Etiology of craniofacial malformations in mouse models of blepharophimosis, ptosis and epicanthus inversus syndrome.

Blepharophimosis, ptosis, epicanthus-inversus syndrome (BPES) is an autosomal dominant genetic disorder characterized by narrow palpebral fissures and eyelid levator muscle defects. BPES is often associated to premature ovarian insufficiency (BPES type I). FOXL2, a member of the forkhead transcription factor family, is the only gene known to be mutated in BPES. Foxl2 is essential for maintenance of ovarian identity, but the developmental origin of the facial malformations of BPES remains, so far, unexplained. In this study, we provide the first detailed account of the developmental processes l

Whole exome sequencing uncovers genetic syndromes and putative candidate genes underlying orofacial clefts presenting with limb abnormalities in a Sub-Saharan African cohort.

BACKGROUND: Orofacial clefts (OFCs) are the most frequent congenital craniofacial anomalies that occur during embryonic development. The incidence is ~ 1 in 700 live births, and it may occur in isolation or with other abnormalities, such as limb deformities. Congenital limb malformations are the second most prevalent birth defect, affecting 1 per 500 to 1000 live births. It can also occur in isolation or as part of a syndrome. This study investigated the genetic etiology of OFCs co-occurring with limb abnormalities in a Sub-Saharan African cohort. METHODS: Nine unrelated probands with concurr

The Shikani Optical Stylet: a useful adjunct to airway management in a neonate with popliteal pterygium syndrome.

The craniofacial abnormalities of popliteal pterygium syndrome present unique challenges to the pediatric anesthetist. Congenital fibrous bands between the maxilla and mandible limit mouth opening and the tongue may be fused to the palate. We describe the use of the Shikani Optical Stylet, a novel fiberoptic endoscope, in the airway management of a neonate with popliteal pterygium syndrome and syngnathia.

Computerized axial tomography of craniofacial malformations. A preliminary report.

Computerized axial tomography is a new radiologic technique that permits noninvasive study of the skull, brain, and cerebral ventricles. This technique has been applied to the study of craniofacial malformations prior to intracranial surgical correction. The assessment of the cerebral ventricles is valuable as a base line study as well as helpful in detecting unsuspected abnormalities in the brain prior to surgery.

Olaparib induces craniofacial cartilage malformations in zebrafish embryos through DNA damage, oxidative stress, and Wnt dysregulation.

Pharmaceutical residues in aquatic environments are a growing concern. Global surveillance has detected these residues in nearly a quarter of the world's rivers, with anticancer drugs drawing particular attention due to their high bioactivity. Olaparib (Ola), a poly(ADP-ribose) polymerase (PARP) inhibitor, is widely used to treat breast and ovarian cancers, but its developmental toxicity to aquatic vertebrates remains unclear. We exposed zebrafish embryos to Ola (5-15 mg/L) from 6 to 120 h post-fertilization (hpf). Ola exposure caused dose-dependent craniofacial cartilage malformations and beh

Complex craniofacial changes in blind cave-dwelling fish are mediated by genetically symmetric and asymmetric loci.

The genetic regulators of regressive craniofacial morphologies are poorly understood. To shed light on this problem, we examined the freshwater fish Astyanax mexicanus, a species with surface-dwelling and multiple independent eyeless cave-dwelling forms. Changes affecting the skull in cavefish include morphological alterations to the intramembranous circumorbital bones encircling the eye. Many of these modifications, however, have evolved separately from eye loss, such as fragmentation of the third suborbital bone. To understand the genetic architecture of these eye-independent craniofacial al

Phenotypic Spectrum and Chromosomal Discordance in Alobar Holoprosencephaly: A Comparative Case Series from a Tertiary Referral Center.

INTRODUCTION: Holoprosencephaly (HPE) is a rare congenital malformation of the forebrain caused by incomplete midline cleavage, often accompanied by craniofacial abnormalities. The condition arises from multifactorial etiologies, including genetic, environmental, and maternal factors. Early prenatal diagnosis is essential for parental counseling, management decisions, and detection of associated anomalies. CASE REPORT: We report two cases of alobar HPE diagnosed in the third trimester by ultrasonography. The first case involved a 41-year-old primigravida at 32 weeks of gestation, with ultraso…

17q12 deletion syndrome mouse model shows defects in craniofacial, brain and kidney development, and glucose homeostasis.

17q12 deletion (17q12Del) syndrome is a copy number variant (CNV) disorder associated with neurodevelopmental disorders and renal cysts and diabetes syndrome (RCAD). Using CRISPR/Cas9 genome editing, we generated a mouse model of 17q12Del syndrome on both inbred (C57BL/6N) and outbred (CD-1) genetic backgrounds. On C57BL/6N, the 17q12Del mice had severe head development defects, potentially mediated by haploinsufficiency of Lhx1, a gene within the interval that controls head development. Phenotypes included brain malformations, particularly disruption of the telencephalon and craniofacial defe

Sequential developmental changes in holoprosencephalic mouse embryos exposed to ethanol during the gastrulation period.

BACKGROUND: Prenatal exposure to ethanol induces holoprosencephalic malformations in both humans and laboratory animals. However, its teratogenic window for inducing holoprosencephaly is narrow, and the teratogenic mechanism is not well understood. In the present study, we examined the morphological changes in the craniofacial structures of mouse embryos/fetuses at intervals following ethanol treatment and evaluated gene expression patterns in the embryos. METHODS: Pregnant C57BL/6J mice were given two doses of ethanol (30 mg/kg in total) on the morning (7:00 and 11:00 AM) of day 7. The fetus
