Central Adiposity, Systemic Inflammation, and Incident Heart Failure: A Causal Mediation Analysis From the Jackson Heart Study.

BACKGROUND: Obesity and systemic inflammation have each been independently associated with an increased risk of heart failure (HF). However, whether inflammatory pathways mediate the association between obesity and the development of heart failure remains unclear. METHODS: We analyzed data from a community-based cohort of 1998 participants to evaluate multiple adiposity indicators in relation to incident HF. hs-CRP (high-sensitivity C-reactive protein) was examined as a mediator using causal mediation analysis to quantify the contribution of systemic inflammation. Kaplan-Meier analyses and mu

Pink1-mediated mitophagy in the endothelium releases proteins encoded by mitochondrial DNA and activates neutrophil responses during inflammation.

Eukaryotic mitochondria are characterized by several features that represent vestiges of their prokaryotic ancestry. One such feature is the N-terminal formylation of proteins encoded by mitochondrial DNA that undergo translation by mitochondrial ribosomes. N-formylated proteins are also released by bacteria and trigger activation of immune cells, such as neutrophils. Growing evidence indicates that circulating levels of mitochondrial formyl proteins are elevated in the serum of patients with excessive inflammatory responses. However, the mechanisms by which they are released into circulation

Influence of Oxidative Stress-Mediated Inflammation on the Pathogenesis of Reproductive Disorders: Exploring the Benefits of Carnosine for Prevention and Treatment of Endometriosis.

Endometriosis is a chronic pathological condition characterized by the growth of endometrial-like tissue outside the uterine cavity and is frequently associated with severe pain, persistent inflammation, and fibrosis within the pelvic region and other parts of the body. The exact causes of endometriosis are not clear, but an innate or adaptive immune response defect has recently been suggested as a factor in the disease's development. Carnosine is a natural dipeptide formed by the ligation of β-alanine and L-histidine and characterized by a multimodal mechanism of action that includes antioxid

Demethoxycurcumin Alleviates Lipopolysaccharide-Induced Acute Lung Injury via Nrf2-Mediated Anti-Inflammation and Ferroptosis Inhibition.

BACKGROUND: Sepsis-induced acute lung injury (SI-ALI) represents a life-threatening condition driven by dysregulated immune responses and redox imbalance. Demethoxycurcumin (DMC), a bioactive analog of curcumin, exhibits marked anti-inflammatory and free radical-scavenging capacities. Despite its potential, the precise molecular pathways through which DMC mitigates SI-ALI pathogenesis have yet to be fully elucidated. METHODS: SI-ALI was induced in mice through lipopolysaccharide (LPS) administration. Lung injury was assessed by histopathological analysis and measurement of wet-to-dry (W/D) we

Attenuation of cGAS-STING signaling-mediated lung inflammation during infection through autophagy induction by bioactive nanodevices.

BACKGROUND: Modulating the cGAS-STING pathway by bioactive nanodevices is a promising strategy for combating infection-associated inflammatory disorders. However, the development of pharmacological inhibitors for cGAS-STING signaling is currently hindered by lacking cell-specific targeting capability. This study aimed to develop a potent, drug-free nanodevice that specifically targets pulmonary macrophages to modulate the cGAS-STING pathway for ameliorating infection-associated detrimental lung inflammation. METHODS: Cigarette smoke extract-modified peptide gold nanoparticle hybrids (CSE-P12)

Cytokine-mediated activation of kidney-infiltrating CD8+ T cells enables their contribution to inflammation in human lupus nephritis.

Proliferative lupus nephritis (LN) is triggered by deposition of autoantibodies in glomeruli and paralleled by a T cell-rich kidney infiltrate. Although these T cells have been attributed to the propagation of tissue injury, it is unclear how they are activated and whether T cell autoreactivity drives local inflammation. Kidney-infiltrating T cells are also observed in urine, where they have high resemblance to interstitial T cells. Therefore, urinary T cells are a proxy for investigating tissue pathogenesis. Here, we analyzed urinary T cells to elucidate whether a kidney-specific T cell autoi

Targeting macrophage-mediated TGF-β/BMP signaling in ankylosing spondylitis: from inflammation to pathological bone formation.

Ankylosing spondylitis (AS) is a chronic, immune-mediated disease characterized by inflammatory arthritis and pathological new bone formation, ultimately leading to spinal fusion and functional impairment. Despite effective suppression of inflammation by biologic agents targeting cytokines such as TNF-α and IL-17A, radiographic progression often continues, highlighting a critical dissociation between inflammatory activity and structural damage. The literature suggests that macrophage polarization and the TGF-β/BMP signaling axis collectively constitute a critical nexus linking inflammation to

Transcriptome and m6A epitranscriptome profiling reveal CCL20 as a key mediator of inflammation in Staphylococcus aureus-induced bovine mastitis.

Mastitis caused by Staphylococcus aureus (S. aureus) poses a significant challenge in dairy farming because of its impact on animal health and milk production. Understanding the regulatory role of N6-Methyladenosine (m6A) modification in this disease remains crucial. Here, we used direct RNA sequencing (DRS) to explore transcriptomic changes in bovine mammary epithelial cells (BMECs) exposed to S. aureus (SA) compared to untreated controls (MEC). We identified 178 differentially expressed isoforms (DEIs) enriched in pathways such as the TNF signaling pathway, Kaposi's sarcoma-associated herpes

Diosmetin alleviates retinal ischemia-reperfusion injury through SIRT1-mediated suppression of oxidative stress, inflammation, and PANoptosis.

BACKGROUND: Retinal ischemia-reperfusion (RI/R) injury causes vision loss and lacks effective treatments. The role of diosmetin (DIO) in this condition, particularly through the SIRT1/Nrf2 pathway and its effect on PANoptosis, is unknown. METHODS: An in vitro oxygen-glucose deprivation/reperfusion (OGD/R) model was developed using R28 retinal precursor cells, while an in vivo rat model of retinal ischemia/reperfusion (RI/R) was created through transient intraocular pressure elevation. DIO was administered in vitro, at the onset of reperfusion, and in vivo, via intraperitoneal injection. Cell

NCOA4-mediated ferroptosis drives cGAS-STING-dependent inflammation in radiation dermatitis: Protective modulation by Cu-ATSM.

BACKGROUND: Radiation dermatitis (RD) remains a major clinical complication of radiotherapy, but its upstream molecular mechanisms are incompletely understood. Increasing evidence suggests that ferroptosis-an iron-dependent form of regulated cell death-may contribute to radiation-induced tissue injury, yet its role in skin pathology and immune activation remains undefined. METHODS: We integrated multi-omics analysis, cellular co-culture, computational modeling, and in vivo validation to investigate the role of NCOA4-mediated ferritinophagy and its interplay with the cGAS-STING pathway in RD.

Inert polystyrene nanoplastics-induced liver injury in mice was implicated in intense oxidative stress and inflammation mediated by ferroptosis.

Polystyrene nanoplastics (PS-NPs) are emerging food safety contaminants. Ferroptosis is iron-dependent cell death, but its role in PS-NPs hepatotoxicity is unclear. Mice received tail-vein injection of PS-NPs (2-8 mg/kg). PS-NPs caused liver injury (elevated transaminases) and possible renal impairment (increased uric acid/creatinine/urea). Hepatic GSH and SOD decreased, IL-1β and TNF-α increased. Mitochondrial shrinkage and cristae loss (ferroptotic features) were observed. PS-NPs upregulated ACSL4, MDA, 4-HNE and TfR, but suppressed FTH1, FPN1, SLC7A11 and GPx4. Thus, ferroptosis mediates PS

Brain-derived HMGB1 mediates periodontal inflammation after ischemic stroke through the STAT3/NLRP3 inflammasome pathway.

Ischemic stroke is known to cause immunosuppression and may exacerbate peripheral conditions such as periodontitis, yet its direct impact on periodontal tissues remains unclear. Using a transient middle cerebral artery occlusion (MCAO) mouse model, we showed that stroke rapidly induced periodontal inflammation, including epithelial disruption, alveolar bone loss, increased osteoclasts, and activation of the NOD-like receptor family pyrin domain containing 3 (NLRP3) inflammasome. Meanwhile, we found that neuronal high-mobility group box 1 (HMGB1) translocated from the nucleus and was released a

Engineering a spleen-selective mRNA-LNPs vaccine by decoupling the inflammation from cellular immunity-mediated cancer immunotherapy.

Rationale: mRNA vaccine-based cancer immunotherapy requires innate immune activation followed by potent cellular immunity. Vectors of lipid nanoparticles (LNPs) with proinflammatory properties activate the innate immune pathway, while excessive inflammatory response of mRNA-LNPs vaccine often results in systemic inflammation, compromising its therapeutic safety. Methods: Here, we engineered a spleen-selective mRNA-LNPs (mRNA-sLNPs) vaccine by decoupling the excessive inflammation from strong cellular immunity through ionizable lipids substituting for potent cancer immunotherapy. Results: The m
