Hirschsprung's disease, Down syndrome, and missing heritability: too much collagen slows migration.

Hirschsprung's disease (HSCR) causes functional intestinal obstruction due to the absence of the enteric nervous system (ENS) in the distal bowel and is usually diagnosed shortly after birth or during childhood. While several genetic and nongenetic factors have been linked to HSCR, the underlying mechanisms that prevent ENS precursors from colonizing distal bowel during fetal development are not completely understood in many affected children. In this issue of the JCI, Soret and colleagues identify a new mechanism that causes HSCR-like disease in mice and involves deposition of excess collagen

The development of excitatory neurons in the chick cloaca.

INTRODUCTION: The enteric nervous system of the chick embryo hindgut is derived from the vagal and sacral neural tube. Nerve cells from thes … al tube was ablated at the 10-12 ss, but not replaced with quail neural tube, thus, only the sacral neural crest cells remained. All embryos were harvested at E12 and E14, embedded in paraffin wax and serially sectioned. Immunohistochemistry was carried out on all embryos using human natural killer-1, quail non-chick perinuclear, choline acetyltransferase (ChA …

The developmental biology of melanocytes and its application to understanding human congenital disorders of pigmentation.

In the past 15 years, a fruitful interplay between the description of mutations associated with human congenital disorders of pigmentation, the discovery of the molecular basis of murine coat color mutants, and the elucidation of determinants of melanocyte development from the neural crest has led to rapid advancement in the understanding of these disorders and the developmental biology underlying them. The insight gained about these human disorders from similar mouse models has facilitated progress that might not otherwise have occurred so rapidly. Clearly more work remains to be done, especi…

[Neurochristopathy in the differential diagnosis of newborn's apnea: case report].

OBJECTIVE: to include neurocristopathy on the etiological workup of neonatal apneas and discuss the importance of polysomnography in this diagnosis. METHOD: case report and critical review of the literature. RESULTS: we report on a newborn that presented respiratory failure in the first hours of life associated to abdominal distention. Continuous ventilatory support was necessary, and the respiratory distress increased during sleep. After polysomnographic confirmation of hypoventilation the newborn was submitted to neuroradiolgic tests and colon byopsy, positive to Hirschsprung's disease. C

Segmentation of the iliac crest from CT-data for virtual surgical planning of facial reconstruction surgery using deep learning.

BACKGROUND AND OBJECTIVES: For the planning of surgical procedures involving the bony reconstruction of the mandible, the autologous iliac crest graft, along with the fibula graft, has become established as a preferred donor region. While computer-assisted planning methods are increasingly gaining importance, the necessary preparation of geometric data based on CT imaging remains largely a manual process. The aim of this work was to develop and test a method for the automated segmentation of the iliac crest for subsequent reconstruction planning. METHODS: A total of 1,398 datasets with manual

Development of a lethal congenital heart defect in the splotch (Pax3) mutant mouse.

OBJECTIVE: The splotch (Sp2h) mutation disrupts the Pax3 gene and is lethal in homozygotes. The aim of the present study was to investigate … e of a cardiac defect and reduction or absence of dorsal root ganglia, which are derivatives of the neural crest. CONCLUSIONS: In this paper, we show that the lethality has a biphasic pattern, and the data strongly suggests that mid-gestation lethality is due to cardiac defects and not the associated neural tube defects. This finding supports the idea that 'c …

On the trail of the 'new head' in Les Treilles.

The vertebrate brain develops in association with neighboring tissues: neural crest, placodes, mesoderm and endoderm. The molecular and evolutionary relationships between the forming nervous system and the other craniofacial structures were at the focus of a recent meeting at the Fondation des Treilles in France. Entitled 'Relationships between Craniofacial and Neural Development', the meeting brought together researchers working on diverse species, the findings of whom provide clues as to the origin and diversity of the brain and facial regions that are involved in forming the 'new head' of v…

Linkage of an X-chromosome cleft palate gene.

Many congenital malformations, such as cleft palate and neural tube defects, have a multifactorial origin involving both environmental and genetic factors. Conditions such as these may be exclusively monogenic, polygenic or environmental, but in most cases both genetic and environmental factors are involved. This study describes the sub-chromosomal localization of a single gene defect causing cleft palate and ankyloglossia (tongue-tied) in a large Icelandic family. This defect is a model for the analysis of other neural-crest malformations that show a more complex multifactorial inheritance pa

Induction of Skin-Derived Precursor Cells from Human Induced Pluripotent Stem Cells.

The generation of full thickness human skin from dissociated cells is an attractive approach not only for treating skin diseases, but also f … d with recombinant noggin and SB431542, an inhibitor of activin/nodal and TGFβ signaling, to induce neural crest progenitor cells. Those cells were then treated with SKP medium that included CHIR99021, a WNT signal activator. The induction efficacy from neural crest progenitor cells to SKPs was more than 97%. No other modifiers tested were able to induce those …

Abnormal development of tracheal innervation in rats with experimental diaphragmatic hernia.

BACKGROUND: We previously demonstrated that tracheobronchial innervation, originated from the vagus nerve and hence of neural crest origin, is deficient in rats with experimental congenital diaphragmatic hernia (CDH). The present study examines the development of this innervation during fetal life in an attempt to understand the nature of these deficiencies. MATERIALS AND METHODS: Pregnant rats were given either 100 mg nitrofen or vehicle on E 9.5. Embryos were recovered on E15 and E18. Control and nitrofen/CDH pups (n = 10 each) were studied on each of these days and compared with our previo…

Cranial anomaly of homozygous rSey rat is associated with a defect in the migration pathway of midbrain crest cells.

Craniofacial development of vertebrates depends largely on neural crest contribution and each subdomain of the crest-derived ectomesenchyme follows its specific genetic control. The rat small eye (rSey) involves a mutation in the Pax-6 gene and the external feature of rSey homozygous embryos exhibits craniofacial defects in ocular and frontonasal regions. In order to identify the mechanism of craniofacial development, we examined the cranial morphology and migration of cephalic crest cells in rSey embryos. The chondrocranial defects of homozygous rSey embryos primarily consisted of spheno-orbi…

Cerebroarthrodigital syndrome: a newly recognized formal genesis syndrome in three patients with apparent arthromyodysplasia and sacral agenesis, brain malformation and digital hypoplasia.

We describe three patients with a complex syndrome of apparent arthromyodysplasia, dyscephaly, sacral agenesis, and hypoplastic digitis. Cause is unknown, but an environmental cause is suspected on the basis of ergotamine exposure in one case and diazoxide intake in another, together with suggestive similarities to anomalies seen in animals treated with these drugs and to calves with the Australian hydranencephaly/arthrogryposis syndrome caused by Akebane or Aino virus. Pathogenetically the primary defect may be a neural tube-neural crest dysplasia with multiple secondary and tertiary manifest…

Schwann cell precursors in health and disease.

Schwann cell precursors (SCPs) are frequently regarded as neural crest-derived cells (NCDCs) found in contact with axons during nerve formation. Nevertheless, cells with SCPs properties can be found up to the adulthood. They are well characterized with regard to both gene expression profile and cellular behavior -for instance, proliferation, migratory capabilities and survival requirements-. They differ in origin regarding their anatomic location: even though most of them are derived from migratory NCCs, there is also contribution of the boundary cap neural crest cells (bNCCs) to the skin and …

[Problems of the immunohistochemical differential diagnosis of neuroendocrine carcinoma and neuroblastomas].

Neurons arising from neural tube or neural crest do not express epithelial markers from the beginning of their differentiation and neuroblastomas arising from these anlages also do not express epithelial markers. On the other hand, neuroendocrine carcinomas, such as small cell carcinomas of the respiratory tract, express epithelial markers in addition to neuronal or neuroendocrine markers and demonstration of epithelial marker has been regarded as subtle evidence to rule out neuroblastomas. However, this general rule can not be applied to olfactory neuroblastomas. Unlike neurons derives from n…

Lifetime imaging reveals long-distance non-neural bioelectric patterns on timescales from seconds to hours.

Translating the bioelectric code remains one of the core challenges to widespread biomedical translation of bioelectric interventions, as we … timation of membrane potential (Vmemoe) to map the bioelectric dynamics of spreading Xenopus laevis neural crest explants (NCEs) over > 17 h. We identify a slow hours-scale bioelectric component and distinct faster minutes-scale and seconds-scale components. These dynamics often span multiple cells, consistent with roles in the collective behavior of NCEs. We …

Nager Syndrome Revisited: Integrating In Vivo and In Vitro Models to Decipher SF3B4-Dependent Tissue Coordination.

Nager syndrome (NS) is a rare congenital disorder primarily characterized by mandibulofacial dysostosis and upper limb anomalies. Pathogenic variants in SF3B4, which encodes a core spliceosomal component, represent the primary known genetic cause of NS. This review synthesizes recent findings from cellular, zebrafish, Xenopus, and mouse models to elucidate how SF3B4 deficiency perturbs neural crest cell (NCC) biology and multi-tissue development. Loss of SF3B4 induces widespread splicing abnormalities, with preferential exon skipping affecting AT-rich and GC-poor exons, thereby altering the ex…

The novel role of foxi3 in zebrafish mandibular development.

Recent studies have identified pathogenic variants in the FOXI3 gene associated with craniofacial microsomia pedigrees. In zebrafish, the foxi1 gene is considered a functional homolog of the mouse Foxi3. However, research on foxi3a and foxi3b, which display homologous genes in the naming of FOXI3 in zebrafish, has predominantly focused on their roles in epidermal ionocyte function. Our study reveals that disruption of foxi3a or foxi3b results in a reduced number of cranial neural crest cells (CNCCs) and hypoplastic mandibular cartilage in zebrafish. These findings introduce a new perspective o…

Aplasia Cutis Congenita Pathomechanisms Reveal Key Regulators of Skin and Skin Appendage Morphogenesis.

Aplasia cutis congenita (ACC) manifests at birth as a defect of the scalp skin. New findings answer 2 longstanding questions: why ACC forms and why it affects mainly the midline scalp skin. Dominant-negative mutations in the genes KCTD1 or KCTD15 cause ACC owing to loss of function of KCTD1/KCTD15 complexes in cranial neural crest cells (NCCs), which normally form midline cranial suture mesenchymal cells that express keratinocyte growth factors. Loss of KCTD1/KCTD15 function in NCCs impairs the formation of normal midline cranial sutures and, consequently, the overlying skin, resulting in ACC.…
