Genetic structure and knockdown resistance (kdr) mutations in Aedes albopictus (Skuse) (Diptera: Culicidae): Implications for dengue fever transmission in southeastern China.

Dengue fever, transmitted by Aedes albopictus in China, is a major public health issue. The emergence of kdr gene mutations in Ae. albopictus has reduced the efficacy of insecticide-based control. We investigated the genetic structure of eight Ae. albopictus populations from China's southeastern coastal region, analyzing genetic diversity, population structure, and the prevalence of kdr mutations in relation to dengue fever incidence. Allelic diversity was moderate, with the number of alleles ranging from 2 to 6 and effective number of alleles from 1.52 to 5.00. Genetic clustering revealed fou

New dimensions in the field of antimalarial research against malaria resurgence.

Malaria is a life threatening disease caused by microscopic parasites called Plasmodium that are transmitted to human beings by mosquitoes. Single celled Eukaryotic plasmodium parasite is responsible to cause malaria in human beings and is transmitted by bite of Anopheles species mosquitoes. Resurgence of malaria towards the end of 20th Century is due to failure of its eradication completely. Parasite recurrence occurs due to high densities of parasite, low immunity and non opimized drug concentration. The ineffective eradications strategies were due to indefinable complex life cycle of Plasmo

The toxicity of angiotensin converting enzyme inhibitors to larvae of the disease vectors Aedes aegypti and Anopheles gambiae.

The control of mosquitoes is threatened by the appearance of insecticide resistance and therefore new control chemicals are urgently required. Here we show that inhibitors of mosquito peptidyl dipeptidase, a peptidase related to mammalian angiotensin-converting enzyme (ACE), are insecticidal to larvae of the mosquitoes, Aedes aegypti and Anopheles gambiae. ACE inhibitors (captopril, fosinopril and fosinoprilat) and two peptides (trypsin-modulating oostatic factor/TMOF and a bradykinin-potentiating peptide, BPP-12b) were all inhibitors of the larval ACE activity of both mosquitoes. Two inhibito…

Management and Treatment of Human Lice.

Of the three lice (head, body, and pubic louse) that infest humans, the body louse is the species involved in epidemics of louse-borne typhus, trench fever, and relapsing fever, but all the three cause pediculosis. Their infestations occur today in many countries despite great efforts to maintain high standards of public health. In this review, literature searches were performed through PubMed, Medline, Google Scholar, and EBSCOhost, with key search words of "Pediculus humanus", "lice infestation", "pediculosis", and "treatment"; and controlled clinical trials were viewed with great interest.

Polyenylcyclopropane carboxylic esters with high insecticidal activity.

BACKGROUND: Pyrethroids are synthetic derivatives of naturally occurring pyrethrum. These molecules are widely used in agriculture for ant, fly and mosquito control and for lawn and garden care. Pyrethroids are the optically active esters of 2,2-dimethyl-3-(2-methylpropenyl)-cyclopropane carboxylic acid, also known as chrysanthemic acid. However, their intense use has resulted in the development of resistance in many insect species. Herein, specific structural modifications of the pyrethroid scaffold and their effect on insecticidal activity, especially on resistant pests strains, are reported

The mosquitocidal activity of isoxazoline derivatives afoxolaner, lotilaner, and fluralaner are not affected by mosquito sugar or antibiotic treatment.

Long-lasting insecticidal nets and indoor residual sprays have played a major role in significantly reducing the burden of malaria. However, the management of mosquitoes resistant to current insecticides continues to be challenging. A promising new strategy is drug-based vector control where drugs are administered to vector hosts (human/animal), which renders the host blood toxic to mosquitoes-thereby reducing mosquito lifespan, fecundity and onward transmission of pathogens. Afoxolaner, fluralaner, and lotilaner are isoxazoline derivatives widely used as systemic insecticides for ectoparasite

Delivery and effectiveness of entomopathogenic fungi for mosquito and tick control: Current knowledge and research challenges.

Insects, ticks, and mites represent a threat to animal health globally, mainly due to their role as vectors of pathogens. Among the most important diseases, those transmitted by mosquitoes (e.g., malaria and arboviral infections) and ticks (e.g., Lyme borreliosis, babesiosis, and viral haemorrhagic fever) have a huge impact on human health. The principal methods available for reducing the public health burden of most vector-borne diseases are vector-based intervention relying to insecticides and acaricides. However, the use of these products is challenged by the introduction of invasive specie

Control of Aedes albopictus Mosquito Larvae with Carpesium abrotanoides L.

As a vital vector of dengue fever, yellow fever, and other mosquito-borne diseases, Aedes albopictus (Diptera: Culicidae) can be broadly distributed worldwide and cause a severe threat to public health. To date, considering the fast-emerging insecticide resistance in the mosquito, the development of new botanical insecticides to control and reduce Ae. albopictus is urgent and crucial. Therefore, to investigate the decoction effect of the plant C. abrotanoides L. on mosquito larvae killing, we detected the mortality of larvae after treatment with different concentrations (60 mg/mL, 120 mg/mL, a…

High concentrations of protein test substances may have non-toxic effects on Daphnia magna: implications for regulatory study designs and ecological risk assessments for GM crops.

Laboratory testing for possible adverse effects of insecticidal proteins on non-target organisms (NTOs) is an important part of many ecological risk assessments for regulatory decision-making about the cultivation of insect-resistant genetically modified (IRGM) crops. To increase confidence in the risk assessments, regulatory guidelines for effects testing specify that representative surrogate species for NTOs are exposed to concentrations of insecticidal proteins that are in excess of worst-case predicted exposures in the field. High concentrations in effects tests are achieved by using prote

Crystal structures of acetylcholinesterase in complex with organophosphorus compounds suggest that the acyl pocket modulates the aging reaction by precluding the formation of the trigonal bipyramidal transition state.

Organophosphorus compounds (OPs), such as nerve agents and a group of insecticides, irreversibly inhibit the enzyme acetylcholinesterase (AChE) by a rapid phosphorylation of the catalytic Ser203 residue. The formed AChE-OP conjugate subsequently undergoes an elimination reaction, termed aging, that results in an enzyme completely resistant to oxime-mediated reactivation by medical antidotes. In this study, we present crystal structures of the non-aged and aged complexes between Mus musculus AChE (mAChE) and the nerve agents sarin, VX, and diisopropyl fluorophosphate (DFP) and the OP-based inse

Engineering and malaria control: learning from the past 100 years.

Traditionally, engineering and environment-based interventions have contributed to the prevention of malaria in Asia. However, with the introduction of DDT and other potent insecticides, chemical control became the dominating strategy. The renewed interest in environmental-management-based approaches for the control of malaria vectors follows the rapid development of resistance by mosquitoes to the widely used insecticides, the increasing cost of developing new chemicals, logistical constraints involved in the implementation of residual-spraying programs and the environmental concerns linked t

CRISPR/Cas9 knock-out of nAChR α6 confers resistance to spinosyns in Frankliniella occidentalis and is associated with a higher fitness cost than target-site mutation G275E.

Thrips are major agricultural pests globally and spinosyn insecticides like spinosad and spinetoram are commonly used for their control. However, numerous cases of resistance have emerged, often linked to mutations in the nicotinic acetylcholine receptor (nAChR) α6 subunit, the main molecular target of spinosyns. In this study, toxicological data for spinosad and spinetoram were obtained from a susceptible strain of Frankliniella occidentalis, as well as two field-collected resistant strains carrying the G275E resistance mutation. Notably, a new candidate resistance mutation never reported bef

Pyrethroid Resistance in Anopheles gambiae s.l.: A Focus on Permethrin and Deltamethrin for Malaria Vector Control.

BACKGROUND: Malaria remains a major public health concern, and sub-Saharan Africa accounts for a significant proportion of global cases. A key challenge in malaria control is the increasing resistance of malaria vectors to commonly used insecticides, particularly pyrethroids. This study assessed the susceptibility of Anopheles gambiae s.l. to permethrin and deltamethrin in Zuba, located in the Gwagwalada Area Council of Abuja, Nigeria. METHODS: Mosquito larvae were collected in June 2024 and reared under controlled insectary conditions until adulthood. Adult mosquitoes were then tested using

First detection of kdr L1014F allele in Anopheles ziemanni and Anopheles pharoensis in Cameroon and distribution of the allele in members of the Anopheles gambiae complex.

BACKGROUND: Knockdown resistance (kdr) is one of the primary resistance mechanisms present in anopheline species. Although this mutation is largely spread across the Anopheles gambiae s.l. members, its prevalence in other species is still not well documented. METHODS: The present study investigated the distribution and allelic frequencies of kdr in An. gambiae s.l., An. pharoensis, and An. ziemanni samples collected in 2022 and 2023 in nine sites spread across five ecogeographical settings in Cameroon. Members of the An. gambiae complex were identified molecularly by polymerase chain reaction

Characterization of the inheritance of field-evolved resistance to diamides in the fall armyworm (Spodoptera frugiperda) (Lepidoptera: Noctuidae) population from Puerto Rico.

The fall armyworm (Spodoptera frugiperda) is one of the most destructive pests of corn. New infestations have been reported in the East Hemisphere, reaching India, China, Malaysia, and Australia, causing severe destruction to corn and other crops. In Puerto Rico, practical resistance to different mode of action compounds has been reported in cornfields. In this study, we characterized the inheritance of resistance to chlorantraniliprole and flubendiamide and identified the possible cross-resistance to cyantraniliprole and cyclaniliprole. The Puerto Rican (PR) strain showed high levels of resis

Effects of deltamethrin exposure on the cytochrome P450 monooxygenases of Aedes albopictus (Skuse) larvae from a dengue-endemic region of northern part of West Bengal, India.

Aedes albopictus is highly prevalent in the northern part of West Bengal and is considered to be responsible for the recent dengue outbreaks in this region. Control of this vector is largely relied on the use of synthetic pyrethroids, which can lead to the development of resistance. In the present study, larvae of three wild Ae. albopictus populations from the dengue-endemic regions were screened for deltamethrin resistance, and the role of cytochrome P450 monooxygenases (CYPs) was investigated in deltamethrin exposed and unexposed larvae. Two populations were incipient resistant, and one popu

Functional Roles of Five Cytochrome P450 Transcripts in the Susceptibility of the Yellow Fever Mosquito to Pyrethroids Revealed by RNAi Coupled With Insecticide Bioassay.

We evaluated the possible roles of five cytochrome P450 transcripts in the susceptibility of both adults and larvae of Aedes aegypti to three pyrethroids using RNA interference (RNAi) coupled with insecticide bioassays. RNAi by feeding larvae with chitosan/dsRNA nanoparticles led to reductions of CYP6AA5, CYP6AL1, CYP9J32, CYP4J16A, and CYP4J16B transcripts by 38.7%, 46.0%, 46.52%, 44.0%, and 41.0%, respectively, and increased larval mortality by 46.0% to permethrin when CYP9J32 was silenced and by 41.2% to cypermethrin when CYP6AA5 was silenced. RNAi by injecting dsRNA in adults led to reduct

What's eating you? head lice (Pediculus humanus capitis).

The head louse (Pediculus humanus capitis) is a blood-sucking arthropod of the suborder Anoplura. Infestation continues in epidemic proportions in children of all socioeconomic groups. Although not implicated as a disease vector, infestation can lead to considerable distress, missed days of school, and secondary infections. Pyrethroids are recommended for treatment, but resistance is common. Newer agents, including benzyl alcohol and spinosad, have been developed to address this gap in care.
