In vivo visualization of olfactory pathophysiology induced by intranasal cadmium instillation in mice.

Intranasal exposure to cadmium has been related to olfactory dysfunction in humans and to nasal epithelial damage and altered odorant-guided behavior in rodent models. The pathophysiology underlying these deficits has not been fully elucidated. Here we use optical imaging techniques to visualize odorant-evoked neurotransmitter release from the olfactory nerve into the brain's olfactory bulbs in vivo in mice. Intranasal cadmium chloride instillations reduced this sensory activity by up to 91% in a dose-dependent manner. In the olfactory bulbs, afferents from the olfactory epithelium could be qu…

Effect of vitamin A deficiency on retinol and retinyl esters contents in rat brain.

In the present study, the pattern of vitamin A (retinol and retinyl esters) contents in discrete brain areas was investigated in Wistar rats … brain rats (4 to 44% of total retinol). The abundant ester in both sexes was the retinyl linoleate. Olfactory bulb and the midbrain contained the highest quantities of retinol and retinyl esters in both sexes. On the other hand, the vitamin A deficient diet significantly decreased the retinoid contents in male brain, in olfactory bulb (-30.7%), hindbrain (-46.2% …

Neural regeneration and functional reconnection following olfactory nerve transection in hamster.

The olfactory sensory neurons in the vertebrate nervous system are unique in that they undergo continuous neurogenesis and replacement. Anatomical studies have shown that transection of the olfactory nerves leads to a degeneration of sensory neurons followed by a neurogenesis and replacement with newly formed cells. Replacement neurons grow axonal processes that are capable of reestablishing morphological connections with cells in the olfactory bulb. To determine the functional capacity of these anatomical reconnections, single unit responses to odor stimuli were recorded from cells in the olf…

Kallmann syndrome: fibroblast growth factor signaling insufficiency?

Kallmann syndrome (KAL) is a developmental disease that combines hypogonadotropic hypogonadism and anosmia. Anosmia is related to the absence or hypoplasia of the olfactory bulbs. Hypogonadism is due to GnRH deficiency and is likely to result from the failed embryonic migration of GnRH-synthesizing neurons. These cells normally migrate from the olfactory epithelium to the forebrain along the olfactory nerve pathway. KAL is phenotypically and genetically heterogeneous. The gene responsible for the X-chromosome linked form of the disease (KAL1) has been identified in 1991. KAL1 encodes anosmin-1…

MRI findings in Kallmann syndrome.

Kallmann syndrome (KS) is a neuronal migration disorder characterised by hypogonadotrophic hypogonadism and anosmia or hyposmia. Five patients with clinical findings suggestive of KS were evaluated with MRI. All patients had abnormalities of olfactory system. Olfactory bulbs were absent in all patients. Olfactory sulci were absent in 3 patients and hypoplastic in 2 patients. Anterior pituitary was hypoplastic in two patients. The MRI findings in KS are characteristic and MRI is a useful adjunct to the diagnosis of KS.

Olfactory recognition of infants in laboratory mice: role of noradrenergic mechanisms.

Noradrenaline depletion of the olfactory bulbs induces cannibalism at parturition in primiparous mice, without producing anosmia or impairment of maternal behaviour. Similar lesions made in multiparous experienced females do not result in cannibalism. The present studies investigated 1) whether a 30-min exposure to pups or to distal cues from pups given to virgin females before noradrenaline depletion of the olfactory bulbs overcame the impairment in recognition at first parturition, and 2) whether noradrenaline-depleted females allowed to care for their pups for the 24 h following parturition…

Identification and distribution of mouse carboxypeptidase A-6.

Carboxypeptidase A-6 (CPA6) was recently discovered in the human genome. To gain information regarding the potential function of this novel protein, the mouse homolog of CPA6 was identified using a combination of bioinformatics and reverse transcriptase-polymerase chain reaction (RT-PCR). In addition, homologs in rat, chicken, and frog were identified using a bioinformatics approach. The distribution of CPA6 mRNA in mouse tissues was examined using RT-PCR and in situ hybridization. A strong RT-PCR signal is detectable in olfactory bulb, and much lower levels are present in other regions such a…

Male hamster copulatory responses to a high molecular weight fraction of vaginal discharge: effects of vomeronasal organ removal.

The importance of the vomeronasal (accessory olfactory) system for the copulatory responses of male hamsters to a high molecular weight fraction (HMF) of vaginal discharge was assessed in animals that had their vomeronasal organs (VNO) removed. These organs were extirpated bilaterally using an oral approach through the palate so as to eliminate the peripheral afferents to the accessory olfactory bulb (AOB) with minimal or no damage to the main olfactory system. The selective peripheral deafferentation procedure was verified by applying horseradish peroxidase intranasally following intraperiton…

The cyclic AMP signaling pathway in the rodent main olfactory system.

Odor perception begins with the detection of odorant molecules by the main olfactory epithelium located in the nasal cavity. Odorant molecules bind to and activate a large family of G-protein-coupled odorant receptors and trigger a cAMP-mediated transduction cascade that converts the chemical stimulus into an electrical signal transmitted to the brain. Morever, odorant receptors and cAMP signaling plays a relevant role in olfactory sensory neuron development and axonal targeting to the olfactory bulb. This review will first explore the physiological response of olfactory sensory neurons to odo…

Molecular physiology of olfaction.

Olfactory reception is mediated by olfactory receptor cells located in the olfactory epithelium. These cells are bipolar neurons that extend a dendrite toward the nasal lumen and an axon toward the olfactory bulb in the brain. The dendrite possesses a group of apical cilia embedded in mucus. Odorant recognition and signal transduction are initiated at the membranes of these chemosensory cilia and culminate in excitation of the olfactory receptor cell. Differential activation by odorants of distinct groups of olfactory receptor cells generates patterns of neuronal activity that encode odor qual…

Myelination of the developing lateral olfactory tract and anterior commissure.

Both the lateral olfactory tract (LOT) and anterior limb of the anterior commissure (AC) carry olfactory information. The LOT forms the projection from the olfactory bulb to the ipsilateral olfactory cortices, while the AC carries odor information across the midline to the contralateral olfactory cortex and bulb. The LOT and AC differ on a number of dimensions, including early development and functional onset. The present work, examining their myelination in mice, reveals additional important differences. For example, the LOT initiates myelination 3-4 days earlier than the AC, evidenced by bot…

Successful treatment of phantosmia with preservation of olfaction.

A 26-year-old woman had an 8-year history of phantosmia in her left nostril. The phantosmia could be eliminated by nostril occlusion or coca … abnormal peripheral olfactory neurons from the nose or the interruption of incoming signals to the olfactory bulb eliminated the phantosmia. This form of therapy for phantosmia offers an alternative to more radical procedures such as olfactory bulbectomy and may offer a significant sparing of olfactory ability.

Age-dependent decrease in glomeruli and receptor cells containing α1-2 fucose glycan in the mouse main olfactory system but not in the vomeronasal system.

Receptor cells of the olfactory epithelium (OE) and vomeronasal organ (VNO) project axons to glomeruli in the main olfactory bulb (MOB) and accessory olfactory bulb (AOB), respectively and undergo continuous turnover throughout life. Alpha1-2 fucose (α1-2Fuc) glycan mediates neurite outgrowth and synaptic plasticity and plays important roles in the formation of the olfactory system during development. We previously confirmed the localization of α1-2Fuc glycan in the olfactory system of 3- to 4-month-old mice but whether such localization persists throughout life remains unknown. Here, the MOB,…

Two GABAergic intraglomerular circuits differentially regulate tonic and phasic presynaptic inhibition of olfactory nerve terminals.

Olfactory nerve axons terminate in olfactory bulb glomeruli forming excitatory synapses onto the dendrites of mitral/tufted (M/T) and juxtaglomerular cells, including external tufted (ET) and periglomerular (PG) cells. PG cells are heterogeneous in neurochemical expression and synaptic organization. We used a line of mice expressing green fluorescent protein under the control of the glutamic acid decarboxylase 65-kDa gene (GAD65+) promoter to characterize a neurochemically identified subpopulation of PG cells by whole cell recording and subsequent morphological reconstruction. GAD65+ GABAergic…
