Intermediate filament proteins are reliable immunohistological biomarkers to help diagnose multiple tissue-specific diseases.

Cytoskeletal networks are proteins that effectively maintain cell integrity and provide mechanical support to cells by actively transmitting mechanical signals. Intermediate filaments, which are from the cytoskeleton family and are 10 nanometres in diameter, are unlike actin and microtubules, which are highly dynamic cytoskeletal elements. Intermediate filaments are flexible at low strain, harden at high strain and resist breaking. For this reason, these filaments fulfil structural functions by providing mechanical support to the cells through their different strain-hardening properties. Inter…

Ribbon Synapses and Retinal Disease: Review.

Synaptic ribbons are presynaptic protein complexes that are believed to be important for the transmission of sensory information in the visual system. Ribbons are selectively associated with those synapses where graded changes in membrane potential drive continuous neurotransmitter release. Defective synaptic transmission can arise as a result of the mutagenesis of a single ribbon component. Visual diseases that stem from malfunctions in the presynaptic molecular machinery of ribbon synapses in the retina are rare. In this review, we provide an overview of synaptopathies that give rise to reti

Parasite microtubule arrays.

Microtubules are a key component of eukaryotic cell architecture. Regulation of the dynamic growth and shrinkage of microtubules gives cells their shape, allows cells to swim, and drives the separation of chromosomes. Parasites have developed intriguingly divergent biology, seemingly expanding upon and reinventing microtubule use in fascinating ways. These organisms affect life on the planet at scales that are often overlooked: there are likely more parasitic than free-living organisms on Earth, and they have a sizeable influence across ecosystems. As parasites can cause devastating diseases,

Cytoskeletal orchestration of glucose uptake in Sertoli cell to support efferocytosis of apoptotic germ cells.

Efferocytosis of non-viable germ cells by Sertoli cells (SCs) constitutes a sentinel for testis homeostasis, yet how SCs signal for the meta … deficiency impairs glucose uptake and lactate production in SCs. Moreover, a defective assembly of cytoskeleton and a loss of blood-testis barrier integrity are also featured by SCs deficient in βII-spectrin. The disruption in glucose metabolism and cytoskeletal organization synergistically lead to a defective efferocytosis. In vivo siRNA-mediated targeting o …

Microtubules self-repair in living cells.

Microtubule self-repair has been studied both in vitro and in vivo as an underlying mechanism of microtubule stability. The turnover of tubulin dimers along the microtubule has challenged the pre-existing dogma that only growing ends are dynamic. However, although there is clear evidence of tubulin incorporation into the shaft of polymerized microtubules in vitro, the possibility of such events occurring in living cells remains uncertain. In this study, we investigated this possibility by microinjecting purified tubulin dimers labeled with a red fluorophore into the cytoplasm of cells expressi

A transient apical extracellular matrix relays cytoskeletal patterns to shape permanent acellular ridges on the surface of adult C. elegans.

Epithelial cells secrete apical extracellular matrices to form protruding structures such as denticles, ridges, scales, or teeth. The mechanisms that shape these structures remain poorly understood. Here, we show how the actin cytoskeleton and a provisional matrix work together to sculpt acellular longitudinal alae ridges in the cuticle of adult C. elegans. Transient assembly of longitudinal actomyosin filaments in the underlying lateral epidermis accompanies deposition of the provisional matrix at the earliest stages of alae formation. Actin is required to pattern the provisional matrix into …

Multivalent Interactions Drive the Toxoplasma AC9:AC10:ERK7 Complex To Concentrate ERK7 in the Apical Cap.

The Toxoplasma inner membrane complex (IMC) is a specialized organelle that is crucial for the parasite to establish an intracellular lifestyle and ultimately cause disease. The IMC is composed of both membrane and cytoskeletal components, further delineated into the apical cap, body, and basal subcompartments. The apical cap cytoskeleton was recently demonstrated to govern the stability of the apical complex, which controls parasite motility, invasion, and egress. While this role was determined by individually assessing the apical cap proteins AC9, AC10, and the mitogen-activated protein kina…

Conformation Control of Amyloid Filaments by Repeated Thermal Perturbation.

We report that repeated thermal perturbation by thermal cycling (TC) accelerates the formation rate of amyloid filaments at microliter volumes (10-200 μL) and produces a new conformation of zigzag-shaped filaments. The amyloid filaments have been synthesized under different TC conditions, such as temperature variations (ΔT = 0-86 °C) and the number of cycles (C# = 30-90). In particular, the filament formation was promoted by TC with ΔT ≥ 30 °C. This indicates that the change in binding energy of β-sheets and the breakage of disulfide bonds induced by TC with large ΔT contributed to the increas

A plague of actin disassembly.

Pathogenic Yersinia species employ several strategies to evade the host immune system, including interfering with cytoskeletal remodeling as a way to block macrophage phagocytosis. The kinase YopO binds directly to monomeric actin and phosphorylates the actin-remodeling protein gelsolin, but the functional importance of this gelsolin modification has not been clear. A combined biochemical, computational, and biophysical study now reveals that YopO-mediated phosphorylation activates host gelsolin, leading to severed actin filaments and disturbed actin dynamics.

Covariant stringlike dynamics of scroll wave filaments in anisotropic cardiac tissue.

It has been hypothesized that stationary scroll wave filaments in cardiac tissue describe a geodesic in a curved space whose metric is the inverse diffusion tensor. Several numerical studies support this hypothesis, but no analytical proof has been provided yet for general anisotropy. In this Letter, we derive dynamic equations for the filament in the case of general anisotropy. These equations are covariant under general spatial coordinate transformations and describe the motion of a stringlike object in a curved space whose metric tensor is the inverse diffusion tensor. Therefore the behavio

Golgi-associated filament networks in duct epithelial cells of rabbit submandibular glands: immunohistochemical light and electron microscopic studies.

The duct epithelial cells of rabbit submandibular glands expressed keratin 8, keratin 14, and actin in the supranuclear region, and these cytoskeletal proteins formed ring structures, approximately 1-2.5 microm in diameter. Ultrastructurally, these ring structures were observed as a 'Golgi-associated filament network' surrounding Golgi apparatuses. Double immunofluorescent studies showed that keratin 8 and keratin 14 formed keratin 8/14 filaments, and that these filaments and actin filaments colocalized as components of the Golgi-associated filament network. Our studies suggested that the Golg

Roles of microfilaments in exocytosis: a new hypothesis.

We observed the dynamic changes in the localization of microfilaments during the exocytic secretion of rat parotid and submandibular gland acinar cells, and obtained results which led us to propose a new concept of microfilament function in exocytosis. With the electron microscopy, NBD-Phallacidin (NBD-PL) fluorescence technique and immunohistochemistry for myosin, microfilaments consisting of F-actin and myosin were localized mainly underneath the luminal plasma membrane. Microfilaments were not detectable around the secretory granules which were stored in the cytoplasm, but were clearly obse

CNP-induced cGMP signaling reduces growth cone stiffness and Ca2+ levels in embryonic DRG neurons.

A cyclic guanosine monophosphate (cGMP) signaling pathway composed of the extracellular ligand C-type natriuretic peptide (CNP), the transmembrane natriuretic peptide receptor 2 (Npr2), and the cGMP-dependent protein kinase I (cGKI) regulates axon bifurcation of embryonic dorsal root ganglion (DRG) neurons in mice. Despite the importance of this process for the development of neuronal connectivity, the underlying mechanisms are only partially understood. Axon bifurcation requires an orchestrated rearrangement of the cytoskeleton in growth cones, the highly motile structures at axon tips. In th…
