This study was aimed at developing a novel platform for tetravalent conjugation of 4-arm polyethylene glycol (PEG) with an antisense oligonucleotide (ASO). The ASO technology has several limitations, such as low cellular uptake, poor nuclease stability, and short half-life. PEG-conjugated ASOs may result in an improvement in the pharmacokinetic behavior of the drug. Moreover, PEGylation can reduce enzymatic degradation and renal excretion of the conjugates, thereby, increasing its blood stability and retention time. In this study, we successfully synthesized PEG-ASO conjugate consisting of 4-a

BACKGROUND: 5-methylcytosine (5-mC) RNA methylation is a prevalent post-transcriptional modification affecting both coding and non-coding RNAs. 5-mC methylation can regulate the expression level of relevant genes and biological processes by affecting RNA stability, translation efficiency, spatial structure, protein interactions, and other processes. Emerging evidence highlights its critical role in cancer biology. MAIN BODY: Abnormal changes in 5-mC methylation lead to aberrant expression of oncogenes or inactivation of suppressor genes, which in turn affects the growth and differentiation of …

RNA 5-methylcytosine (m5C) modification plays a critical role in multiple diseases; however, its function in hyperuricaemic nephropathy (HN) remains unclear. Here, we show that renal m5C levels are markedly decreased and the m5C methyltransferase NOP2/Sun RNA methyltransferase 5 (NSUN5) is downregulated in HN mice. Conditional knock-in (cKI) mice and mouse tubular epithelial cells overexpressing NSUN5 exhibit resistance to uric acid (UA), thereby alleviating kidney injury, inflammation, and fibrosis in HN. Bisulphite sequencing coupled with RNA sequencing identifies stearoyl-CoA desaturase-1 (

Long non-coding RNAs (lncRNAs) have emerged as critical regulators of the cancer epitranscriptome, orchestrating post-transcriptional gene regulation through dynamic interactions with RNA-modifying enzymes and RNA-binding proteins. Acting as molecular guides, scaffolds, decoys, and organizational hubs, lncRNAs regulate the deposition, removal, and interpretation of epitranscriptomic marks, including N6-methyladenosine (m6A), 5-methylcytosine, pseudouridine, and adenosine-to-inosine editing. Through these interactions, lncRNAs influence RNA stability, splicing, translation, localization, and de…

Age-related decline in oocyte quality increases the risk of infertility, miscarriage, and birth defects. Mitochondrial dysfunction is a key contributor to this decline. Here, we report that oocyte-specific deletion of Uba3, which encodes the catalytic subunit of the E1 NEDDylation-activating complex, causes sterility in mice. Fully grown, germinal vesicle-stage Uba3 conditional knockout oocytes exhibit mitochondrial dysfunction, including elevated reactive oxygen species, impaired oxidative phosphorylation, and depletion of mitochondrially encoded RNA transcripts. Proteomic analysis identified

Circular RNA (circRNA) represents a promising class of RNAs for sustained protein expression and vaccine applications. However, current lipid nanoparticle (LNP) platforms remain poorly optimized for efficient intramuscular circRNA delivery, as they are primarily designed for the delivery of small interfering RNA and linear messenger RNA with strong off-target expression in the liver. In this study, based on the clinical benchmark SM-102 LNP, we systematically optimized lipid components and microfluidic parameters for better intramuscular circRNA delivery. Through in vitro and in vivo screening

Ubiquitination dynamically regulates protein stability and signaling networks through post-translational modifications. Makorin ring finger protein 1 (MKRN1) is a unique member of the MKRN family that functions as E3 ubiquitin ligase and RNA-binding protein. It maintains cellular homeostasis via a dual regulatory mechanism involving ubiquitination and RNA biology. This review summarizes the regulatory mechanisms of MKRN1 in core pathways, such as p53-p21, Wnt/β-catenin, transforming growth factor beta, phosphatidylinositol 3 kinase/protein kinase, nuclear factor kappa B, and various pathologic

Autophagy is a conserved degradation process crucial to maintaining the primary function of cellular and organismal metabolism. Impaired autophagy could develop numerous diseases, including cancer, cardiomyopathy, neurodegenerative disorders, and aging. N6-methyladenosine (m6A) is the most common RNA modification in eukaryotic cells, and the fate of m6A modified transcripts is controlled by m6A RNA binding proteins. m6A modification influences mRNA alternative splicing, stability, translation, and subcellular localization. Intriguingly, recent studies show that m6A RNA methylation could alter

The timing, dosage and location of gene expression are fundamental determinants of brain architectural complexity. In neurons, this is, primarily, achieved by specific sets of trans-acting RNA-binding proteins (RBPs) and their associated factors that bind to specific cis elements throughout the RNA sequence to regulate splicing, polyadenylation, stability, transport and localized translation at both axons and dendrites. Not surprisingly, misregulation of RBP expression or disruption of its function due to mutations or sequestration into nuclear or cytoplasmic inclusions have been linked to the

Genes encoding toxin-antitoxin (TA) systems are near ubiquitous in bacterial genomes and they play key roles in important aspects of bacterial physiology, including genomic stability, formation of persister cells under antibiotic stress, and resistance to phage infection. The CptIN locus from Eubacterium rectale is a member of the recently-discovered Type III class of TA systems, defined by a protein toxin suppressed by direct interaction with a structured RNA antitoxin. Here, we present the crystal structure of the CptIN protein-RNA complex to 2.2 Å resolution. The structure reveals a new het

Ectromelia virus encodes a protein which is homologous to the product of the vaccinia virus host range gene, K1L, except for eight conservative and two non-conservative substitutions and an additional threonine residue at the carboxyl terminus. Unlike the vaccinia virus gene, the ectromelia virus homolog failed to support optimal virus replication in RK-13 cells and appeared to be expressed 20-fold less efficiently. This lower level of expression was not due to the genetic background of the virus, K1L RNA transcription, sequence of the K1L RNA leader, or stability of K1L RNA or protein. Infect

Circular RNAs (circRNAs) represent an emerging class of noncoding RNAs characterized by their covalently closed-loop structure and generated through the back-splicing event. This structural configuration provides circRNAs with enhanced stability and unique functional properties. Functionally, circRNAs act as protein scaffolds, enhance protein activity, can be translated, and frequently serve as competing endogenous RNAs (ceRNAs), particularly by sponging microRNAs (miRNAs) and modulating downstream gene expression. Since these regulatory capacities have significant implications in cancer, neur

The RNA exosome plays a critical role in surveilling nuclear mRNA biogenesis and regulating co-translational mRNA decay in the cytoplasm. Unlike canonical translation, repeat-associated non-AUG (RAN) translation of a GGGGCC hexanucleotide repeat expansion (HRE) within an intron of the C9orf72 locus leads to the synthesis of neurotoxic dipeptide-repeat (DPR) proteins, contributing to the pathogenesis of frontotemporal dementia and amyotrophic lateral sclerosis (C9-ALS/FTD). However, it remains unclear whether aberrant RAN translation is monitored and regulated co-translationally or how C9orf72

BACKGROUND: Apolipoprotein B (apoB) is essential for lipoprotein assembly and secretion and plays a central role in the development of cardiovascular disease and metabolic dysfunction-associated steatotic liver disease. Although apoB protein degradation during very-low-density lipoprotein maturation has been extensively studied, the regulation of Apob mRNA stability under physiological and pathological conditions remains unexplored.

Mesothelioma is a rare and aggressive cancer linked to asbestos exposure and characterized by rapid metastasis and poor prognosis. Inhibition of adenosine deaminase acting on dsRNA 2 (ADAR2) RNA binding but not ADAR2 editing has shown antitumor effects in mesothelioma. Natural compounds from the Traditional Chinese Medicine (TCM) database were docked to the RNA-binding interface of ADAR2's second dsRNA binding domain (dsRBD2), and their drug-likeness and predicted safety were assessed. RNA N6-methyladenosine (m6A) deposition by the METTL3-METTL14 complex regulates RNA metabolism and thereby influences cell differentiation and stress responses. Dysregulated m6A deposition has been implicated in multiple cancers, where METTL3-METTL14 supports oncogenic programs. Yet the molecular basis of RNA recognition by this complex remains unclear, in part due to the lack of an RNA-bound structure of the full assembly. Here, we combined native mass spectrometry (MS), molecular modeling, biochemical assays, and cell-based studies to define how METTL3-METTL14 engages RNA and how this proces
