A simple and reliable high performance liquid chromatographic (HPLC) method has been developed and validated for the analysis of malondialdehyde (MDA) in human seminal plasma. After human seminal plasma was hydrolyzed, MDA, one of the hydrolysis products, reacted with thiobarbituric acid (TBA) to form MDA (TBA)2, a red-colored adduct with a maximum absorbance at 532 nm. HPLC separation of the adduct in human seminal plasma was performed on a Lichrospher C18 column. A mobile phase composed of 0.025 mol/L KH2PO4 (pH 6.2)-methanol in 58:42 (v/v) was found to be the most suitable ratio for this se

The biochemical modification of the metals and metalloids mercury, tin, arsenic, antimony, bismuth, selenium, and tellurium via formation of volatile metal hydrides and alkylated species (volatile and involatile) performs a fundamental role in determining the environmental processing of these elements. In most instances, the formation of such species increases the environmental mobility of the element, and can result in bioaccumulation in lipophilic environments. While inorganic forms of most of these compounds are well characterized (e.g., arsenic, mercury) and some of them exhibit low toxici

Human eosinophils produce upon treatment with 5-oxo-eicosatetraenoic acid or (5S,15S)-dihydroxyeicosatetraenoic acid a potent eosinophil-chemotactic eicosanoid, 5-oxo-15-hydroxy-(6E,8Z,11Z,13E)-eicosatetraenoi c acid (5-oxo-15-HETE). 5-Oxo-15-HETE induces human eosinophil (Eo) chemotaxis at nanomolar concentrations with an efficacy in vitro comparable to that seen for platelet activating factor. Comparison of Eo chemotactic activities of several structurally related eicosanoids with different substituents and/or double bound geometry led to the conclusion that maximal potency and efficacy of e

Stearic acid, phosphatidylcholine, and phosphatidylglycerol nitroxide spin-labels were used to probe the effect of 1-hexanol, urethane, diethyl ether, and ethanol on lipid-protein interactions in nicotinic acetylcholine receptor (nAcChoR) rich membranes from Torpedo nobiliana. For stearic acid spin-labeled at the C-14 position of the sn-1 acyl chain, 1-hexanol induced little change (over a wide concentration range, 0-16.7 mM) in either the ESR line shape or the proportion of motionally restricted spectral component from labels probing the protein interface. The main effect of 1-hexanol was lim

Glycosylation, the enzymatic addition of carbohydrate moieties to proteins and lipids, is a fundamental posttranslational modification that profoundly influences cellular processes. Altered glycosylation patterns represent a characteristic of malignant transformation, affecting tumor progression, immune evasion, metastasis, and response to therapy. The emerging field of glycomics aims to systematically characterize these glycan structures, providing insights into their biological roles and clinical relevance. Mass spectrometry (MS) has become an indispensable tool in cancer glycomics due to it

Sevoflurane (sevo) exhibits cardioprotective effects against myocardial ischemia/reperfusion injury (MIRI); however, its precise molecular mechanism of action remains elusive and warrants further in-depth investigation. A rat model of MIRI and a hypoxia/reoxygenation (H/R) model in the H9c2 cardiomyocytes were established separately, followed by sevo treatment. Hematoxylin-eosin staining, Perls staining, transmission electron microscope, FerroOrange staining, and Liperfluo staining were used to assess myocardial pathological damage, iron deposition, and oxidative stress. BACKGROUND: Utpalashatphala ghrita (USG) is a traditional ayurvedic medicine containing piperine as its main chemical constituent responsible for the activity. Murchita is an ayurvedic process to detoxify and purify the lipids (ghee, oil). It involves heating ghee with specific ayurvedic ingredients for a specified duration and temperature. It is essential to understand the stability of piperine during and after the murchita process.
OBJECTIVE: The objective of this study was to develop and validate an LCMS method to assess the stability of piperine after the murchita process.
METHOD: LCMS s

Plasma membrane vesicles (PMVs) are powerful model systems for studying plasma membrane biophysics, but their production typically relies on chemicals, such as dithiothreitol (DTT) and paraformaldehyde (PFA). DTT is detrimental to protein structure and can alter membrane composition, whereas PFA is a well-established fixative that is widely used in biological studies. Taking this into consideration, we introduced a DTT-free method for generating PMVs using PFA alone. Using flow cytometry and fluorescence microscopy, we validated its robustness across multiple cell types. Measurements with the

Brassica crops, encompassing globally important vegetables and oilseeds, face severe threats from diverse biotic and abiotic stresses. Plant secondary metabolites constitute the chemical foundation of defence, and metabolomics has emerged as an effective systems biology tool for comprehensively dissecting stress-induced metabolic changes. Recent progress in applying metabolomics to elucidate defence mechanisms in Brassica crops is systematically synthesised here. Major stresses confronting Brassica crop production and the metabolic basis of plant defence are first outlined. Current analytical

Sickle cell disease (SCD) is an autosomal recessive hemoglobin disorder that is mainly characterized by the presence of hemoglobin S (HbS; point mutation [Glu6Val] in the beta-globin gene). Under deoxygenated conditions, HbS polymerizes and serves as the primary trigger of oxidative stress in red blood cells (RBCs), promoting polymerization of Band 3, a major membrane scaffold protein that links the lipid bilayer to the spectrin-ankyrin cytoskeletal network. Phosphorylation at key residues within the cytosolic domain of Band 3 induces conformational changes that weaken ankyrin binding and enha

Bacterial lipopolysaccharide (LPS) is among the most potent pathogen-associated molecular patterns (PAMPs). In animals, LPS can induce endotoxic shock, a widely used model of sepsis. Studies of this system have yielded important insights into innate immunity, including key immune sensors and signaling pathways. However, how inflammatory pathways are spatially coordinated across cell types and organs in vivo during endotoxic shock remains poorly understood. Here, we systematically analyzed LPS-induced lethality in mice to define the coordinated contributions of cell death pathways, inflammatory

Plaque heterogeneity underlies the propensity of atherosclerotic lesions to rupture and trigger cardiovascular events. Most proteomic studies examine bulk changes, obscuring key spatial differences in protein abundance. We report a high-resolution spatial proteomics workflow exploring the molecular landscape of human plaques and a murine myocardium. By combining laser capture microdissection with high-sensitivity ion-mobility mass spectrometry, spatial profiling of cellular and extracellular matrix (ECM) proteomes was achieved. Over 2700 proteins were detected from 50,000 μm2 areas, revealing

Excessive dietary dextrose intake and its misuse in sports raise significant concerns about health outcomes across life stages. To investigate how biological maturity influences small intestinal responsiveness to dextrose overexposure, we subjected 1-month-old (young maturing) and 14-month-old (middle-aged) male Wistar rats to a 20% or 60% dextrose drinking regimen for 8 weeks. The middle-aged rats receiving the 60% dextrose solution exhibited the most pronounced alterations, including compromised barrier integrity, reduced crypt proliferation, and accelerated enterocyte apoptosis and necrosis

The increasing spread of mobile colistin resistance (mcr) genes is becoming a major concern in the treatment of infections caused by multidrug-resistant Gram-negative bacilli. Colistin is often used as a last treatment option, but the emergence of mcr genes is reducing its effectiveness. These genes are most commonly found in bacteria, such as Escherichia coli, Klebsiella pneumoniae, and Salmonella, and have also been reported, though less frequently, in organisms like Pseudomonas aeruginosa. This review provides an overview of the occurrence, diversity, and mechanisms of mcr genes in Gram-neg

Recently, photodynamic therapy (PDT) has gained considerable attention as a promising therapeutic approach for the treatment of psoriasis. Unfortunately, the activation of high mobility group box 1 protein (HMGB1) by PDT triggers innate and adaptive immune responses, which exacerbate skin inflammation. Herein, we combined glycyrrhizic acid (GA), a natural anti-inflammatory compound and immunomodulator derived from the herb Glycyrrhiza uralensis Fisch., with PDT actuated by the photosensitizer chlorin e6 (Ce6) by co-loading them in GA-based lipid nanoparticles coated with a catechol-modified qu

BACKGROUND: Type 2 diabetes (T2D) is a prevalent and costly disease, with sedentary behaviour and physical inactivity as modifiable contributors. Mobile health (mHealth) applications provide complementary strategies for T2D management, but their impact on clinical outcomes remains unclear.
OBJECTIVE: This study evaluated the efficacy of an mHealth programme promoting "sit less and move more" at work, prescribed in clinical practice, on clinical and cardiovascular risk factors in office employees with T2D.
METHODS: A randomized controlled trial compared usual care (n = 25) with a 13-week mHea

The envelope (E) protein of the SARS coronavirus forms a pathogenic cation-selective channel across intracellular membranes. The ion-conduction mechanism of the hydrophobic transmembrane domain of the protein, ETM, has been elusive despite recent determination of its high-resolution structures in the closed and open states. Here, we investigate the structural mechanism of SARS ETM by mutating two residues, T11 and N15. T11A is the second most common mutation in Omicron variants and has attenuated channel activity and cell lethality compared to wild-type ETM, whereas the N15A mutant is absent f
