Transcriptomic and Metabolomic Profiling Reveals the Antiproliferative Mechanism of Goose Serum and Plasma in SW1990 Cells.

Goose blood has anticancer properties and was recorded in ancient China, but the specific molecular mechanisms underlying this effect still require further exploration. In this study, SW1990 cells were treated with goose serum or plasma, and transcriptome analysis was performed to explore the function of goose blood on cancer cells. Metabolomic profiling was also performed on goose serum, goose plasma, chicken serum, and chicken plasma to identify the bioactive substances responsible for the anticancer effect. The study examined the effects of goose plasma and serum on SW1990 cells and compare

Targeting and monitoring cancer cells with cold atmospheric plasma and PET Imaging: a new theranostic approach.

Cold atmospheric plasma (CAP) has emerged as a promising tool for biomedical applications, including cancer research. In this study, we developed a micro‑sized CAP device designed for minimally painful subdermal plasma delivery in BALB/c mice without the need for anesthesia. The in vitro effects of CAP were evaluated in 4T1 breast cancer cells, and in vivo responses were assessed in tumor‑bearing mice. To quantitatively monitor treatment outcomes, 18F‑FDG PET imaging was employed to measure metabolic activity and residual tumor burden. CAP‑treated mice demonstrated a measurable reduction in tu

Positive selection of IgG over IgM plasma cells through BCR isotype-specific antigen presentation and signaling.

During a primary immune response, B cells can undergo isotype switching from an immunoglobulin M (IgM) B cell receptor (BCR) to an IgG BCR. B cells that have switched to IgG give rise to more bone marrow long-lived plasma cells (PCs) compared with those expressing IgM, but how BCR isotype-driven bias occurs remains unclear. Here, we found that IgG1-expressing germinal center B cells presented higher levels of antigen to T follicular helper cells, resulting in greater proliferation of IgG1 PCs than IgM PCs. BCR signaling through IgM induced more Bim-dependent apoptosis in IgM PCs, together lead…

Protocol for isotype-resolved 3D imaging and quantification of bone marrow plasma cells in murine femurs.

Bone marrow plasma cells maintain durable antibody responses, but their tissue localization and morphology are difficult to quantify in situ. Here, we present a protocol for isotype-resolved 3D imaging and quantification of bone marrow plasma cells in murine femurs. We describe steps for fixation, optimal cutting temperature (OCT) coumpound embedding, longitudinal opening of mouse femurs, whole-mount immunostaining, optical clearing, and confocal 2D/3D imaging. We detail an Imaris-based workflow for reproducible single-cell segmentation, isotype-resolved counting, and the extraction of morphol…

Celiac disease plasma cells recognize the active form of transglutaminase 2 with and without bound substrate.

Autoantibodies to the enzyme transglutaminase 2 (TG2) are characteristic of celiac disease, and TG2-specifc plasma cells are abundant in the celiac gut lesion. The production of antibodies by effector B cells (i.e. plasma cells) are believed to result from collaboration between TG2-specific B cells and gluten-specific CD4+ T cells - a collaboration facilitated by formation of TG2-gluten enzyme-substrate complexes. Since binding of substrate is associated with conformational changes in TG2, we sought to address whether the conformation of TG2 influences plasma cell reactivity. Using effector mo…

Cold atmospheric plasma restores fulvestrant sensitivity by inhibiting CCND3 in the drug-resistant breast cancer cells.

Cold atmospheric plasma (CAP) has demonstrated anti-proliferative activity in various cancer cells, yet its efficacy against drug-resistant cancer cells remains largely unexplored. This study investigates CAP's potential to restore drug sensitivity in fulvestrant-resistant breast cancer cells. Fulvestrant-resistant T47D/fulR and MCF-7/fulR cell lines were developed through 32-week drug exposure. CAP treatment effectively inhibited growth of both resistant cell lines and restored sensitivity to fulvestrant when used as pretreatment. Genome-wide expression analysis revealed that CAP significantl

Long-lived IgE plasma cells persist in secondary lymphoid tissues using a navitoclax-sensitive survival program.

Long-term allergies can exhibit persistent concentrations of circulating immunoglobulin E (IgE). Here, we examined the lifespan of IgE antibody-secreting cells (ASCs) to determine whether the IgE that sustains allergies receives contributions from long-lived cells or relies more heavily on constant ASC production. In mouse aeroallergy, IgE ASCs localized to the lungs, mediastinal lymph nodes, spleen, and bone marrow (BM). IgE ASC production continued for months after allergen exposure ceased. We identified long-lived IgE ASCs residing predominantly outside the BM, with a half-life exceeding 49

Blocking plasma cell fate enhances antigen-specific presentation by B cells to boost anti-tumor immunity.

B cells engage in anti-tumor immunity but how they contribute to cancer suppression remains unclear. We report that inhibiting plasma cell differentiation either in IgMi mice lacking Igh elements needed for antibody secretion or in mice with B cell-specific knockout of Blimp-1 (Blimp-1 BcKO) promotes rather than inhibits antitumor immunity and increases numbers of activated B cells. Deficiency of Blimp-1 in tumor-infiltrating B cells generates a unique transcription profile associated with expansion of mutated clones targeting cognate tumor cells. Major histocompatibility complex class II (MHC

Protein-Binding Patterns Drive the Transport of Mercury in Plasma and Red Blood Cells in Rats.

The fate of metals within biological systems is determined by their associated binding partners, specifically protein ligands. However, the mechanisms by which these protein targets regulate the transport of metal pollutants remain unclear. Here, protein-binding patterns were identified as drivers of the transport of mercuric compounds (methylmercury and inorganic mercury) in the bloodstream. We systematically investigated the transport of mercury (Hg) in rats following oral administration and analyzed the time-resolved patterns of Hg-binding proteins in the blood. Our findings demonstrated th

Chronic endometritis identified by plasma cells can often be diagnosed in patients with recurrent implantation failure.

PURPOSE: Is plasma cell (PC)-diagnosed chronic endometritis a common phenomenon in patients with recurrent implantation failure, and what can be learned from it? METHODS: In this retrospective case-control study at Wiesbaden Fertility Centre, 147 patients with recurrent implantation failure (RIF) underwent endometrial biopsy (EB) between January 2017 and December 2021 to rule out chronic endometritis (CE). For diagnosis, samples were labelled immunohistochemically with anti-CD138 and anti-MUM1 antibodies for the detection of PCs. The cut-off for the diagnosis of CE was at least five PCs per 1

Dissecting Causal Relationships Between Immune Cells, Plasma Metabolites, and PCOS: Evidence From Mediating Mendelian Randomization Analysis.

BACKGROUND: The relationship between Polycystic ovary syndrome (PCOS) and immune dysregulation, along with metabolic disturbances, remains unclear. This study used Mendelian Randomization (MR) to investigate causal relationships between immune cells, PCOS, and possible metabolite mediators. METHODS: We explored the genetic-level relationship between immune cells and PCOS, focusing on metabolites as potential mediators. Data from genome-wide association studies (GWAS) included 731 immune cell types (n=3757), 1400 plasma metabolites (n=8299), and PCOS cases (n=797) versus controls (n=140,558).

Gastrointestinal germinal center B cell depletion and reduction in IgA + plasma cells in HIV-1 infection.

UNLABELLED: Gastrointestinal (GI) B cells and plasma cells (PCs) are critical to mucosal homeostasis and the host response to HIV-1 infection. Here, high resolution mapping of human B cells and PCs sampled from the colon and ileum during both viremic and suppressed HIV-1 infection identified a reduction in germinal center (GC) B cells and follicular dendritic cells (FDCs) during HIV-1 viremia. IgA + PCs are the major cellular output of intestinal GCs and were significantly reduced during viremic HIV-1 infection. PC-associated transcriptional perturbations, including type I interferon signaling…

Accelerated plasma-cell differentiation in Bach2-deficient mouse B cells is caused by altered IRF4 functions.

Transcription factors BACH2 and IRF4 are both essential for antibody class-switch recombination (CSR) in activated B lymphocytes, while they oppositely regulate the differentiation of plasma cells (PCs). Here, we investigated how BACH2 and IRF4 interact during CSR and plasma-cell differentiation. We found that BACH2 organizes heterochromatin formation of target gene loci in mouse splenic B cells, including targets of IRF4 activation such as Aicda, an inducer of CSR, and Prdm1, a master plasma-cell regulator. Release of these gene loci from heterochromatin in response to B-cell receptor stimula…

Translocation (11;14) is a common cytogenetic abnormality in clonal plasma cells in monoclonal immunoglobulin deposition disease.

Monoclonal Immunoglobulin deposition disease (MIDD) is characterised by deposits of intact monoclonal light chains in the kidney leading to renal dysfunction. In this study, we retrospectively investigated the underlying plasma cell cytogenetic abnormalities in MIDD. CyclinD1 (11;14) translocation was identified in 12/27 (45%) patients. Among the patients without translocation, del13q and hyperdiploidy were the most common abnormalities. Patients in the non-t (11;14) group had a higher baseline light-chain ratio, higher proteinuria and lower eGFR as compared to patients with t (11;14). Haemato

Thiopurines impair the apical plasma membrane expression of CFTR in pancreatic ductal cells via RAC1 inhibition.

BACKGROUND AND AIMS: Thiopurine-induced acute pancreatitis (TIP) is one of the most common adverse events among inflammatory bowel disease patients treated with azathioprine (AZA), representing a significant clinical burden. Previous studies focused on immune-mediated processes, however, the exact pathomechanism of TIP is essentially unclear. METHODS: To model TIP in vivo, we triggered cerulein-induced experimental pancreatitis in mice receiving a daily oral dose of 1.5 mg/kg AZA. Also, freshly isolated mouse pancreatic cells were exposed to AZA ex vivo, and acinar cell viability, ductal and
