Cessation of fluoxetine treatment increases alcohol seeking during relapse and dysregulates endocannabinoid and glutamatergic signaling in the central amygdala.

Administration of selective serotonin reuptake inhibitors (SSRIs), typically used as antidepressants, induces long-lasting behavioral changes associated with alcohol use disorder (AUD). However, the contribution of SSRI (fluoxetine)-induced alterations in neurobiological processes underlying alcohol relapse such as endocannabinoid and glutamate signaling in the central amygdala (CeA) remains largely unknown. We utilized an integrative approach to study the effects of repeated fluoxetine administration during abstinence on ethanol drinking. Gene expression and biochemical and electrophysiologic

Temporally specific miRNA expression patterns in the dorsal and ventral striatum of addiction-prone rats.

MicroRNAs (miRNAs) within the ventral and dorsal striatum have been shown to regulate addiction-relevant behaviours. However, it is unclear how cocaine experience alone can alter the expression of addiction-relevant miRNAs within striatal subregions. Further, it is not known whether differential expression of miRNAs in the striatum contributes to individual differences in addiction vulnerability. We first examined the effect of cocaine self-administration on the expression of miR-101b, miR-137, miR-212 and miR-132 in nucleus accumbens core and nucleus accumbens shell (NAcSh), as well as dorsom

Drug versus sweet reward: greater attraction to and preference for sweet versus drug cues.

Despite the unique ability of addictive drugs to directly activate brain reward circuits, recent evidence suggests that drugs induce reinforcing and incentive effects that are comparable to, or even lower than some nondrug rewards. In particular, when rats have a choice between pressing a lever associated with intravenous cocaine or heroin delivery and another lever associated with sweet water delivery, most respond on the latter. This outcome suggests that sweet water is more reinforcing and attractive than either drug. However, this outcome may also be due to the differential ability of swee

Epigenetic drugs and psychedelics as emerging therapies for alcohol use disorder: insights from preclinical studies.

Alcohol use disorder (AUD) is a public health issue that affects millions of people worldwide leading to physical, mental and socio-economic consequences. While current treatments for AUD have provided relief to individuals, their effectiveness on the long term is often limited, leaving a number of affected individuals without sustainable solutions. In this review, we aim to explore two emerging approaches for AUD: psychedelics and epigenetic drugs (i.e., epidrugs). By examining preclinical studies, different animal species and procedures, we delve into the potential benefits of each of these

Self-Deception in Addiction Rehabilitation: Impulsivity and Self-Efficacy as Predictors of Manipulation and Mystification.

Substance use disorder (SUD) is a global problem with serious psychological, physiological, and social consequences. Seeking professional help is often delayed due to a lack of self-recognition of addiction, frequently sustained by self-deception. Although self-deception is a core feature of SUD, the mechanisms underlying it remain insufficiently studied. This research examines the role of impulsivity and self-efficacy in predicting self-deceptive behaviors-manipulation and mystification-in individuals undergoing addiction rehabilitation. The sample consisted of 122 Spanish participants from t

Olanzapine Attenuates the Morphine-Induced Conditioned Place Preference: The Involvement of the D2-Like Dopamine Receptors.

OBJECTIVE: Opioid use disorder is associated with persistent molecular and cellular changes in the brain, leading to compulsive drug-seeking behaviors. This study aimed to evaluate the effects of olanzapine (OLZ), a D2-like dopamine receptor (D2R) antagonist, on morphine dependence using the conditioned place preference (CPP) model. METHODS: Morphine-induced CPP was established by subcutaneous (sc) administration of morphine (5 mg/kg) for three consecutive days. OLZ at doses of 1.5, 3, and 4.5 mg/kg was administered intraperitoneally (ip) during different phases of CPP: acquisition, expressio…

Novel opioid cyclic tetrapeptides: Trp isomers of CJ-15,208 exhibit distinct opioid receptor agonism and short-acting κ opioid receptor antagonism.

BACKGROUND AND PURPOSE: The κ opioid receptor antagonists demonstrate potential for maintaining abstinence from psychostimulant abuse, but existing non-peptide κ-receptor selective antagonists show exceptionally long activity. We hypothesized that the L- and D-Trp isomers of CJ-15,208, a natural cyclic tetrapeptide reported to be a κ-receptor antagonist in vitro, would demonstrate short-acting, dose-dependent antagonism in vivo, preventing reinstatement of cocaine-seeking behaviour. EXPERIMENTAL APPROACH: Affinity, selectivity and efficacy of the L-Trp and D-Trp isomers for opioid receptors w

Sex and the unspoken in male street prostitution.

Although the overwhelming majority of male prostitutes work through agencies or by placing their own ads, most studies of male prostitution focus upon young men who work on the street. Remarkably, these studies seldom identify the dynamics of poverty and street-level violence as important elements of their examination. Investigations of male sex work-few though they are-focus almost exclusively upon sexual aspects of "the life." Despite the importance of these networks in shaping the contours of street life, and often in enabling one's very survival, the primary research focus has remained on

A positive allosteric modulator of α7 nicotinic receptor reduces levodopa-induced dyskinesias in hemi-parkinsonian mice.

Parkinson's disease is a prevalent neurodegenerative disorder characterized by motor impairments including rigidity, bradykinesia and tremor. L-3,4-dihydroxyphenylalanine (L-DOPA) keeps being the standard treatment for Parkinson's disease. But long-term treatment often leads to L-DOPA induced dyskinesias (LIDs): abnormal involuntary movements (AIMs) that significantly impact patients' quality of life. Drugs acting on nicotinic acetylcholine receptors (nAChRs) have emerged as a potential treatment for managing LIDs, since nicotine, shows promise in alleviating LIDs in animal models. Positive al

Given that drug re-exposure may potentiate relapse to drug-seeking behavior, the ability of R,S-GVG to block METH-triggered reinstatement offers further support for its use in the successful management of METH dependence.

The effect of dextromethorphan on reward-related behaviors: A systematic review of preclinical and clinical evidence.

INTRODUCTION: Extant literature suggests that anhedonia, defined as a loss of the ability to feel pleasure or interest, is subserved by dysregulation of reward processing in the central nervous system. Dextromethorphan (DXM), an uncompetitive N-Methyl-d-Aspartate (NMDA) receptor antagonist and sigma-1 (σ1) receptor agonist, is a glutamatergic modulator with antidepressant properties. The effect of DXM on reward-related outcomes remains inadequately characterized. Herein, we conducted a systematic review of extant literature reporting on the effects of DXM on reward-related behaviors in both pr

Serotonin transporter gene polymorphism and the phenotypic heterogeneity of adult ADHD.

The present study investigates possible associations between the 5-HTT control region polymorphism (5-HTTLPR) with adult ADHD, including subtypes, severity, temperament profile and comorbidities. The polymorphic site was genotyped in 312 adult patients with ADHD and 236 controls, all of them Brazilians of European descent. The interviews followed the DSM-IV criteria, using the K-SADS-E for ADHD and oppositional defiant disorder, SCID-I and MINI for comorbidities and the TCI for temperament dimensions. The 5-HTTLPR polymorphism was not associated with ADHD. Carriers of the S allele presented sl

Obsessive-compulsive aspects as predictors of poor response to treatments in patients with chronic migraine and medication overuse.

Patients with chronic migraine (CM) and medication overuse (MO) have a high frequency of psychiatric comorbidity or psychopathological traits, the presence of which may have important implications for the course of the CM and the MO, both for response to treatment and possible relapses. Overuse of symptomatic drugs is regarded as one of the most important risk factor for the transformation of episodic migraine into CM and drug-seeking tendency due to fear of headache in chronic migraine patients shares with obsessive-compulsive disorder (OCD) the compulsive quality of the behavior. Aim of this

A single-center, retrospective analysis evaluating the utilization of the opioid risk tool in opioid-treated cancer patients.

The Opioid Risk Tool (ORT) is a screening tool used to assess risk of opioid misuse by stratifying aberrant drug-seeking behaviors and/or identifying known risk factors for drug abuse. The objectives of this study were to risk stratify opioid misuse in a cancer pain population and determine the most common patient risk factors associated with misuse utilizing the ORT. This was a retrospective analysis conducted at an academic comprehensive cancer center. Patients were referred by an oncologist or hematologist to an outpatient palliative care clinic. One-hundred and fourteen patients with cance…

Cocaine-induced sensitization and glutamate plasticity in the nucleus accumbens core: effects of sex.

BACKGROUND: The development and persistence of addiction is mediated in part by drug-induced alterations in nucleus accumbens (NAc) function. AMPA-type glutamate receptors (AMPARs) provide the main source of excitatory drive to the NAc and enhancements in transmission of calcium-permeable AMPARs (CP-AMPARs) mediate increased cue-triggered drug-seeking following prolonged withdrawal. Cocaine treatment regimens that result in psychomotor sensitization enhance subsequent drug-seeking and drug-taking behaviors. Furthermore, cocaine-induced locomotor sensitization followed by 14 days of withdrawal

Evidence for Modulation of Substance Use Disorders by the Gut Microbiome: Hidden in Plain Sight.

The gut microbiome modulates neurochemical function and behavior and has been implicated in numerous central nervous system (CNS) diseases, including developmental, neurodegenerative, and psychiatric disorders. Substance use disorders (SUDs) remain a serious threat to the public well-being, yet gut microbiome involvement in drug abuse has received very little attention. Studies of the mechanisms underlying SUDs have naturally focused on CNS reward circuits. However, a significant body of research has accumulated over the past decade that has unwittingly provided strong support for gut microbio
