Lipidomic profiling of plasma and urine from patients with Gaucher disease during enzyme replacement therapy by nanoflow liquid chromatography-tandem mass spectrometry.

Gaucher disease (GD) is a rare genetic disorder that arises from lipid species, especially monohexosylceramide (MHC), accumulating in different organs. GD results from a β-glucocerebrosidase deficiency, causing metabolic or neurologic complications. This study comprehensively profiled lipids from patients and healthy controls to discover active lipid species related to GD. Most studies have evaluated lipids from one type of biological sample, such as plasma, urine, or spinal fluid, which are the main sources of lipids in human bodies. The purpose of this study, however, was to collect and asse

Metabolomic and lipidomic plasma profiles according to metabolic dysfunction-associated steatotic liver diseases (MASLD) stages in the absence of type 2 diabetes (T2D).

INTRODUCTION: Amino acids (AAs), tricarboxylic acid (TCA) cycle intermediates, and acylcarnitines (ACs) can reflect energetic metabolism. Metabolic dysfunction-associated steatotic liver (MASLD) has been associated with the modification of plasma AAs, ACs and TCA cycle intermediates' profiles, but the changes in advanced fibrosis without type 2 diabetes (T2D) are not well studied. OBJECTIVES: The objective of this pilot study was to describe the targeted plasma metabolomic profile in individuals with advanced fibrosis to test research hypotheses concerning hepatic energy metabolism. METHODS:

Lipidomic Comparison of Detergent-Soluble and Detergent-Resistant Lipid Domains Isolated From Rat Cerebellum and the Effect of Hypoxia-Ischemia Using UPLC-HDMSE.

Hypoxic ischemic encephalopathy (HIE) due to insufficient oxygen or blood flow to the brain can result in inflammation, impaired neurodevelopment, or death. Changes to microdomain lipid composition may contribute to altered structure and function of key regulatory processes; however, knowledge regarding changes to membrane lipid composition after HIE is incomplete. Here, we describe the application of untargeted lipidomics to investigate the impact of hypoxia-ischemia on detergent-resistant membrane (DRM) and detergent-soluble membrane (DSM) domains fractionated from the cerebellum in a rat mo…

Partial body mass regain attenuates lipid utilization and alters the hepatic lipidome linked to HDL dysfunction during metabolic syndrome in male rats.

Increases in body mass are associated with the development of metabolic dysfunction-associated steatotic liver disease (MASLD). Caloric restriction (CR) is the primary non-pharmacological defense against MASLD; however, poor CR maintenance induces partial body mass regain (PR), leading to MASLD progression. We conducted an in vivo study to investigate the effects of PR following 2 weeks of CR on the hepatic lipidome during Metabolic Syndrome (MetS) using the male Otsuka Long Evans Tokushima Fatty (OLETF) rat and the Long Evans Tokushima Otsuka (LETO) rat as strain control (Ctrl). PR reduced he

LDL subspecies and lipidome change by evolocumab add-on therapy to empagliflozin in patients with type 2 diabetes: A prespecified secondary analysis of a randomized clinical trial.

BACKGROUND: Sodium-glucose cotransporter-2 inhibitors (SGLT2i) and proprotein convertase subtilisin/kexin type 9 inhibitors (PCSK9i) reduce cardiovascular risk in type 2 diabetes. Whether these benefits are partly mediated by modifications in low-density lipoprotein (LDL) phenotype, beyond changes in LDL-cholesterol levels, remains unknown. METHODS: In this prespecified analysis of the EXCEED-BHS3 randomized trial, 110 individuals with type 2 diabetes were assigned to 16 weeks of empagliflozin (E) or empagliflozin plus evolocumab (EE). Changes in LDL subspecies profiles were assessed in the f

Integrative lipidomic and transcriptomic analysis unraveled polystyrene nanoplastics-induced liver injury via oral and inhalation exposure: All roads lead to Rome?

The ubiquitous environmental presence of nanoplastics (NPs) necessitates urgent investigation into their biological impacts. As the primary target organ for accumulated NPs, the liver faces substantial health risks, but the differential hepatotoxic effects of different exposure routes remain unknown. In this study, a four-week exposure experiment in mice using polystyrene nanoplastics (PS-NPs) through oral and inhalation routes were conducted. Multidimensional assessments revealed exposure route-specific pathological patterns: oral administration primarily caused histopathological damage, wher

Conductive cooling and Moringa oleifera supplementation of heat stressed sows during late gestation and lactation alters the cheek cell lipidome of neonatal pigs.

Exposure to maternal heat stress during gestation and lactation reduces litter growth. Heat stress disrupts lipid metabolism and elevates the risk of oxidative damage to lipids. However, it remains unclear whether maternal heat stress during gestation and lactation affects lipid profiles of offspring. Buccal cheek cells, which can be collected noninvasively, provide a valuable source for assessing the metabolic response to such environmental challenges. A 2 × 2 factorial study was conducted to investigate the impact of maternal cooling and supplementation with Moringa oleifera, a high-antioxid

Comparison of the transcriptome, lipidome, and c-di-GMP production between BCGΔBCG1419c and BCG, with Mincle- and Myd88-dependent induction of proinflammatory cytokines in murine macrophages.

We have previously reported the transcriptomic and lipidomic profile of the first-generation, hygromycin-resistant (HygR) version of the BCGΔBCG1419c vaccine candidate, under biofilm conditions. We recently constructed and characterized the efficacy, safety, whole genome sequence, and proteomic profile of a second-generation version of BCGΔBCG1419c, a strain lacking the BCG1419c gene and devoid of antibiotic markers. Here, we compared the antibiotic-less BCGΔBCG1419c with BCG. We assessed their colonial and ultrastructural morphology, biofilm, c-di-GMP production in vitro, as well as their tra

Lipidomic profiling of the hepatic esterified fatty acid composition in diet-induced nonalcoholic fatty liver disease in genetically diverse Collaborative Cross mice.

Non-alcoholic fatty liver disease (NAFLD), one of the most common forms of chronic liver disease, is characterized by the excessive accumulation of lipid species in hepatocytes. Recent studies have indicated that in addition to the total lipid quantities, changes in lipid composition are a determining factor in hepatic lipotoxicity. Using ultra-high performance liquid chromatography coupled with electrospray tandem mass spectrometry, we analyzed the esterified fatty acid composition in 24 strains of male and female Collaborative Cross (CC) mice fed a high fat/high sucrose (HF/HS) diet for 12 w

Lipidomic analysis identifies age-disease-related changes and potential new biomarkers in brain-derived extracellular vesicles from metachromatic leukodystrophy mice.

BACKGROUND: Recent findings show that extracellular vesicle constituents can exert short- and long-range biological effects on neighboring cells in the brain, opening an exciting avenue for investigation in the field of neurodegenerative diseases. Although it is well documented that extracellular vesicles contain many lipids and are enriched in sphingomyelin, cholesterol, phosphatidylserines and phosphatidylinositols, no reports have addressed the lipidomic profile of brain derived EVs in the context of Metachromatic Leukodystrophy, a lysosomal storage disease with established metabolic altera

Early signature in the blood lipidome associated with subsequent cognitive decline in the elderly: A case-control analysis nested within the Three-City cohort study.

BACKGROUND: Brain lipid metabolism appears critical for cognitive aging, but whether alterations in the lipidome relate to cognitive decline remains unclear at the system level. METHODS: We studied participants from the Three-City study, a multicentric cohort of older persons, free of dementia at time of blood sampling, and who provided repeated measures of cognition over 12 subsequent years. We measured 189 serum lipids from 13 lipid classes using shotgun lipidomics in a case-control sample on cognitive decline (matched on age, sex and level of education) nested within the Bordeaux study cen…

New insights in hemp chemical composition: a comprehensive polar lipidome characterization by combining solid phase enrichment, high-resolution mass spectrometry, and cheminformatics.

The chemical composition of Cannabis sativa L. has been extensively investigated for several years; nevertheless, a detailed lipidome characterization is completely lacking in the literature. To achieve this goal, an extraction and enrichment procedure was developed for the characterization of phospholipids and sulfolipids. Firstly, a study on the solid-liquid extraction was performed, to maximize the recovery of the considered lipids; the best procedure consisted of a simple extraction with a mixture of methanol and chloroform (1:1, v/v). The hemp extracts were analyzed by ultra-high-performa

New insight into the role of lipids in the formation and retention of key aroma compounds in micro-pressure stewed beef based on lipidomics and lipid-derived free radical analysis.

Lipids play crucial roles in both the formation and retention of aroma in meat products. However, the effect of micro-pressure stewing on beef lipids involved in aroma production remains unclear. The lipid profiles, free radicals, and key aroma compounds in micro-pressure stewed beef were analyzed. A total of 29 aroma compounds with relative odor activity values (ROAVs) exceeding 1 were identified, with (E, E)-2,4-decadienal, (E, E)-2,4-nonadienal, and 1-octen-3-ol representing the predominant odorants. Phospholipids and triglycerides with C18 acyl groups might predominantly contributed to the

Targeted Lipidomics Reveal the Effects of Different Phospholipids on the Phospholipid Profiles of Hepatic Mitochondria and Endoplasmic Reticulum in High-Fat/High-Fructose-Diet-Induced Nonalcoholic Fatty Liver Disease Mice.

The lipid alternation in mitochondria and endoplasmic reticulum (ER) might be indicative of their abnormal morphology and function, which contribute to development of nonalcoholic fatty liver disease (NAFLD). However, the influence of dietary phospholipids (PLs) on the PL composition of the organellar membrane is largely unknown. High-fat/high-fructose (HFHF)-diet-induced NAFLD mice were administrated with different PLs (2%, w/w) with specific fatty acids and headgroups, including eicosapentaenoic acid (EPA)-phosphatidylcholine (PC)/phosphatidylethanolamine (PE)/phosphatidylserine (PS), docosa

Lipidomics study of the protective effects of isosteviol sodium on stroke rats using ultra high-performance supercritical fluid chromatography coupling with ion-trap and time-of-flight tandem mass spectrometry.

Isosteviol sodium (STV-Na) was reported to possess significant protective effects on ischemic stroke in recent years. However, the protective mechanism of STV-Na against stroke was still unclear. In this work, an untargeted lipidomics approach based on the ultra high-performance supercritical fluid chromatography coupling with ion-trap and time-of-flight tandem mass spectrometry (UHSFC-IT-TOF/MS) was employed to investigate the lipid profiles of stroke rats with STV-Na treatment for the first time. The possible mechanism of STV-Na was further elucidated. The UHSFC-IT-TOF/MS-based method achiev…
