Skeletal muscle insulin unresponsiveness during chronic hyperdynamic sepsis in the dog.

Recent reports from our laboratory and others have documented changes in insulin unresponsiveness and electrolyte and hormonal changes characteristic of hypodynamic shock states in anesthetized animals. Since most acute shock protocols do not adequately mimic the clinical profile of sepsis, the present study was undertaken to document the hemodynamic and metabolic changes associated with chronic hyperdynamic peritonitis in dogs. Mongrel dogs of either sex weighing 20 +/- 2 kg were surgically instrumented with an electromagnetic aortic flow probe for monitoring cardiac output determinations, an

Porcine malignant hyperthermia: role of skeletal muscle in increased oxygen consumption.

During malignant hyperthermia (MH) induced by halothane and succinylcholine, oxygen consumption (VO2) of whole body, muscle and the splanchnic area was calculated from measurements of blood flow and arteriovenous oxygen content difference. Caudal body blood flow was isolated and measured (six animals) for determination of average muscle VO2 in the hind limbs, buttocks, and flanks. The increase in muscle VO2 was extrapolated to total skeletal muscle and compared with the increase in whole body VO2 (same six animals). The average increase in VO2 for both total muscle and whole body during the pe

Intracellular calcium movements of frog skeletal muscle during recovery from tetanus.

Radioautographs of (45)Ca-labeled frog skeletal muscles have been prepared using freeze-dry and vapor fixation techniques to avoid displacement of the isotope during the preparation of the radioautographs. (45)Ca has been localized in resting muscles exposed to (45)Ca Ringer's for 5 min or 5 hr and in isotopically labeled muscles recovering from tetanic stimulation at room temperature or at 4 degrees C. In muscles soaked at rest for 5 min (45)Ca was present almost exclusively in the terminal cisternae. In all other muscles there were three sites at which the isotope was concentrated: (a) the t

Unraveling skeletal muscle senescence in an in vitro cell culture model.

INTRODUCTION: Skeletal muscles are crucial for voluntary movement, posture, breathing, joint stability, and therefore are essential for maintaining overall health and quality of life. Aging, sarcopenia, cancer, immobilization, and several muscle pathologies can have an impact on muscle function, and muscle cell senescence is associated with these conditions. METHODS: Here we aimed to develop a primary cell culture model to study skeletal muscle cell senescence. We induced cell cycle arrest in cultured embryonic chick muscle cells using a low concentration of cytosine arabinoside (Ara-C) for 9

Redundant YAP and TAZ functions are essential for skeletal muscle regeneration.

Muscle stem cells orchestrate skeletal muscle regeneration through complex fate decisions. The transcriptional co-activators Yes-associated protein 1 (YAP) and WW domain-containing transcription regulator 1 (TAZ) contribute to multiple stages of myogenesis, yet their individual contributions to regeneration remain unclear due to substantial functional overlap. We genetically titrated YAP and TAZ expression in MuSCs with double knockout and single allele mutants by crossing Pax7 CreERT mice with TAZ flox/flox ;YAP flox/flox mice. Conditional deletion of both YAP and TAZ in muscle stem cells sev

Study on the spatiotemporal development of skeletal muscle in embryonic Pekin ducks.

This study aimed to investigate the developmental patterns of breast muscle and leg muscle in embryonic Pekin ducks, and to screen potential genes regulating their independent development and growth differences. Muscle weight and histological characteristics were measured at eight embryonic stages (E11, E13, E15, E17, E19, E21, E23, E25; n = 10). Transcriptome analysis was performed on breast muscle at E11, E13, E17, and E21, and on leg muscle at E17 and E21. The results showed that breast muscle weight exhibited an S-shaped growth pattern, while leg muscle weight increased significantly in a

Activation of ATP Consumption Is Necessary to Stimulate Glycogenolysis in Skeletal Muscle.

Glycogenolysis is an important contributor to ATP production in contracting skeletal muscle. However, the activation of glycogen phosphorylase in resting muscle does not by itself trigger significant glycogenolysis. To elucidate the mechanisms underlying this regulation, we analyzed the functional coupling between glycogenolysis and glycolysis using a mathematical model of energy metabolism in mammalian fast-twitch (white) skeletal muscle. A system-level analysis of the model reveals an important role for inorganic phosphate (a substrate for glycogen phosphorylase) in regulating glycogenolysis

Skeletal muscle-kidney crosstalk in a cohort of critical illness survivors.

BACKGROUND: The skeletal muscles and kidneys are frequently affected during critical illness; however, their crosstalk remains poorly explored, especially in the long-term evolution. Therefore, we investigated the crosstalk between skeletal muscle and kidney function in COVID-19 survivors. METHODS: A cross-sectional analysis of a prospective cohort study with survivors of moderate to severe COVID-19 hospitalization. Skeletal muscle assessments included handgrip strength, calf circumference, ultrasound-measured quadriceps thickness, and gait speed test. Sarcopenia was diagnosed by modified EWG

GPAT3 is associated with lipid deposition and AMPK signaling in skeletal muscle.

As metabolic disorders associated with excessive lipid deposition in skeletal muscle rise, strategies to alleviate this accumulation have emerged as a major focus in metabolic research. Glycerol-3-phosphate acyltransferase 3 (GPAT3) is a key enzyme in triglyceride biosynthesis; however, its role in muscle lipid metabolism remains unclear. In this study, we overexpressed or knocked down GPAT3 in C2C12 myoblasts to evaluate its effects on muscle lipid metabolism and then validated the findings in GPAT3 knockout (KO) mice. GPAT3 deficiency significantly reduced lipid deposition in muscle cells. I

Isolation of Immune Cells from Skeletal Muscles for Flow Cytometry.

Inflammation is a characteristic of skeletal muscle wound healing and of many muscle diseases. Analysis and isolation of the immune cell populations infiltrating skeletal muscles and their molecular signatures allow optimal use of anti-inflammatory treatments and identification of novel therapeutic targets. Here we describe a protocol for unbiased isolation of skeletal muscle immune cells. This technique allows for downstream assessment of immune cell populations or their isolation for downstream molecular or cellular assays from individual skeletal muscles during normal wound healing or disea

[Functional analysis of the mutant channels associated with skeletal muscle channelopathies].

Skeletal muscle channelopathies are rare genetic disorders caused by mutations in voltage-gated ion channel genes that regulate sarcomere excitability, including the CLCN1 gene encoding ClC-1, the KCNJ2 gene encoding Kir2.1, the SCN4A gene encoding Nav1.4, and the CACNA1S gene encoding Cav1.1. More than one hundred heterozygous missense mutations have been identified in SCN4A, representing a broad spectrum of clinical phenotypes, including sodium channel myotonia (SCM), paramyotonia congenita (PMC), hyperkalemic periodic paralysis (HyperPP) and hypokalemic periodic paralysis (HypoPP). Impact of Acute Endurance Exercise on Alternative Splicing in Skeletal Muscle.

PURPOSE: Alternative splicing (AS) is a highly conserved post-transcriptional mechanism, generating mRNA variants to diversify the proteome. Acute endurance exercise appears to transiently perturb AS in skeletal muscle, but transcriptome-wide responses are not well-defined. We aimed to better understand differential AS (DAS) and differential isoform expression (DIE) in skeletal muscle by comparing short-read (SRS) and long-read RNA sequencing (LRS) data. METHODS: Publicly accessible SRS of clinical exercise studies were extracted from the Gene Expression Omnibus. Oxford Nanopore LRS was perfo

Myostatin Exhibits an Evolutionarily Conserved Circadian Pattern in Skeletal Muscles.

INTRODUCTION: Myostatin (MSTN), a transforming growth factor-beta (TGF-β) superfamily member, is an evolutionarily conserved negative regulator of skeletal muscle mass. Loss of MSTN commonly promotes augmentation in skeletal muscle mass in all animal species examined. Recent studies have demonstrated that circadian clock proteins play a critical role in the regulation of muscle mass and function, in part by modulating the expression of key muscle-related genes. While myostatin has an important role in sustaining skeletal muscle protein turnover, it is unknown if circadian clock proteins regula

Microscopic and molecular aspects of skeletal muscle alterations in cerebral palsy.

Cerebral palsy (CP), the most prevalent childhood-onset motor disability, frequently entails progressive musculoskeletal complications. This comprehensive review synthesizes existing knowledge of microscopic and molecular alterations in CP skeletal muscle. Considerable methodological variability, heterogeneous patient cohorts, and inconsistent control groups significantly complicate comparative interpretations across studies. Nonetheless, some structural abnormalities consistently emerge, including increased variability in muscle fibre size, altered fibre type distribution, long sarcomeres at

Evaluating skeletal muscle wasting and weakness in models of critical illness.

Skeletal muscle wasting and weakness are common complications associated with admission to the intensive care unit (ICU), with the loss of muscle mass and function increasing mortality and contributing to physical impairments post-discharge. While our understanding of the pathophysiology of this condition, commonly termed 'ICU-acquired weakness' (ICU-AW), has advanced considerably, no effective therapies are available. ICU-AW broadly encompasses a range of muscle-related impairments in this setting, including, but not limited to, critical illness myopathy and sepsis-induced myopathy. Pre-clini

BOLA3 as a key protein for the treatment of diabetic skeletal muscle atrophy.

OBJECTIVE: Skeletal muscle is crucial for glucose metabolism, but diabetes impairs this function, leading to muscle atrophy. Although the mutation of BOLA Family Member 3 (BOLA3) resulted in disease Multiple Mitochondrial Dysfunctions Syndrome, the role of which in diabetic muscle atrophy is unclear. METHODS: Firstly, datasets were downloaded and assessed from skeletal muscle tissue with diabetic individuals in GEO database and key genes were screened using weighted gene co-expression network analysis (WGCNA) and machine learning algorithms. Subsequently, external validation datasets were app
