Carbonic anhydrase inhibitors as antitumor/antimetastatic agents: a patent review (2008-2018).

INTRODUCTION: Human carbonic anhydrases (CA, EC 4.2.1.1) IX and XII are tumor-associated proteins, being part of the molecular machinery that tumor cells build as adaptive responses to hypoxia and acidic conditions characteristic of the 'glycolytic shift' of many tumors. A wealth of research depicts CA IX and CA XII as biomarkers and therapeutic targets for various cancer types. AREAS COVERED: The review presents an overview of the role of CA IX and CA XII in hypoxic tumors physio-pathology as well as the principal molecular, structural, and catalytic features of both isozymes. The review the

Stabilizer Slicing: Coherent Error Cancellations in Low-Density Parity-Check Stabilizer Codes.

Coherent errors are a dominant noise process in many quantum computing architectures. Unlike stochastic errors, these errors can combine constructively and grow into highly detrimental overrotations. To combat this, we introduce a simple technique for suppressing systematic coherent errors in low-density parity-check stabilizer codes, which we call stabilizer slicing. The essential idea is to slice low-weight stabilizers into two equally weighted Pauli operators and then apply them by rotating in opposite directions, causing their overrotations to interfere destructively on the logical subspac

Structural basis for maintenance of bacterial outer membrane lipid asymmetry.

The Gram-negative bacterial outer membrane (OM) is a unique bilayer that forms an efficient permeation barrier to protect the cell from noxious compounds 1,2 . The defining characteristic of the OM is lipid asymmetry, with phospholipids comprising the inner leaflet and lipopolysaccharides comprising the outer leaflet 1-3 . This asymmetry is maintained by the Mla pathway, a six-component system that is widespread in Gram-negative bacteria and is thought to mediate retrograde transport of misplaced phospholipids from the outer leaflet of the OM to the cytoplasmic membrane 4 . The OM lipoprotein

Functional Connectivity Hubs and Networks in the Awake Marmoset Brain.

In combination with advances in analytical methods, resting-state fMRI is allowing unprecedented access to a better understanding of the network organization of the brain. Increasing evidence suggests that this architecture may incorporate highly functionally connected nodes, or "hubs", and we have recently proposed local functional connectivity density (lFCD) mapping to identify highly-connected nodes in the human brain. Here, we imaged awake nonhuman primates to test whether, like the human brain, the marmoset brain contains FC hubs. Ten adult common marmosets (Callithrix jacchus) were accli

Identifying Predictors of Central Sleep Apnea/Cheyne-Stokes Breathing in Chronic Heart Failure: a Pathophysiological Approach.

Chronic heart failure (CHF) is a major health problem associated with increased mortality, despite modern treatment options. Central sleep apnea (CSA)/Cheyne-Stokes breathing (CSB) is a common and yet largely under-diagnosed co-morbidity, adding significantly to the poor prognosis in CHF because of a number of acute and chronic effects, including intermittent hypoxia, sympathetic overactivation, disturbed sleep architecture and impaired physical tolerance. It is characterized by repetitive periods of crescendo-decrescendo ventilatory pattern, alternating with central apneas and hypopneas. The

Computational genetics analysis of grey matter density in Alzheimer's disease.

BACKGROUND: Alzheimer's disease is the most common form of progressive dementia and there is currently no known cure. The cause of onset is not fully understood but genetic factors are expected to play a significant role. We present here a bioinformatics approach to the genetic analysis of grey matter density as an endophenotype for late onset Alzheimer's disease. Our approach combines machine learning analysis of gene-gene interactions with large-scale functional genomics data for assessing biological relationships. RESULTS: We found a statistically significant synergistic interaction among

Structures of Burkholderia thailandensis nucleoside kinase: implications for the catalytic mechanism and nucleoside selectivity.

The nucleoside kinase (NK) from the mesophilic Gram-negative bacterium Burkholderia thailandensis (BthNK) is a member of the phosphofructokinase B (Pfk-B) family and catalyzes the Mg(2+)- and ATP-dependent phosphorylation of a broad range of nucleosides such as inosine (INO), adenosine (ADO) and mizoribine (MZR). BthNK is currently used in clinical practice to measure serum MZR levels. Here, crystal structures of BthNK in a ligand-free form and in complexes with INO, INO-ADP, MZR-ADP and AMP-Mg(2+)-AMP are described. The typical homodimeric architecture of Pfk-B enzymes was detected in three d

Oral biofilm architecture on natural teeth.

Periodontitis and caries are infectious diseases of the oral cavity in which oral biofilms play a causative role. Moreover, oral biofilms are widely studied as model systems for bacterial adhesion, biofilm development, and biofilm resistance to antibiotics, due to their widespread presence and accessibility. Despite descriptions of initial plaque formation on the tooth surface, studies on mature plaque and plaque structure below the gum are limited to landmark studies from the 1970s, without appreciating the breadth of microbial diversity in the plaque. We used fluorescent in situ hybridizatio

Immediate implants placed into infected sockets: a case report with 3-year follow-up.

The esthetics and functional integrity of the periodontal tissue may be compromised by dental loss. Immediate implants became a viable option to maintain the periodontal architecture because of their anatomic compatibility with the dental socket and the possibility of eliminating local contamination. This article describes the procedure of immediate implant placement in the anterior maxilla replacing teeth with chronic periapical lesions, which were condemned due to endodontic lesions persisting after failed endodontic treatment and endodontic surgery, and discusses the relationship between th

Coherent quantum state storage and transfer between two phase qubits via a resonant cavity.

As with classical information processing, a quantum information processor requires bits (qubits) that can be independently addressed and read out, long-term memory elements to store arbitrary quantum states, and the ability to transfer quantum information through a coherent communication bus accessible to a large number of qubits. Superconducting qubits made with scalable microfabrication techniques are a promising candidate for the realization of a large-scale quantum information processor. Although these systems have successfully passed tests of coherent coupling for up to four qubits, commu

Definition of a VR-based protocol to treat social phobia.

Social phobia is an anxiety disorder that is accessible to two forms of treatment yielding scientifically validated results: drugs and cognitive-behavioral therapies. Graded exposure to feared social situations is fundamental to obtain an improvement of the anxious symptoms. Traditionally, exposure therapies are done either in vivo or by imagining the situations. In vivo exposure is sometimes difficult to control and many patients have some difficulties in using imagination. Virtual reality (VR) seems to bring significant advantages. It allows exposures to numerous and varied situations. This

Designing An electronic patient record with multiple real time decision support modules for managing diseases.

Physician compliance with clinical guidelines varies between 30% and 50% despite knowledge of and acceptance of guidelines. In an attempt to improve the use of guidelines there is increasing interest in the development of clinical decision support systems. We are interested in designing a system that is intended for interdisciplinary use in primary, secondary, and tertiary level situations. Since 1995 we have been developing an electronic oral health record containing a decision support system for caries management. As the addition of different decision support modules was contemplated, the ab

A new human in vitro model of cytotypic and testosterone-producing organoids derived from testicular tissue of transgender women.

STUDY QUESTION: Can testicular tissue from trans women (trans tissue) be used to create human testicular organoids? SUMMARY ANSWER: Testosterone-producing and cytotypic human testicular organoids with bicompartmental architecture can be successfully generated from trans tissue. WHAT IS KNOWN ALREADY: Testicular organoids are a promising tool for studying testicular function and the effects of toxicants. Immature testicular cells are currently the most efficient at forming organoids that closely recapitulate seminiferous tubule-like architecture and functions. However, the scarcity of immatur

Mesoporous Thin Film Architectures: Addressing Material Demands through Molecular Self-Assembly.

Mesoporous thin films spark interest across a wide range of disciplines due to their tunable nanostructures, large internal surface areas, and strong compatibility with planar optical, electronic, and microfluidic devices. While attention in the porous materials community has shifted toward macroporous or disordered nanoporous systems, a resurgence in mesoporous thin film research is underway, driven by new molecular self-assembly methods, advanced materials chemistry, and improved characterization techniques. The integration of high-χN block copolymer design, kinetically persistent micelle te

Automated workflow for genotyping individual transposon library variants.

BACKGROUND: The fields of genome engineering and synthetic biology rely on high-throughput screening to accelerate research and increase reproducibility. This often requires the use of costly equipment such as flow cytometers, specialized microfluidic devices or liquid handlers. Despite the availability of population-level screening methods, genotyping individual clones in transposon libraries remains a necessary but frequently labor-intensive task well suited for liquid-handling platforms. Although open-source automation is democratizing access to liquid handlers, it requires a minimum level

Identifying a new candidate space crop: the alpine strawberry Fragaria vesca.

New food systems are needed for multi-year space missions. Supporting long missions exclusively with preserved food is logistically impractical and would have negative physical and psychological impacts on crew members. One option is to grow crops of fresh food on spacecraft or in off-Earth habitats. Strawberry (Fragaria sp.) has been identified as a potential space crop due to its physical, physiological and sensory properties. We assessed the potential of the alpine strawberry, F. vesca, as a new space crop by comparing F. vesca accession Hawaii 4 (F7) to NASA's space strawberry candidate F.

Building a deeply AI over a substrate you cannot retrain: the Clara Nexus reference implementation of layered intelligence theory.

Layered intelligence theory (LIT) specifies eight design principles for what its author terms a Deeply AI-a cognitive architecture whose parameters and surrounding structure jointly instantiate the preconditions for reorganisation, reflection, and falsifiable learning. The specification is silent on how a builder without write access to those parameters might proceed. This paper reports a working reference implementation, the Clara platform, instantiating those principles over hosted frontier large language models. The reference implementation is the platform as a whole rather than any single

Deep Research Agents: Major Breakthrough or Incremental Progress for Medical AI?

Deep research agents are autonomous large language model-based systems capable of iterative web search, retrieval, and synthesis. They are increasingly positioned as the next major leap in medical artificial intelligence. In this viewpoint, we argue that while these agents mark progress in information access and workflow automation, they represent an incremental evolution rather than a paradigm shift. We review current applications of deep research agents in biomedical scenarios, including literature review generation, clinical evidence synthesis, guideline comparison, and patient education. A

Root system architecture remodeling under oxygen-mediated subsurface drip irrigation: a mechanistic hypothesis framework linking multiphase mass transfer to root foraging decisions.

As climate variability and water scarcity intensify, subsurface drip irrigation (SDI) has emerged as a key water-saving technology across arid and semiarid regions. However, in practice, the wetted soil volume around each emitter remains close to saturation, disrupting the continuity of the soil gas phase and restricting oxygen diffusion into the root zone. Subsequently, the water and oxygen supplies may compete for the same pore space, which is an intrinsic constraint of SDI. Aerated SDI, particularly systems that use micro- and nanobubbles (MNBs), has shown potential to improve crop yield an

Beyond classical models: LLM-driven survival analysis for breast cancer prognosis using European cancer registry data.

BACKGROUND: Survival analysis is a fundamental tool in clinical prognosis, yet traditional statistical models often struggle to capture complex, high-dimensional relationships in modern healthcare data. Recent advances in Large Language Models (LLMs) offer new opportunities for flexible and context-aware modeling. At the same time, access to real-world clinical data remains restricted due to privacy constraints, motivating the use of synthetic datasets as a privacy-preserving alternative for model development and evaluation. METHODS: We propose a survival analysis framework based on fine-tune
