Diffuse large B-cell lymphoma (DLBCL) stands as the most prevalent subtype of non-Hodgkin's lymphoma and exhibits significant heterogeneity. Various forms of programmed cell death (PCD) have been established to have close associations with tumor onset and progression. To this end, this study has compiled 16 PCD-related genes. The investigation delved into genes linked with prognosis, constructing risk models through consecutive application of univariate Cox regression analysis and Lasso-Cox regression analysis. Furthermore, we employed RT-qPCR to validate the mRNA expression levels of certain

PURPOSE: NK cells are essential for the detection, identification and prediction of cancer. However, so far, there is no prognostic risk model based on NK cell-related genes to predict the prognosis and treatment outcome of DLBCL patients. This study aimed to explore a risk assessment model that could accurately predict the prognosis and treatment efficacy of DLBCL.
METHODS: Bioinformatics analysis of the expression profiles of DLBCL samples in the GEO database was performed. Cox regression and LASSO regression analysis were used to determine NK cell-related genes associated with patient's pr

BACKGROUND: First-line treatment of diffuse large B-cell lymphoma (DLBCL) achieves durable remission in approximately 60% of patients. In relapsed or refractory disease, only about 20% achieve durable remission with salvage chemoimmunotherapy and consolidative autologous stem cell transplantation (ASCT). The ZUMA-7 (axicabtagene ciloleucel [axi-cel]) and TRANSFORM (lisocabtagene maraleucel [liso-cel]) trials demonstrated superior event-free survival (and, in ZUMA-7, overall survival) in primary-refractory or early-relapsed (high-risk) DLBCL with chimeric antigen receptor T-cell therapy (CAR-T)

Multiple myeloma (MM) and diffuse large B-cell lymphoma (DLBCL) comprise a large fraction of hematologic malignancies diagnosed each year. However, the co-occurrence of these conditions in the same patient is rare. CD19- and B-cell maturation antigen-targeted chimeric antigen receptor (CAR) T-cell therapies have been approved in recent years with promising responses. Here, we present a patient who presented following a bone marrow biopsy that revealed MM with 20% lambda-restricted plasma cells with no evidence of lymphoma involvement in the marrow. A subsequent lymph node biopsy of a right thi

Cell-free DNA (cfDNA) is a fragment of DNA circulating in the blood, and its concentration is often elevated in cancer patients. To investigate the relationships between serum cfDNA concentration and clinical characteristics, including prognosis, we measured serum cfDNA concentration in 114 newly diagnosed lymphoma patients. The cfDNA concentrations in diffuse large B cell lymphoma (DLBCL) (62.5 ng/mL) and follicular lymphoma patients (51.6 ng/mL) were significantly elevated compared to healthy individuals (7.5 ng/mL, P < 0.001). In DLBCL, patients with elevated serum cfDNA (> 38.9 ng/mL) at d

BACKGROUND: Diffuse large B-cell lymphoma (DLBCL) is an aggressive cancer of the lymphatic system. About 30% to 40% of people with DLBCL experience relapse and 10% are refractory to first-line treatment usually consisting of R-CHOP chemotherapy. Of those eligible for second-line treatment, commonly consisting of salvage chemotherapy followed by autologous stem-cell transplantation (ASCT), around 50% experience relapse. With a median overall survival of less than six to 12 months, the prognosis of individuals who relapse or are refractory (r/r) to advanced lines of treatment or of those who are

BACKGROUND: Given the absence of consensus diagnostic criteria for giant cell arteritis, clinicians may encounter difficulty with identification of new-onset headache in patients older than age 50 years presenting with visual changes and elevated inflammatory markers, particularly if temporal artery biopsies are performed and negative.
CASE PRESENTATION: We present a case of a 57-year-old white man with headache, diplopia, and jaw paresthesia initially diagnosed and managed as steroid-refractory biopsy-negative giant cell arteritis. Further investigation disclosed evidence of soft tissue infi

INTRODUCTION: Diffuse large B-cell lymphoma can be cured in 60% - 70% of patients. Autologous stem cell transplantation is the standard treatment for relapsed disease. This high-intensity treatment after first complete remission in patients with high International Prognostic Index remains controversial and was performed in our department during some years.
MATERIAL AND METHODS: Retrospective study, review of clinical records.
RESULTS: This study evaluates the outcome of 113 patients transplanted between 1992 and 2012. Considering status before transplantation patients were divided in groups:

Regulatory T (Treg) cells are essential for maintaining immune tolerance. High Treg frequencies have been reported in peripheral blood and tissue samples of patients with solid tumors while their role in lymphomas, including diffuse large B-cell lymphoma (DLBCL) has not been clearly established. In this study, we analyzed the circulating Treg numbers in 27 patients with newly diagnosed DLBCL and 17 healthy individuals. Tregs were detected by flow cytometry based on CD4(+) CD25(high) FoxP3(+) phenotype. In addition, the expression of CD45RA, HLA-DR, CD62L, CD39, and CTLA4 was analyzed. The numb

We describe the case of a 64-year-old man who developed diffuse large B-cell lymphoma (DLBCL) in less than a year after he was diagnosed and treated for T-cell prolymphocytic leukemia (T-PLL). At the time of diagnosis of T-PLL he had a white blood cell count (WBC) of 38.2×10(9)/L and only few small lymph nodes were identified on physical examination. Hepatosplenomegaly or skin lesions were not present. Peripheral blood examination was remarkable for 91% circulating prolymphocytes, which by flow cytometry immunophenotypic analysis were CD2, CD3, CD5, and CD7 positive and coexpressed CD4 and CD8

BACKGROUND: Vaccine development targeting the novel immunogenic Per ARNT Sim Domain containing 1 (PASD1) cancer testis antigen represents an attractive therapeutic approach for the significant number of patients with diffuse large B-cell lymphoma who are refractory to conventional treatment. Since CD4-positive T helper cells have crucial roles in promoting and maintaining immune responses to tumor antigens, the presence of a CD4-positive T-helper immune response to the PASD1 antigen in patients with diffuse large B-cell lymphoma was investigated in the current study.

BACKGROUND: Primary cutaneous diffuse large B-cell lymphoma, leg type (PCDLBCL-LT), is a rare subset of primary cutaneous lymphoma known for its aggressive behavior, poor prognosis, and limited treatment options, particularly in the relapsed or refractory setting. We report a case of a patient diagnosed with PCDLBCL-LT who achieved a clinically significant response and sustained remission for seven months with acalabrutinib, a covalent inhibitor of Bruton's tyrosine kinase (BTK), before disease progression.

The efficacy of epcoritamab in refractory diffuse large B-cell lymphoma (DLBCL) has been established. However, its safety in dialysis-dependent patients is unclear. A 53-year-old man with refractory DLBCL who had received a series of different chemotherapies became dependent on dialysis because of worsening preexisting chronic kidney disease. Epcoritamab was initiated at a standard dose, and a complete response was achieved after the third cycle. However, it was discontinued after the seventh cycle because of the development of lower leg necrosis due to pre-existing arteriosclerosis. Epcoritam

BACKGROUND: Although CD19 CAR-T cell therapy has improved outcomes in relapsed or refractory diffuse large B-cell lymphoma (DLBCL), responses are variable and predictive biomarkers are lacking. The triglyceride glucose-body mass index (TyG-BMI)-a surrogate of insulin resistance-has shown prognostic value in solid tumors, but its relevance to DLBCL and CAR-T efficacy remains unknown.
METHODS: We retrospectively analyzed 204 newly diagnosed DLBCL patients treated with rituximab-containing regimens, stratified by median TyG-BMI. Prognostic associations were assessed by Kaplan-Meier and multivari

Prediction of the outcome of large B-cell lymphomas/high-grade B-cell lymphomas (DLBCLs/HGBCLs) is based on clinical parameters and molecular testing, for example, for rearrangements of MYC (MYC-R) and MYC-R in combination with BCL2 and BCL6 translocations (double/triple hit). However, the group of DLBCL/HGBCL with poor outcome is not confined to MYC-R lymphomas, and fluorescence in situ hybridization (FISH) testing for MYC-R misses several high-risk lymphomas. We aimed to understand if artificial intelligence (AI) trained to identify MYC-R will delineate a poor prognostic subgroup. We generat

BACKGROUND: This study challenges the static and linear assumptions of traditional prognostic models for diffuse large B-cell lymphoma (DLBCL), such as the International Prognostic Index (IPI).
RESEARCH DESIGN AND METHODS: Analyzing 664 DLBCL patients treated with R-CHOP, the research explored the time-dependent and nonlinear effects of established risk factors.
RESULTS: While the IPI maintained strong overall prognostic stability, its individual components exhibited dynamic behavior. ECOG performance score, beta-2 microglobulin (β2 M), and lactate dehydrogenase (LDH) were key predictors of

B-cell lymphomas (BCLs) are a heterogeneous group of blood cancers whose treatment has been significantly transformed by immunotherapy. The use of the anti-CD20 monoclonal antibody rituximab, combined with chemotherapy (R-CHOP), has markedly improved patient outcomes. However, some subtypes, particularly Diffuse Large B-Cell Lymphoma (DLBCL), remain challenging to treat due to frequent relapses and/or early resistance to therapy. Standard frontline immunochemotherapy is successful in about 60% of DLBCL cases, whereas 30-40% experience relapse or refractory disease, thus highlighting the urgent

Protein-losing enteropathy (PLE) is a rare complication of systemic lupus erythematosus (SLE). A 38-year-old woman with well-controlled SLE developed abdominal pain, lower-extremity edema, anemia, and hypoalbuminemia. The patient was then diagnosed with PLE. Despite treatment with glucocorticoids and cyclophosphamide, persistent hypoalbuminemia was observed. A biopsy was performed using double-balloon enteroscopy due to small bowel lesions, which confirmed the diagnosis of diffuse large B-cell lymphoma. To the best of our knowledge, this is the first reported case of diffuse large B-cell lymph

Hemophagocytic lymphohistiocytosis (HLH) is a rare but life-threatening hyperinflammatory syndrome characterised by uncontrolled immune activation and cytokine release, resulting in multiorgan dysfunction and high mortality if not recognised and treated promptly. In adults, secondary HLH is frequently triggered by infection, autoimmune disease, or malignancy, with haematological malignancies carrying a particularly poor prognosis. The non-specific nature of its presentation often leads to diagnostic delay, as it can closely mimic sepsis and other inflammatory conditions. We present the case of

Diffuse large B-cell lymphoma (DLBCL) can present with hyperlactatemia or even lactic acidosis (LA) as a rare but often underrecognized complication among hematologists. Patients diagnosed with DLBCL complicated by LA generally exhibit high mortality and a poor prognosis, even with therapeutic intervention. This report details two cases of DLBCL concurrently presenting with LA and provides a literature review. Our analysis of 19 published studies revealed a survival advantage in patients with an initial lactate levels of 5-14.9 mmol/L compared to those with levels of 15-24.9 mmol/L. Furthermor
