed with acidic washing fluid (pH 5). A 65-year-old man was severely sprayed on his face just after the start of the operation. He died half an hour later from acute respiratory failure. High serum concentrations of ionized fluoride indicated massive exposure to hydrofluoric acid (HFA). Pathological

Mechanisms in fluoride-induced interleukin-8 synthesis in human lung epithelial cells.

Sodium fluoride (NaF) has previously been reported to induce a strong IL-8 response in human epithelial lung cells (A549) via mechanisms that seem to involve the activation of G proteins. In the present study the signal pathways downstream of the G proteins have been examined. NaF induced a weak, but sustained increase in PKC activity. In contrast, the PKC activator TPA induced a relatively strong, but transient effect and augmented the NaF-induced PKC activity. TPA induced a marked IL-8 response compared to NaF. PDB, another PKC activator, was less effective, but augmented the IL-8 response t

Amine and tin fluoride inhibition of Streptococcus sanguis adhesion under continuous flow.

This study evaluated the ability of topically applied amine fluoride (AmF) and AmF-tin fluoride to inhibit the adhesion of Streptococcus sanguis within a parallel plate flow cell system. One of three AmF compounds and two tin fluoride preparations significantly reduced the net bacterial adhesion to conditioned glass over a 1-h period. Tin(IV) fluoride inhibited S. sanguis adhesion to the greatest extent, and this was shown to be dependent on the formation of the conditioning film prior to agent application.

Fluoride use by gender, age and dental fear among patients in a private practice.

The present study involved 145 consecutive self-referred patients (89 women, 56 men) at a private general practice, aimed at identifying whether differences in anxiety levels between women and men were related to self-administered fluoride prevention. Use of fluoride was assessed using a questionnaire to the use of tablets, rinse or other methods in addition to fluoride toothpaste. In logistic regression models, with fluoride use as the dependent variable, neither dental anxiety, measured by the Dental Anxiety Scale or by the Dental Fear Survey, nor age, were related to the use of fluorides. Detection of apoptotic changes in HeLa cells after treatment with paracetamol and sodium fluoride.

Apoptosis is genetically programmed cell death, an irreversible process of cell senescence with characteristic features (cell shrinkage, chromatin condensation, DNA fragmentation, apoptotic bodies) different from necrosis. Several effective in vitro methods for qualitative and quantitative detection of apoptotic events have been developed. Chromatin degradation, reductions in cell volume and other apoptosis-associated changes in cell morphology and physiology can be quickly analysed by multiparametric flow cytometry (FC) using small numbers of intact cells. One further method used for morpholo

Variation of the total amount of fluoride in blood of young women during the menstrual cycle.

We have previously reported the distribution and the forms of fluorides in the blood of men. This study is concerned with the distribution of fluorides in the blood of young women and sex-related differences in the fluoride metabolism related to the calcium metabolism keeping the postmenopausal osteoporosis in mind. The present study shows that women, aged 18 to 20 years, retain more fluorides in blood compared with men and that the nonionic fluoride level in blood varies during a menstrual cycle, whereas the ionic fluoride level is constant. Since fluoride has strong affinity for calcium, beh

[Wedge-shaped defects of the neck of the tooth in regular brushing with an amine fluoride gel].

Gingival recession and wedge-shaped defects are caused mainly by incorrect tooth brushing procedures. The aim of this epidemiologic study was to determine if in addition there is a relationship to the regular use of an amine fluoride gel (pH 5.2) applied by a toothbrush. In a regression analysis, the effect of age, sex, education, toothbrushing frequency, gingival bleeding (PBI) and use of the gel on the occurrence of wedge-shaped defects was investigated. Similarly, the influence of these variables on the occurrence of gingival retraction without any hard tissue defect was investigated. Both

Comparative study of the effect of sodium fluoride and sodium monofluorophosphate on glucose homeostasis in the rat.

The effect of acute and chronic administration of sodium fluoride (NaF) or sodium monofluorophosphate (MFP) on the glucose homeostasis of the rat are compared. The oral administration of a single dose of 40 mumol/100 g b.w. of either compound produced similar increases in plasma glucose (up to 1.8 g/l) and diffusible fluoride (up to 130 mumol/l). In long-term experiments (three months of duration), treatment with NaF (a 5 mmol/l solution as the water supply) produced, in the first month of experiment, abnormal glucose tolerance tests and increased plasma diffusible fluoride levels (range: 2-12

Effect of slow-release sodium fluoride on cancellous bone histology and connectivity in osteoporosis.

We have previously demonstrated that a treatment regimen of slow-release sodium fluoride (SRNaF) and continuous calcium citrate increases lumbar bone mass, improves cancellous bone material quality, and significantly reduces vertebral fracture rate in osteoporotic patients. In order to assess whether such treatment also improves trabecular structure, we quantitated cancellous bone connectivity before and following 2 years of therapy with SRNaF in 23 patients with osteoporosis and vertebral fractures. In addition, we performed bone histomorphometry on the same sections used for connectivity mea
