Historical reconstruction studies are important for understanding the evolutionary mechanisms associated with different parasite-host systems. Platyhelminths of the classes Monopisthocotyla and Polyopisthocotyla (formerly Monogenoidea or Monogenea) have proven to be excellent models for historical reconstruction studies due to their exceptional parasite specificity, suggesting that cospeciation events are the main pattern observed in these parasite-host systems (i.e., codivergence). However, recent studies indicate that not only cospeciation but also duplication, host-switch, and loss (extinct

New morphological and molecular data generated for hemiuroid trematodes from fishes of coastal waters of Cat Ba Island, Northern Vietnam. The trematode Stomachicola muraenesocis was detected in the intestines of eels Pisodonophis boro. Vietnamese specimens of S. muraenesocis were identical to trematodes from Iran by morphology and molecular data and have some differences with specimens from South Korea by 28S rRNA gene sequence data. Results of p-distance estimation and phylogenetic analysis of Hemiuroidea specimens of Stomachicola differ from other representatives of the superfamily at the le

Toxocara vitulorum is an intestinal nematode of cattle, water buffaloes, and other bovids. This parasite can cause high morbidity and mortality in calves and economic losses. Although T. vitulorum has been reported in cattle in Mexico, there are no records in water buffaloes. This study details the morphological and molecular identification of T. vitulorum obtained from three buffalo calves in Tabasco, Mexico. Nematodes collected from buffalo calves were studied using morphological techniques and partial sequences of the 28S ribosomal RNA (28S) gene and the cytochrome c oxidase 1 mitochondrial

Hassalstrongylus Durette-Desset, 1971 (Nematoda: Heligmonellidae), includes 19 species that are distributed from the southwestern United States to central-western Argentina. Hassalstrongylus aduncus is a parasitic nematode of rodents from the subfamilies Arvicolinae, Murinae, and Sigmodontinae, and has been recorded from southern Virginia and Oklahoma to Costa Rica. This species was described by Chandler in 1932; the morphology of the synlophe was not included. Subsequently, in 1972, Durette-Desset described only the synlophe of the middle region of the body in both sexes. Despite its wide geo

Lungworm infection, or verminous pneumonia, is a parasitic disease that causes serious problems in small and large ruminants. Despite the fact that nematodes of the genus Dictyocaulus in cattle and sheep are the main cause of this disease, there are few studies on the natural infections of South American camelids. For this reason, this study aims to report the natural infection by Dictyocaulus filaria in vicunas (Vicugna vicugna) for the first time. During a shearing season (chaku) in Cuzco, Peru, two accidentally killed adult vicunas were submitted to the IVITA-Marangani research center in Cu

BACKGROUND: Schistosomiasis remains a public health issue and the need for accurate and affordable diagnostics is crucial in the elimination of the disease. While molecular diagnostics are highly effective, they are expensive, with the main costs been associated with DNA extraction. The DNA dipstick is a rapid, affordable and simple purification method that allows DNA to be extracted from diagnostic samples within 30 s. We aimed to optimise the DNA dipstick method for samples from mice and egg-spiked human samples.
METHODS: Urine, blood and faeces were collected from mice exposed to Schistoso

Dirofilaria repens is a parasitic nematode causing vector-borne disease (dirofilariasis), considered an emerging problem in veterinary and human medicine. Although main hosts are carnivores, particularly dogs, D. repens shows high zoonotic potential. The disease spreads uncontrollably, affecting new areas. Since there is no vaccine against dirofilariasis, the only way to limit disease transmission is an early diagnosis. Currently, diagnosis depends on the detection of microfilariae in the host bloodstream using modified Knott's test or multiplex PCR. However, the efficacy of tests relying on m

BACKGROUND: Multiplex polymerase chain reaction (PCR) and PCR-restriction fragment length polymorphism (RFLP) for nuclear phosphoenolpyruvate carboxykinase (pepck) and polymerase delta (pold), respectively, have been used to differentiate Fasciola hepatica, F. gigantica, and hybrid Fasciola flukes. However, discrimination errors have been reported in both methods. This study aimed to develop a multiplex PCR based on a novel nuclear marker, the fatty acid binding protein type I (FABP) type I gene.
METHODS: Nucleotide sequence variations of FABP type I were analyzed using DNA samples of F. hepa

Trichobilharzia species are parasitic flatworms (called schistosomes or flukes) that cause important diseases in birds and humans, but very little is known about their molecular biology. Here, using a transcriptomics-bioinformatics-based approach, we explored molecular aspects pertaining to the nutritional requirements of Trichobilharzia szidati ('visceral fluke') and T. regenti ('neurotropic fluke') in their avian host. We studied the larvae of each species before they enter (cercariae) and as they migrate (schistosomules) through distinct tissues in their avian (duck) host. Cercariae of both

BACKGROUND: Infections with helminths cause an enormous disease burden in billions of animals and plants worldwide. Large scale use of anthelmintics has driven the evolution of resistance in a number of species that infect livestock and companion animals, and there are growing concerns regarding the reduced efficacy in some human-infective helminths. Understanding the mechanisms by which resistance evolves is the focus of increasing interest; robust genetic analysis of helminths is challenging, and although many candidate genes have been proposed, the genetic basis of resistance remains poorly

In the present paper, species of the Proteocephalus-aggregate de Chambrier, Zehnder, Vaucher, and Mariaux, 2004 (Cestoda: Proteocephalidae) reported from centrarchid and percid fishes in North America are reviewed, and their taxonomic status is critically assessed based on a study of type specimens and new material from Canada and the United States. The following 3 species, supposedly strictly specific to their fish definitive hosts, are recognized as valid: (1) Proteocephalus fluviatilis Bangham, 1925 (new synonyms Proteocephalus osburni Bangham, 1925 and Proteocephalus microcephalus Haderlie

Clinostomum spp. (Digenea: Clinostomidae) are a group of trematodes commonly found in the buccal cavity and oesophagus of a variety of piscivorous birds. The metacercariae, colloquially known as "yellow grubs," have been reported from a diverse group of freshwater fishes worldwide. In the catfish farming region of the southeastern USA, piscivorous birds present a continuous challenge for aquaculturists in the form of fish depredation and the introduction of trematodes into these static, earthen pond systems. Clinostomum spp. are commonly encountered in farm-raised catfish. While generally cons

The use of copro-DNA detection methods for the diagnosis of canine Echinococcus multilocularis infection was evaluated with a focus on DNA extraction techniques: two commercial kits and a modified alkaline-sodium dodecyl sulfate (SDS) technique. Dog feces (0.2 g) mixed with a protoscolex or with 1 or 10 eggs of E. multilocularis were subjected to DNA detection following extraction by these methods. DNA was extracted from all protoscolex samples by all methods, but success for samples with eggs depended on extraction technique with the modified technique showing success on all samples. Followin

Myxobolus tsangwuensis Chen, 1954 is a common parasite infecting the gills of common carp Cyprinus carpio (L.). It was described simply in the original description and its molecular data was absent, which makes the accurate diagnosis challenging. Here we supplemented its description based on the morphological, histological and molecular data. It was characterized by the presence of small round or ellipsoidal plasmodia in the gills of host and histology showed the plasmodia developed in the capillary network of the gill lamella. Mature spores of M. tsangwuensis were ellipsodal in frontal view a

The Schistosoma mansoni transcriptome revealed new members of the dynein light chain family (DLC/LC8). The antigenicity and immunogenicity of these proteins, and their potential as vaccine candidates were investigated. Two DLC genes (DLC12_JI392413.1 and DLC13_JI387686.1) were cloned and the recombinant proteins produced in E. coli. The immunization of mice with the rDLCs, using alhydrogel as adjuvant, resulted in high titers of antibodies, indicated that these proteins are highly immunogenic. The anti-DLCs antibodies presented cross reactivity with both recombinant antigens and also recognize

Trichinella spiralis is a tissue-dwelling nematode parasite. A loop-mediated isothermal amplification (LAMP) assay was developed and validated for the sensitive and rapid detection of T. spiralis larvae in muscle samples. Sixteen sets of primers were designed to recognise distinct sequences of a conserved gene, a 1.6kb repetitive element of the Trichinella genome. One set of primers was selected as the most appropriate for rapid detection. The specificity and sensitivity of the primers in LAMP reactions for T. spiralis larvae and muscle samples of mice infected with T. spiralis were determined

Echinococcus multilocularis, causing alveolar echinococcosis in humans, is a highly pathogenic emerging zoonotic disease in central Europe. The gold standard for the identification of this parasite in the main host, the red fox, namely identification of the adult parasite in the intestine at necropsy, is very laborious. Copro-enzyme-linked immunosorbent assay (ELISA) with confirmatory polymerase chain reaction (PCR) has been suggested as an acceptable alternative, but no commercial copro-ELISA tests are currently available and an in-house test is therefore required. Primers targeting the gene encoding the small subunit rRNA were designed to amplify DNA from Schistosoma mansoni with high specificity. Three PCR systems were developed: conventional PCR, two-step nested PCR (NPCR) and single-tube nested PCR (STNPCR). The limits of detection of parasite DNA for the conventional PCR, NPCR and STNPCR were 10 pg, 0.1 fg and 1 fg, respectively. The assays were highly specific for S. mansoni and did not recognise DNA from closely related non-schistosome trematodes. Using pools of Biomphalaria molluscs, PCR, NPCR and STNPCR were positive in 6/16 (37.5%), 15/16 (93.8
