Efficacy of Procox® oral suspension for dogs (0.1% emodepside and 2% toltrazuril) against experimental nematode (Toxocara cati and Ancylostoma tubaeforme) infections in cats.

Two exploratory studies were performed to determine the optimum therapeutic dose of Procox(®) for the removal of experimental infection with mature adult Toxocara (T.) cati and Ancylostoma (A.) tubaeforme in kittens. Procox(®) is a new oral suspension containing a combination of the nematocidal and coccidiocidal active principles emodepside (0.1 %) and toltrazuril (2 %).In the first study, 18 eight-weeks-old kittens were inoculated with 450 L3 larvae of T. cati. 56 days after infection, the kittens were allocated to three treatment groups and were treated with 0.5 mg emodepside/kg body weight

[Prevalence and factors associated with geohelminth infections in children living in municipalities with low HDI in North and Northeast Brazil].

Stool surveys were conducted to estimate the prevalence and identify risk factors for geohelminth infections among children in ten Brazilian municipalities with low human development indices (HDI). Socioeconomic and environmental data were obtained from the children's parents or guardians, and stool samples were examined. The proportion of geohelminth infections according to target variables was calculated. Risk factors were evaluated using multilevel logistic regression. Of the 2,523 children, 36.5% were infected with one or more geohelminths (Ascaris lumbricoides, 25.1%; hookworm, 15.3%; Tri

Helminthic infections in school children in Puducherry, South India.

BACKGROUND/PURPOSE: Intestinal parasites are widely prevalent in developing countries, probably due to poor sanitation and inadequate personal hygiene. It is estimated that as much as 60% of the world's population is infected with intestinal parasites, which may play a significant role in morbidity due to intestinal infections. This study was designed to estimate the prevalence of helminthic infections among school children in Puducherry, South India. METHODS: The prevalence of helminthic infections was studied in 1,172 children in public schools in Puducherry (Pondicherry), South India, betw

The impact of concurrent and treated Ancylostoma ceylanicum hookworm infections on the immunogenicity of a recombinant hookworm vaccine in hamsters.

BACKGROUND: The effect of concurrent (active) and treated hookworm infections on the immunogenicity of vaccination with the recombinant fusion protein Ay-Ancylostoma-secreted protein 2 was analyzed in the Golden Syrian hamster. METHODS: Hamsters were infected with the hookworm Ancylostoma ceylanicum and vaccinated with the recombinant protein, with Quil A used as adjuvant. As controls, hookworm-infected hamsters were treated with the anthelmintic drug pyrantel pamoate before vaccination. Naive hamsters (i.e., those with neither previous hookworm infections nor treatment) were also vaccinated.

Human enteric infection with Ancylostoma caninum: hookworms reappraised in the light of a "new" zoonosis.

Recent studies in northeastern Australia indicate that enteric infection with Ancylostoma caninum is a leading cause of human eosinophilic enteritis. Much more frequent accompaniments of this infection are obscure abdominal pain with or without blood eosinophilia, while a large part of the population is probably infected asymptomatically. These conclusions are based on extensive serological investigations in patients and control subjects, as well as 15 cases in which single, adult hookworms were identified in situ in patients. In no case has more than one worm been identified, and none has bee

[Report on intestinal parasitosis in adults in an urban population of the Central African Republic].

The authors point out the frequency of intestinal parasitism in Bangui, biggest town and capital of the CAR. Out of 3,352 samples, 1,570 were found positive. The main parasites found were: Ankylostoma duodenale, 26,7%; Schistosoma mansoni, 20,8%; Entamoeba histolytica, 18,2%. The authors underline the polyparasitism. They also emphazide the differences with another study carried out in Algiers and surrounding areas.

Feline respiratory tract disease in Louisiana.

An investigation of feline respiratory tract disease (FRD) was made involving 65 affected cats (group 1) and 65 control cats (group 2) matched for age, sex, and residence. Feline calicivirus strains (FCV), feline herpesvirus-1 (FHV-1), or both, were isolated from 59% of group 1 cats and from 7% of group 2 cats. Isolation of these viruses showed a significantly greater (P < 0.05) relative risk for group 1 cats. Virus isolations were three times more frequent from males than from females older than 1 year (both groups), but were equally distributed between males and females younger than 1 year.

ASSOCIATION BETWEEN MACROCYCLIC LACTONE AND BENZIMIDAZOLE RESISTANCE IN THE DOG HOOKWORM ANCYLOSTOMA CANINUM.

Anthelmintic resistance is a growing threat to the treatment and control of parasitic nematode infections in both humans and animals. In the canine hookworm, Ancylostoma caninum, multi-anthelmintic drug-resistant (MADR) isolates have been identified, but the molecular mechanisms underlying resistance and the potential for these mechanisms to promote cross-resistance between drug classes remain unclear. In this study, we report evidence for a previously uncharacterized association between resistance to macrocyclic lactones (MLs) and benzimidazoles (BZs) in A. caninum. In a wild-type isolate wit

Development of multi-ARMS-qPCR method for detection of hookworms from cats and dogs.

Hookworms are blood-sucking nematodes that infect dogs, cats, and humans, causing iron-deficiency anemia, abdominal pain, diarrhea, and skin inflammation. Amplification refractory mutation system (ARMS) is a modified technology based on allele-specific PCR, which is widely used in mutation detection and genotyping. However, no data about ARMS application in hookworm detection. This study aims to establish a multi-ARMS-qPCR method for the detection of three hookworm species from dogs and cats. A universal forward primer and three specific primers (ARMS-Cey, ARMS-Can, and ARMS-Tub) were designed

Old friends and friendly fire: Pregnancy, hookworm infection, and anemia among tropical horticulturalists.

OBJECTIVES: Despite public health concerns about hookworm infection in pregnancy, little is known about immune profiles associated with hookworm (Necator americanus and Ancylostoma duodenale) infection during pregnancy. Fetal tolerance requirements may constrain maternal immune response to hookworm, thereby increasing susceptibility to new infections or increasing hemoglobin loss. To explore this possibility, we study systemic immune response and hemoglobin levels in a natural fertility population with endemic helminthic infection. METHODS: We used Bayesian multilevel models to analyze mixed

Development and Evaluation of a Multiplex Quantitative Real-Time Polymerase Chain Reaction for Hookworm Species in Human Stool.

Hookworm disease caused by Necator americanus, Ancylostoma duodenale, and Ancylostoma ceylanicum affects half a billion people worldwide. The prevalence and intensity of infection of individual hookworm species are vital for assessing morbidity and generating targeted intervention programs for their control. The present study aims to evaluate a multiplex real-time quantitative PCR (qPCR) assay to determine the prevalence and egg intensity of all three hookworm species and compare this with standard microscopy and published genus-based conventional and real-time multiplex qPCRs. Performance of

Ac-AP-12, a novel factor Xa anticoagulant peptide from the esophageal glands of adult Ancylostoma caninum.

Immunoscreening an Ancylostoma caninum cDNA library with canine hookworm-infected dog serum resulted in the isolation of a 461 bp cDNA encoding Ac-AP-12, a new 9.1 kDa anticoagulant peptide (100 amino acids) with 43-69% amino acid homology to other nematode anticoagulant peptides (NAPs) from Ancylostoma hookworms. Messenger RNA transcription and expression of Ac-AP-12 was unique to the adult stage of A. caninum. The yeast expressed recombinant Ac-AP-12 demonstrated potent anticoagulant activity on human blood plasma in a concentration dependent manner, and was shown to specifically inhibit hum

Resistance of dogs to reinfection with Ancylostoma ceylanicum following anthelmintic therapy.

A model of human hookworm infection has been developed which shows that dogs with chronic hookworm infection are considerably resistant to reinfection one month after the termination of the primary infection with anthelmintics. Challenge and control dogs were infected with 1,800 larvae and the infection was followed for six weeks. When compared with control dogs, faecal egg excretion and intestinal adult worm burdens in challenge dogs were reduced by 85% and 77%, respectively. Infection had no significant effect on haemoglobin concentrations, total white cell counts, platelet levels or spontan

Dose-titration studies of ivermectin against experimental Ancylostoma caninum and Uncinaria stenocephala infections.

Dose-titration trials of ivermectin were conducted on pups with dual experimental infections of 4th-stage larvae or adult Ancylostoma caninum and Uncinaria stenocephala. Ivermectin was administered orally or subcutaneously at dosages of 0.006, 0.012, or 0.024 mg/kg of body weight. Maximal or near maximal (greater than or equal to 96% to 100%) anthelmintic effect was observed for both stages of development for each hookworm species by either route of administration at a dosage of 0.024 mg/kg. Responses for all of the aforementioned categories were linearly related to increasing log dosage of iv

Hookworm burdens and faecal egg counts: an analysis of the biological basis of variation.

The relationships between various measures of faecal egg output and hookworm burdens were investigated in 84 villagers from West Bengal with mixed Necator americanus and Ancylostoma duodenale infections. The pattern of day to day variability in egg counts from individuals can be characterized by the linear relationship between the logarithms of the variances and means. Egg output is shown to be non-periodic in long runs of day-to-day records. The distribution of worm numbers per host is well described by the negative binomial probability model, and the relationship between per capita egg outpu
