BACKGROUND: General therapeutic options for patients with acute myelogenous leukemia (AML) are reviewed and specific new therapies are described.
METHODS: Data in this review came from the published literature and the M. D. Anderson Cancer Center's acute leukemia database.
RESULTS: Outcome following standard therapy of AML is so variable that is best to speak of a range of outcomes determined by various prognostic factors. Trisomy of chromosome 12 is one of the commonest cytogenetic abnormalities in the karyotype in chronic lymphocytic leukemia (CLL). It is associated with atypical morphology of lymphocytes, progressing disease and poor survival. Granulocytic sarcoma (GS) is an increasingly common relapse feature of acute myeloid leukemia (AML), late in the disease course or post bone marrow transplantation (BMT). A case of juvenile myelomonocytic leukemia (JMML, previously referred to as JCML) in a neonate with Noonan syndrome (NS) is described. The boy presented with bilateral congenital hydrothoraces, nonimmune hydrops, dysmorphic facies, persistent thrombocytopenia, and leukocytosis. PROBLEM: Leukemia inhibitory factor (LIF) is a pleiotropic cytokine of the interleukin-6 family and has different biological actions in various tissue systems. Although named for its ability to inhibit proliferation of a myeloid leukemic cell line by inducing differentiation, it also regulates the growth and differentiation of embryonic stem cells, primordial germ cells, peripheral neurons, osteoblasts, adipocytes, and endothelial cells. Rat basophilic leukemia (RBL) cells, decorated with IgE, have been shown to bind irreversibly to antigen-coated substrates. In this paper we measured RBL cell deformability and demonstrated that this irreversible binding is not due to a compliant cellular rheology of these cells. An 82-year-old patient with untreated chronic lymphatic leukaemia known for 6 years developed large livid red plaques with central blisters and subsequent ulceration of the lower legs. Reactive angioendotheliomatosis was diagnosed based on histological immunohistological and electronmicroscopic examination, since proliferating endothelial cells with intravascular fibrin thrombi were identified. When the leukaemia was treated the skin eruption healed completely.

Adult T-cell leukemia (ATL) was first reported in Japan, where it has a high incidence in the southwestern region. The retrovirus, human T-lymphotropic virus type I (HTLV-I), is found to be the causative agent of ATL. In ATL-endemic areas, the rate of HTLV-I carriers is high.
