Integrative analyses of potential biomarkers and pathways for non-obstructive azoospermia.

Background: Non-obstructive azoospermia (NOA) is the most severe form of male infertility. Currently, the molecular mechanisms underlying NOA pathology have not yet been elucidated. Hence, elucidation of the mechanisms of NOA and exploration of potential biomarkers are essential for accurate diagnosis and treatment of this disease. In the present study, we aimed to screen for biomarkers and pathways involved in NOA and reveal their potential molecular mechanisms using integrated bioinformatics. Methods: We downloaded two gene expression datasets from the Gene Expression Omnibus (GEO) database.

Corticotropin-releasing hormone and proopiomelanocortin gene expression is altered selectively in the male rat fetal thymus by maternal alcohol consumption.

The present study was carried out to investigate how hormonal changes caused by chronic alcohol exposure of rats during the late period of gestation are coordinated with neuroendocrine functions of the fetal thymus, namely thymic expression of CRH and POMC genes. Alcohol consumption by pregnant dams led to a 5-fold elevation of plasma corticosterone (CORT) levels and significantly decreased fetal CORT levels. This generally inverse correlation between maternal and fetal CORT was absent in alcohol-consuming dams and their male fetuses on day 19 of gestation. These day 19 fetuses also had an att

Protein stabilization: a common consequence of mutations in independently derived v-Myc alleles.

Myc is overexpressed in many cancers as a result of gene rearrangement or amplification, but coding sequence changes which cluster in the N-terminal transactivation domain also appear to play a role in tumour progression. The prototypic v-Myc gene of MC29 virus differs from avian c-Myc by a series of mutations, including a change at a regulatory phosphorylation site within the mutational hotspot (thr-61) which is known to potentiate transformation in vitro. We now show that the mutation at thr-61 stabilizes the v-Myc protein (turnover difference) and that this single mutation is both necessary

Human melanocytes expressing MC1R variant alleles show impaired activation of multiple signaling pathways.

Variant alleles of the human MC1R gene are strongly associated with red hair color, fair skin and poor tanning ability (RHC-trait). Recently, we demonstrated that melanocytes harboring RHC-associated alleles have markedly reduced surface expression and/or impaired G-protein coupling of the corresponding receptor protein. The consequences of such a deficit on MC1R-mediated signaling pathways have now been quantitatively evaluated utilizing strains of human primary melanocytes homozygous for RHC-associated variant alleles and comparing responses to wild-type strains. The ability of melanocortin

CD4 gene transcription is transiently repressed during differentiation of myeloid cells to macrophage-like cells.

The CD4 glycoprotein, which serves as receptor for human immunodeficiency virus (HIV), is expressed in several types of cells of hematopoietic origin, including T lymphocytes and monocytes. Triggering differentiation of peripheral blood monocytes, monocytic U-937 or promyelocytic HL-60 precursor cells to macrophage-like cells by phorbol ester treatment transiently induced both a rapid reduction in surface CD4, demonstrated by flow-cytometry analysis, and a gradual loss of CD4 mRNA, revealed by Northern-blot analysis. Experiments in HL-60 cells to determine the cause of the observed decay in CD

5-methoxyresorcinol mitigates postmenopausal osteoporosis through regulation of the PI3K-AKT-GSK3β signaling pathway and ROS homeostasis.

OBJECTIVES: Osteoporosis is a systemic skeletal disease characterized by excessive osteoclast-mediated bone resorption. This study aimed to evaluate the therapeutic potential of 5-methoxyresorcinol (MR) in osteoporosis and to clarify its underlying mechanisms. METHODS: Network pharmacology analysis was performed to identify shared targets between MR and osteoporosis. In vitro experiments using bone marrow-derived macrophages were performed to assess the effects of MR on osteoclast differentiation and function. In addition, the in vivo efficacy of MR was evaluated in an ovariectomized (OVX) mo

Transcription profiles of chicken liver and spleen in response to infection with avian pathogenic Escherichia coli at different stages.

Avian pathogenic Escherichia coli (APEC) is an extraintestinal pathogen that causes diverse local and systemic infections in poultry and poses a potential zoonotic threat. Although extensive research has addressed its antimicrobial resistance, virulence factors, and host immune responses, the temporal transition from early infection to recovery remains poorly characterized. Here, we established a systemic infection model in broilers by inoculating the pectoral muscle with E. coli O18:K1 and monitored sequential changes in organ weights and indices, serum biochemistry, and histopathology. 5,6,7,4'-Tetramethoxyflavone, a Dietary Polymethoxyflavone, Exerts Antitumor Effect on HeLa Xenografts in Mice.

5,6,7,4'-Tetramethoxyflavone (TMF), a naturally occurring polymethoxyflavone (PMF) abundant in Citrus species, has demonstrated potent antitumor activity against HeLa cells in vitro. To extend these findings, this study systematically investigated its therapeutic efficacy and safety profile in a HeLa tumor xenograft model. Here, we first found that TMF induced apoptosis in HeLa cancer cells both in vitro and in vivo. Proteomics analysis identified 19 differentially expressed proteins (DEPs) from HeLa cancer cells after TMF treatment, including downregulation of HSP60, sTNF-R1, JNK, TAK1 (S412)

Construction of Gene Regulatory Networks Based on Spatial Multi-Omics Data and Application in Tumor-Boundary Analysis.

BACKGROUND/OBJECTIVES: Cell-cell communication (CCC) is a critical process within the tumor microenvironment, governing regulatory interactions between cancer cells and other cellular subpopulations. Aiming to improve the accuracy and completeness of intercellular gene-regulatory network inference, we constructed a novel spatial-resolved gene-regulatory network framework (spGRN). METHODS: Firstly, the spatial multi-omics data of colorectal cancer (CRC) patients were analyzed. We precisely located the tumor boundaries and then systematically constructed the spGRN framework to study the network

Lateral cerebellar nucleus stimulation promotes motor recovery and suppresses neuroinflammation in a fluid percussion injury rodent model.

BACKGROUND: Many traumatic brain injury (TBI) survivors live with persistent disability from chronic motor deficits despite contemporary rehabilitation services, underscoring the need for novel treatment. OBJECTIVE/HYPOTHESIS: We have previously shown that deep brain stimulation (DBS) of the lateral cerebellar nucleus (LCN) can enhance post-stroke motor recovery and increase the expression of markers of long-term potentiation in perilesional cerebral cortex. We hypothesize that a similar beneficial effect will be for motor deficits induced by unilateral fluid percussion injury (FPI) in rodent

Signaling and drug sensitivity.

Even though alterations in receptor and nonreceptor kinases are involved in the development of human cancer, many cancer cell lines still retain their responsiveness to growth factors. We have investigated the hypothesis that cellular signaling events regulate the sensitivity of cancer cells to chemotherapeutic agents. In 2008 human ovarian carcinoma cells, activation of a number of different transduction pathways resulted in a 2 to 4-fold increase in the sensitivity to cisplatin. These signaling events include pathways activated by the epidermal growth factor (EGF) receptor, tumor necrosis fa

The global regulator LaeA controls expression of genes involved in diverse primary and secondary metabolites biosynthesis and their secretion in fungi.

LaeA is a global transcriptional regulator that differentially modulates expression of an impressive variety of secondary metabolite gene clusters in fungi, acting as a positive regulator fo some while repressing others. This regulator controls the production of fungal cellulolytic/ligninolytic enzymes and citric acid secretion in Ascomycetes. Also, it regulates the biosynthesis of siderophores and the antitumor agent ganoderic acid in Basidiomycetes. This regulator contains an S-adenosylmethionine-binding motif, suggesting a function as a methyltransferase, although its specific methylation s

Candidate Key Proteins in Tinnitus-A Bioinformatic Study of Synaptic Transmission in the Inferior Colliculus.

Proteins involved in synaptic transmission in normal hearing, acoustic stimulation, and tinnitus were identified using protein-protein interaction (PPI) networks. The gene list for tinnitus was compiled from the GeneCards database using the keywords "synaptic transmission" AND "inferior colliculus" AND "tinnitus" (Tin). For comparison, two gene lists were built using the keywords "auditory perception" (AP) and "acoustic stimulation" (AS). The STRING and the Cytoscape data analyzer were used to identify the top two high-degree proteins (HDPs) and the corresponding high-score interaction protein

Transduction of leptin growth signals in placental cells is independent of JAK-STAT activation.

We have characterized the transduction pathways induced by leptin in the placenta, using human BeWo cells that express endogenous leptin receptors and synthesize leptin in a regulated manner. We first examined if the JAK-STAT phosphorylation cascade was functional in these cells. Phosphorylated JAK2 was primarily bound to a short 106kDa leptin receptor isoform and to a lesser extent to a 210kDa molecule. Leptin neither enhanced JAK2 phosphorylation nor activated STAT3 and STAT1 proteins indicating that JAK2 is constitutively activated and that the JAK-STAT transduction pathway is not recruited

Male and Female Mice Show Similar Fear Memory Performance Despite Hippocampal Immediate Early Gene Expression Differences During Encoding and Consolidation.

Accurate and efficient memory processing is essential for survival. A body of ongoing work in both human subjects and animal models suggests that memory processing may differ substantially between males and females. In mice, contextual fear memory (CFM) encoding, consolidation, and recall have been well studied, and the mouse hippocampus and amygdala have been implicated in these processes. The present pilot study addresses whether the activation of these brain regions differs substantially between male and female mice at each stage of CFM processing. We find that male and female mice show no

Post-weaning folate deficiency induces a depression-like state via neuronal immaturity of the dentate gyrus in mice.

Folate deficiency has been suggested as a risk factor for depression in preclinical and clinical studies. Several hypotheses of mechanisms underlying folate deficiency-induced depressive symptoms have been proposed, but the detailed mechanisms are still unclear. In this study, we assessed whether post-weaning folate deficiency affect neurological and psychological function. The low folate diet-fed mice showed depression-like behavior in the forced swim test. In contrast, spontaneous locomotor activity, social behavior, coordinated motor skills, anxiety-like behavior and spatial memory did not

Reduced c-Fos expression in medullary catecholaminergic neurons in rats 20 h after exposure to chronic intermittent hypoxia.

Persons affected by obstructive sleep apnea (OSA) have increased arterial blood pressure and elevated activity in upper airway muscles. Many cardiorespiratory features of OSA have been reproduced in rodents subjected to chronic-intermittent hypoxia (CIH). We previously reported that, following exposure to CIH, rats have increased noradrenergic terminal density in brain stem sensory and motor nuclei and upregulated expression of the excitatory α(1)-adrenergic receptors in the hypoglossal motor nucleus. This suggested that CIH may enhance central catecholaminergic transmission. We now quantified

G(alphaq)-protein carboxyl terminus imitation polypeptide GCIP-27 attenuates cardiac hypertrophy in vitro and in vivo.

1. Various G(q)-protein-coupled receptors, such as alpha(1)-adrenoceptors, angiotension AT(1) receptors, endothelin ET(A) receptors, neuropeptide Y(1) receptors etc., contribute to cardiac hypertrophy. In G-protein signalling pathways, the carboxyl terminus of the G(alpha) subunit plays a vital role within G-protein-receptor interaction. The present study was designed to explore the effects of the synthetic G(alphaq) carboxyl terminal imitation peptide GCIP-27 on cardiac hypertrophy. 2. Hypertrophy of rat cultured cardiomyocytes was induced by noradrenaline (NA) or angiotensin (Ang) II in vitr
