Development and validation of genomic and epigenomic signatures associated with tumor immune microenvironment in hepatoblastoma.

Источник пометил публикацию как отозванную

BACKGROUND: This study aimed to probe and verify aberrantly methylated and expressed genes in hepatoblastoma and to analyze their interactions with tumor immune microenvironment. METHODS: Aberrantly methylated and expressed genes were obtained by comprehensively analyzing gene expression and DNA methylation profiles from GSE81928, GSE75271 and GSE78732 datasets. Their biological functions were predicted by the STRING and Metascape databases. CIBERSORT was utilized for inferring the compositions of tumor-infiltrating immune cells (TIICs) in each sample. Correlation between hub genes and immune

Restoring the Immunity in the Tumor Microenvironment: Insights into Immunogenic Cell Death in Onco-Therapies.

Immunogenic cell death (ICD) elicited by cancer therapy reshapes the tumor immune microenvironment. A long-term adaptative immune response can be initiated by modulating cell death by therapeutic approaches. Here, the major hallmarks of ICD, endoplasmic reticulum (ER) stress, and damage-associated molecular patterns (DAMPs) are correlated with ICD inducers used in clinical practice to enhance antitumoral activity by suppressing tumor immune evasion. Approaches to monitoring the ICD triggered by antitumoral therapeutics in the tumor microenvironment (TME) and novel perspective in this immune sy…

The emerging role of miRNA in the perturbation of tumor immune microenvironment in chemoresistance: Therapeutic implications.

Chemoresistance is a major hindrance in cancer chemotherapies, a leading cause of tumor recurrence and cancer-related deaths. Cancer cells develop numerous strategies to elude immune attacks and are regulated by immunological factors. Cancer cells can alter the expression of several immune modulators to upregulate the activities of immune checkpoint pathways. Targeting the immune checkpoint inhibitors is a part of the cancer immunotherapy altered during carcinogenesis. These immune modulators have the capability to reprogram the tumor microenvironment, thereby change the efficacy of chemothera…

The Prognostic Value of Immune Factors in the Tumor Microenvironment of Penile Squamous Cell Carcinoma.

The host's immune system plays a pivotal role in many tumor types, including squamous cell carcinomas (SCCs). We aim to identify immunologic … significantly unfavorable for DSS (HR 5.0, p < 0.01). These results emphasize the complexity of the tumor microenvironment in penile cancer and point toward several possible immunotherapy targets. Here described immune factors can aid risk-stratification and should be evaluated in clinical immunotherapy studies to ultimately lead to patient tailored treatment.

The changes of angiogenesis and immune cell infiltration in the intra- and peri-tumoral melanoma microenvironment.

Malignant melanoma (MM) urgently needs identification of new markers with better predictive value than currently-used clinical and histological parameters. Cancer cells stimulate the formation of a specialized tumor microenvironment, which reciprocally affects uncontrolled proliferation and migration. However, this microenvironment is heterogeneous with different sub-compartments defined by their access to oxygen and nutrients. This study evaluated microvascular density (MVD), CD3+ lymphocytes (TILs) and FOXP3+ T-regulatory lymphocytes (Tregs) on formalin-fixed paraffin-embedded tissue section…

The Involvement of RCAS1 in Creating a Suppressive Tumor Microenvironment in Patients with Salivary Gland Adenocarcinoma.

The tumor microenvironment is the tissue that determines the growth and progression of the tumor as well as its ability to initiate metastases. The aim of the present study has been to evaluate the role of RCAS1 in creating the suppressive tumor microenvironment in cases of parotid adenocarcinoma. The tissue samples of salivary gland adenocarcinomas and their stroma and the palatine tonsils which constituted the reference tissue sample group were obtained during routine surgical procedures. The immunoreactivity of RCAS1, CD3, CD25, CD68, CD69, and Foxp3 antigens was then evaluated by using the…

Impact of the tumor microenvironment on prognosis in follicular lymphoma is dependent on specific treatment protocols.

BACKGROUND: The clinical behavior of follicular lymphoma is largely determined by properties of the non-malignant tumor microenvironment. The precise nature of the cell populations is still unclear and published data on their prognostic significance are highly conflicting. This may be partly due to heterogeneous composition and treatments. DESIGN AND METHODS: Pre-treatment biopsy samples of patients with follicular lymphoma treated in an EORTC/BNLI trial comparing fludarabine to cyclophosphamide, vincristine and prednisone (CVP) chemotherapy could be retrieved for 61 patients in five European…

p38α orchestrates BRAFV600E-melanoma progression and immunotherapy response via tumor fitness and immune microenvironment modulation.

Understanding how tumor cells shape the tumor microenvironment (TME) and evade the host immune response is critical for developing novel therapeutic combinations. In this study, we identify tumor cell p38α as a therapeutic target that restricts antitumor immunity and contributes to resistance against immune-checkpoint inhibitors (ICIs). Genetic analyses of mouse and human melanomas reveal alterations in the mitogen-activated protein kinase (MAPK)-stress pathway, including upregulation of MAPK14. We demonstrate a dual role for p38α in ultraviolet-B (UVB)-induced Braf V600E melanoma, acting as a…

Multiplex hyperspectral IHC: A 12-plex assay for high-dimensional analysis of the tumor microenvironment.

The immune component of the tumor microenvironment (TME) is highly prognostic of patient outcome, further understanding of TME and the spatial organization of the immune cells within the TME is needed for better patient prognosis and treatment planning. Current brightfield immunohistochemistry techniques quantify immune cell counts and density but generally cannot assess the spatial relationship between tumor and immune cells due to the limited number of markers on a single slide. Here, we present a brightfield-based multiplexed immunohistochemistry workflow that combines multiple labels per s…

Smart Interactive System for the Enhanced Bioimaging and Multi-Drug Synergistic Therapy in Tumor-Microenvironment.

Tumor microenvironment (TME) interactive platforms are promising for accurate diagnostics and therapeutics. Herein, a smart interactive system (G⊂H) consisting of a hypoxia-acid dual triggered multifunctional guest (G) and an amphipathic metal-organic capsule (H) was successfully established for bioimaging and treatment. Hydrophobic G is encapsulated in cavity of H to form the homogeneous soluble G⊂H, which enhances its accumulation in tumor cells, controls the interactive regulation between G⊂H and TME, and facilitates the sensitivity of tumor cell therapy. When triggered in TME, G⊂H selectiv…

Epigenetic Priming Sensitizes Metastatic Breast Cancer to Checkpoint Blockade by Rewiring the Tumor-Immune Microenvironment.

Immune checkpoint blockade (ICB) has shown limited efficacy in metastatic breast cancer. Resistance mechanisms involve suppressive myeloid and stromal populations in the tumor microenvironment (TME). In this issue of Cancer Research, Gonzalez and colleagues integrate single-cell transcriptomics, spatial proteomics, functional immunology, and mathematical modeling to dissect how combined treatment with the histone deacetylase (HDAC) inhibitor entinostat and anti-PD-1/anti-CTLA-4 blockade (EPC) sensitizes these refractory tumors. Work using metastatic mouse models shows that the antitumor respon…

Spermine oxidase-DOX conjugates reshape tumor microenvironment via carbonyl stress to potentiate bladder cancer chemotherapy.

The tumor's antioxidant defenses restrict the efficacy of standard therapies, resulting in compromised treatment responses and systemic tolerance. Conventional redox-disrupting strategies, designed to generate reactive oxygen species (ROS), are limited by both their reliance on single generation methods and the rapid clearance. Herein, we report an innovative strategy that employs spermine oxidase (SMOX) to catalyze the production of endogenous and highly cytotoxic acrolein, thereby creating a destructive carbonyl stress microenvironment to potentiate antineoplastic drug's efficacy. Tumor cell derived CCL20 exacerbates the immunosuppressive microenvironment by recruiting CCR6+ Tregs in pancreatic cancer.

Pancreatic cancer has a dismal prognosis, largely due to resistance to all current therapeutic modalities, including prevailing immunotherapy. Deciphering the mechanisms underlying the immunosuppressive tumor microenvironment is pivotal for developing effective therapeutic strategies. In this study, we found that the expression of CCL20 is negatively correlated with the infiltration of CD8+ T cells, and consistently, patients with higher CCL20 expression have a worse prognosis. CCL20, as a secreted ligand protein regulated by SP5, can bind to its cognate receptor CCR6 in the tumor microenviron…
