Time Course of Electrocortical Processing of Food Stimuli in Adolescent Anorexia Nervosa.

OBJECTIVE: Anorexia nervosa (AN) typically manifests during adolescence and is linked to significant psychological and physical sequelae. A better understanding of the underlying maintenance mechanisms of adolescent AN is therefore crucial. Cognitive behavioral theories emphasize the influence of cognitive biases in the processing of disorder-relevant stimuli. METHOD: The current study investigated attentional bias to high- and low-calorie food in female adolescents with AN (n = 31) and without AN (n = 35). An event-related potentials study utilizing prolonged stimulus presentation in an atte

Molecular and functional asymmetry in Cckar-expressing vagal sensory neurons.

The vagus nerves are important carriers of sensory information from the viscera to the brain. Emerging evidence suggests that sensory signaling through the right, but not the left, vagus nerve evokes striatal dopamine release and reinforces appetitive behaviors. However, the extent to which differential gene expression within vagal sensory neurons may underlie this asymmetric reward-related signaling is unknown. Here, we use single-nucleus RNA sequencing, in situ hybridization, and calcium imaging to identify a single cluster of neurons, defined by co-expression of Chrna3 (nicotinic acetylchol…

[Clinical practice guidelines for Prader-Willi syndrome].

Prader-Willi syndrome (PWS) is the first multi-systemic genetic disorder known to be caused by imprinting defect. The clinical manifestations of PWS vary with age. At the prenatal stage, decreased fetal movements are frequent. The major clinical manifestations during neonatal period include hypotonia, weak cry, poor suck and feeding difficulties. Growth retardation and delayed language and motor development are observed during infancy. Short stature, small hands and feet, cognitive deficiency are noticed in the childhood. At adolescence, prominent growth retardation, obesity, gonadal dysplasia

GEOFFREY HARRIS PRIZE LECTURE 2018: Novel pathways regulating neuroendocrine function, energy homeostasis and metabolism in humans.

The discovery of leptin, an adipocyte-secreted hormone, set the stage for unraveling the mechanisms dictating energy homeostasis, revealing adipose tissue as an endocrine system that regulates appetite and body weight. Fluctuating leptin levels provide molecular signals to the brain regarding available energy reserves modulating energy homeostasis and neuroendocrine response in states of leptin deficiency and to a lesser extent in hyperleptinemic states. While leptin replacement therapy fails to provide substantial benefit in common obesity, it is an effective treatment for congenital leptin d

Cancer-induced anorexia and malaise are mediated by CGRP neurons in the parabrachial nucleus.

Anorexia is a common manifestation of chronic diseases, including cancer. Here we investigate the contribution to cancer anorexia made by calcitonin gene-related peptide (CGRP) neurons in the parabrachial nucleus (PBN) that transmit anorexic signals. We show that CGRPPBN neurons are activated in mice implanted with Lewis lung carcinoma cells. Inactivation of CGRPPBN neurons before tumor implantation prevents anorexia and loss of lean mass, and their inhibition after symptom onset reverses anorexia. CGRPPBN neurons are also activated in Apcmin/+ mice, which develop intestinal cancer and lose we

New centrally acting agents for appetite control: from biological mechanisms to clinical efficacy.

BACKGROUND: Obesity is one of the major problems of health policy in different countries. Pharmacological attempts have been made to help affected people without a definitive solution. Some agents--either with peripheral or central effect--are available in the market. On July 2012, the FDA approved two novel preparations for obese patients: (1) topiramate-phentermine--the first one an anticonvulsant and the second one a sympathomimetic amine--and (2) lorcaserin, a 5-HT2CR agonist. Both preparations emerged as new options for weight management. SCOPE: Based on the complex biology of eating beh

Evaluation of the safety of a combination of oral administration of phenylbutazone and firocoxib in horses.

Simultaneous administration of a nonselective COX inhibitor and a COX-2 specific NSAID has not been previously reported in horses. The goal of this study was to determine the safety of a 10-day dosage regimen of phenylbutazone and firocoxib, both at their standard dosages, in horses. Six horses were administered 2.2 mg/kg of phenylbutazone and 0.1 mg/kg of firocoxib by mouth, daily for 10 days. Horses were assessed daily for changes in behavior, appetite, fecal consistency, signs of abdominal pain, and oral mucous membrane ulceration. Horses were assessed prior to and on the last day of treatm

Behavior of toads, Bufo bufo, in a dynamic environment.

Susceptibility to motion sickness was tested by exposing free moving toads to rotation of a stimulator modeled after an amusement park Ferris Wheel. The stimulator provided a gentle stimulation of frequency 0.25 Hz and centrifugal acceleration 0.143 g during 120 min or more without external visual cues. No emetic or prodromal behavioral response was elicited during or after rotation. During rotation the amount of motor activity in most toads increased evidently. The most active toads attempted to climb out of the test chamber. It was inferred that experimental rotation was rather a stressful s

A review of modafinil film-coated tablets for attention-deficit/hyperactivity disorder in children and adolescents.

Modafinil, a wakefulness-promoting agent unrelated to classical sympathomimetic stimulants, has been studied in a total of 933 children and adolescents as a treatment for attention-deficit/hyperactivity disorder (ADHD). Several studies, including three double-blind, placebo-controlled studies with intent-to-treat analyses, have demonstrated the efficacy of modafinil film-coated tablets in reducing symptoms of ADHD and associated problem behaviors in children and adolescents. Modafinil is generally well tolerated, with adverse events (such as insomnia, headache, loss of appetite, weight loss, a

Electrophysiological characterization of responses from gustatory receptor neurons of sensilla chaetica in the moth Heliothis virescens.

Discrimination of edible and noxious food is crucial for survival in all organisms. We have studied the physiology of the gustatory receptor neurons (GRNs) in contact chemosensilla (insect gustatory organs) located on the antennae of the moth Heliothis virescens, emphasizing putative phagostimulants and deterrents. Sucrose and the 2 bitter substances quinine and sinigrin elicited responses in a larger proportion of GRNs than inositol, KCl, NaCl, and ethanol, and the firing thresholds were lowest for sucrose and quinine. Variations in GRN composition in individual sensilla occurred without any

Preferential saline or water intake by pinealectomized, adrenalectomized, and pinealectomized-adrenalectomized male rats.

Because the pineal gland appears to be involved in the regulation of body electrolytes, saline or water preference was investigated in pinealectomized, adrenalectomized, pinealectomized-adrenalectomized, and sham-operated (SO) control rats. The pinealectomized, adrenalectomized, and pinealectomized-adrenalectomized rats exhibited increased saline consumption during the two postoperative weeks when compared with the SO-control animals. Water intake was similar for pinealectomized and SO-control rats, but decreased for pinealectomized-adrenalectomized, and adrenalectomized rats. Although total f

Chronic administration of sulbutiamine improves long term memory formation in mice: possible cholinergic mediation.

Thiamine deficiency in both man and animals is known to produce memory dysfunction and cognitive disorders which have been related to an impairment of cholinergic activity. The present experiment was aimed at testing whether, inversely, chronic administration of large doses of sulbutiamine would have a facilitative effect on memory and would induce changes in central cholinergic activity. Accordingly mice received 300 mg/kg of sulbutiamine daily for 10 days. They were then submitted to an appetitive operant level press conditioning test. When compared to control subjects, sulbutiamine treated

Activation of trace amine-associated receptor 1 attenuates schedule-induced polydipsia in rats.

Trace Amine Associated Receptor 1 (TAAR1) is a novel pharmacological target. TAAR1 are well-documented to play a modulatory role in the dopaminergic system. In spite of a growing number of studies of TAAR1 effects, little is still known about the behavioral pharmacology of TAAR1 ligands, including effects of repeated TAAR1 agonist administration. The present study appears to be the first that estimated the action of TAAR1 agonists on schedule-induced polydipsia, a type of adjunctive behavior, which is considered to be useful for evaluating certain aspects of obsessive-compulsive and related di

Behavioral problems may be associated with multilevel lumbosacral stenosis in military working dogs.

Behavioral and spinal problems have been reported to be important causes of early retirement in military working dogs, however studies on possible relationships between these two problems are currently lacking. The aim of this retrospective, observational, cross-sectional study was to test associations between reported behavioral problems and computed tomographic (CT) diagnoses of lumbosacral (LS) stenosis in a sample of military working dogs. Data archives at a tertiary referral military working dog hospital were searched for dogs that had LS CT scans acquired during the period of June 2013-

Neurochemical mechanism of action of drugs which modify feeding via the serotoninergic system.

The neurochemical mechanisms by which drugs acting on central serotoninergic system modify feeding were reviewed. Fenfluramine, a clinically effective appetite suppressant, releases serotonin from nerve terminals and inhibits its reuptake, and considerable evidence suggests that these effects mediate its anorectic activity. The D isomer of fenfluramine is particularly specific in affecting serotonin mechanisms and causing anorexia. Transmitters other than serotonin such as acetylcholine, catecholamines and GABA are also affected by systemic administration of fenfluramine, but some of these eff

Evaluation of the Relationship Between Orexin A, Peptide YY, AgRP, and POMC Levels and Sleep Disorders in Children with Malnutrition.

BACKGROUND: Malnutrition and sleep disturbances are common in childhood and are associated with neuropeptides that regulate appetite and circadian rhythms. Hypothalamic peptides such as orexin A, agouti-related protein (AgRP), proopiomelanocortin (POMC), and peptide YY (PYY) play important roles in energy balance and eating behavior; however, their specific functions in pediatric malnutrition remain unclear. This study aimed to determine the levels of these peptides in malnourished children and to examine their relationship with eating and sleep behaviors. METHODS: This case-control, cross-se

Cues play a critical role in estrous cycle-dependent enhancement of cocaine reinforcement.

While preclinical work has aimed to outline the neural mechanisms of drug addiction, it has overwhelmingly focused on male subjects. There has been a push in recent years to incorporate females into existing addiction models; however, males and females often have different behavioral strategies, making it important to not only include females, but to develop models that assess the factors that comprise female drug addiction. Traditional self-administration models often include light or tone cues that serve as discriminative stimuli and/or consequent stimuli, making it nearly impossible to dise

Mu Opioid Receptors in Gamma-Aminobutyric Acidergic Forebrain Neurons Moderate Motivation for Heroin and Palatable Food.

BACKGROUND: Mu opioid receptors (MORs) are central to pain control, drug reward, and addictive behaviors, but underlying circuit mechanisms have been poorly explored by genetic approaches. Here we investigate the contribution of MORs expressed in gamma-aminobutyric acidergic forebrain neurons to major biological effects of opiates, and also challenge the canonical disinhibition model of opiate reward. METHODS: We used Dlx5/6-mediated recombination to create conditional Oprm1 mice in gamma-aminobutyric acidergic forebrain neurons. We characterized the genetic deletion by histology, electrophys
