Feline nasal lymphoma (FNL) is the most common nasal tumor in cats. Although some negative prognostic factors for FNL have been previously reported, further studies on its molecular pathophysiology are needed. In the present study, we conducted comprehensive investigations of gene expression profiles (GEPs) in FNL. GEPs were compared between 17 FNL tissues and peripheral blood mononuclear cell (PBMC) samples of 3 healthy cats by RNA-sequencing. Then, we extracted differentially expressed genes (DEGs), and the differences in their mRNA expression levels were validated by RT-qPCR using 31 FNL, 3

BACKGROUND: Increasing evidence indicates that tumor cellular senescence can impair antitumor immunity and promote skin cutaneous melanoma (SKCM) progression. However, effective methods for assessing tumor cellular senescent status and tumor immune microenvironment (TIME) status remain lacking. This study intends to establish a novel Senescence-TIME Risk Score (STIRS) based on senescence and TIME related genes to predict prognosis and immunotherapy responsiveness in SKCM patients, thereby providing new strategies for current clinical personalized treatment.
METHODS: We identified distinct sen

Salivary adenoid cystic carcinoma (SACC) is an immunologically cold malignancy with limited response to current immunotherapies. Integrated transcriptomic profiling of peripheral blood, primary tumors, lung metastases, and a two-donor single-cell dataset revealed compartment-specific expression signatures and, through computational inference, systemic immune dysregulation marked by hematopoietic suppression, T-cell exhaustion, compensatory myelopoiesis, and an immature B-cell expansion. To explore the transcriptional basis of this peripheral immune aberration, blood-derived RNA-seq was interro

PURPOSE: Suture placement and alkali burn to the cornea are often used to induce inflammatory corneal neovascularization (CorNV) models in animals. This study compares the changes in genome-wide gene expression under these two CorNV conditions in mice.
METHODS: CorNV were induced in Balb/c mice by three interrupted 10-0 sutures placed at sites about 1 mm from the corneal apex, or by alkali burns that were 2 mm in size in the central area of the cornea. At the points in time when neovascularization progressed most quickly, some eyeballs were subjected to histological staining to examine CorNV

Disulfidptosis is a newly defined disulfide stress-induced cell death. However, there are gaps in the prognostic role of disulfidptosis-related genes (DRGs) and their correlation with the tumor microenvironment (TME) in gastric cancer (GC). In here, we systematically investigated DRG changes at genomic and transcriptional levels, prognostic value, and their expression patterns in GC. Fifteen DRGs were used to identify the different subtypes, and the differences in prognosis and immune infiltration among the subtypes were examined. We identified 3 distinct molecular subtypes and observed profou

BACKGROUND: The clinical heterogeneity of colon adenocarcinoma (COAD) complicates patient prognosis and treatment. While metabolic reprogramming is a key driver of tumor progression, the role of histidine metabolism in the COAD immune microenvironment remains unclear.
METHODS: Through an integrated analysis of public single-cell and bulk transcriptomic data, we mapped the COAD cellular atlas and assessed histidine metabolism activity. A prognostic Histidine Metabolism-related Model (HRM) was constructed using an ensemble of 101 machine learning algorithms, and its biological underpinnings wer

This research aimed to determine genes associated with M1 TAMs (tumour-associated macrophages) and to develop an M1 TAMs-related signature for predicting GC (Gastric cancer)'s prognosis and therapeutic effect. Based on the GC dataset in TCGA, we constructed a prognostic signature using M1 TAMs-related genes and validated it using data from the GEO dataset. To evaluate the predictive power of the signature, the survival curves, ROC curves, Cox regression analysis, nomograms and calibration curves were constructed. Differences in immune infiltration, immunotherapy response, and chemotherapy sens

The heterogeneity of pancreatic adenocarcinoma (PAAD) tumors is complex. The purpose of this work was to use immunologic signature gene sets (ISGS) activity alterations to identify subtypes of PAAD and to develop valid prognostic indicators. We discovered tumor-related differentially expressed gene sets and their associated subtypes by analyzing changes in ISGS activity via the cancer genome Atlas and the genotype-tissue expression datasets. To evaluate the relationship between these subtypes and clinical characteristics, the immunological microenvironment, and tumor immune dysfunction and exc
