Anxiolytic and sedative activities of aqueous leaf extract of Dichrocephala integrifolia (Asteraceae) in mice.

ETHNOPHARMACOLOGICAL RELEVANCE: Dichrocephala integrifolia (Asteraceae) is commonly used in the folk medicine in Cameroon to treat various ailments including headaches, jaundice, filariasis, neuritis and children's night agitation. It is also known to have anxiolytic properties. Since there were no scientific data supporting the above claims, we therefore aimed to evaluate the anxiolytic and sedative effects of the aqueous leaf extract of D. integrifolia. MATERIALS AND METHODS: The extract used was obtained after maceration of leaves of D. integrifolia in water at room temperature. The anxiol

Behavioral effects of the beta3 adrenoceptor agonist SR58611A: is it the putative prototype of a new class of antidepressant/anxiolytic drugs?

A large body of evidence corroborates the notion that deficiencies of serotonergic system are likely involved in the pathogenesis of both de … doses of SR58611A was followed by increased exploratory behavior in Swiss male mice exposed to the elevated plus maze test. These effects are mediated by beta(3) adrenoceptors since i.p. pretreatment with the selective beta(3) adrenoceptor antagonist SR59230A (5 mg/kg) blocked the effects of SR58611A. Finally, also the 5HT antagonist methysergide (2 mg/kg) prevented the …

Enhanced innate fear and altered stress axis regulation in VGluT3 knockout mice.

Glutamatergic neurons, characterized by vesicular glutamate transporters (VGluT1-3) provide the main excitation in the brain. Their disturbances have been linked to various brain disorders, which could be also modeled by the contextual fear test in rodents. We aimed to characterize the participation of VGluT3 in the development of contextual fear through its contribution to hypothalamic-pituitary-adrenocortical axis (HPA) regulation using knockout (KO) mice. Contextual fear conditioning was induced by foot shock and mice were examined 1 and 7 d later in the same environment comparing wild type

Psychological Stress Overactivates IL-23/Th17 Inflammatory Axis and Increases cDC2 in Imiquimod-Induced Psoriasis Models of C57BL/6 Mice.

Psychological stress often accompanies psoriasis, and both conditions involve an overactive IL-23/Th17 inflammatory axis. However, the mechanism behind the comorbidity of psoriasis and psychological stress remains unclear. This study established a mouse model of comorbid psoriasis and psychological stress to investigate the impact on skin inflammation and the IL-23/Th17 axis. Chronic restraint stress and imiquimod cream were used to induce psychological stress and psoriasis. Behavioural tests included the open-field test and elevated plus maze, while psoriasis severity was assessed using the P

Therapeutic Potential of Black Seed and Opium Poppy Oils in Mitigating Morphine Withdrawal Syndrome in an Animal Model.

BACKGROUND: Dependence and withdrawal syndrome caused by opioids are the most important and unambiguous factors in the clinical use of narcotic painkillers. Some evidence indicates the beneficial effects of herbal fragments in attenuating this complication. In the current study, the impact of black seed oil (BSO) and opium poppy oil (OPO) was investigated on the symptoms of morphine withdrawal syndrome in mice. METHODS: For three days, morphine was administrated to induce dependence in mice. To induce the withdrawal syndrome, on the 4th day, naloxone was injected. Thirty minutes before naloxo

PM2.5 exposure contributes to anxiety and depression-like behaviors via phenyl-containing compounds interfering with dopamine receptor.

As a global problem, fine particulate matter (PM2.5) really needs local fixes. Considering the increasing epidemiological relevance to anxiety and depression but inconsistent toxicological results, the most important question is to clarify whether and how PM2.5 causally contributes to these mental disorders and which components are the most dangerous for crucial mitigation in a particular place. In the present study, we chronically subjected male mice to a real-world PM2.5 exposure system throughout the winter heating period in a coal combustion area and revealed that PM2.5 caused anxiety and

Liver tryptophan 2,3-dioxygenase: a determinant of anxiety-like behaviour - studies with chronic nicotine administration in rats.

Deletion of the tryptophan 2,3-dioxygenase ( TDO2 ) gene induces an anxiolytic-like behaviour in mice and TDO inhibition by allopurinol elicits an antidepressant-like effect in rats exposed to restraint stress. Chronic nicotine administration inhibits TDO activity, enhances brain serotonin synthesis and exerts anxiolytic- and antidepressant-like effects in rodent models. There is a strong association between anxiety, depression and tobacco use, which is stronger in women than in men. The present study aimed to examine the relationship between behavioural measures of anxiety and depression with

Topiramate effects on ethanol and sucrose binge-like intake, anxiety- and compulsive-like behavior in non-dependent male and female C57BL/6J mice.

RATIONALE: Repetitive binge episodes are early-stage patterns in the addiction cycle associated with elevated anxiety and compulsive traits. Despite their high comorbidity and the growing off-label use of topiramate for binge-related disorders, its effects in non-dependent individuals remain poorly characterized and studies systematically comparing both sexes under identical experimental conditions remain scarce. OBJECTIVES: To evaluate the effects of topiramate on ethanol (EtOH) and sucrose binge-like intake, anxiety- and compulsive-like responses in non-dependent male and female C57BL/6J mi

Co-segregation of hyperactivity, active coping styles, and cognitive dysfunction in mice selectively bred for low levels of anxiety.

We established mouse models of extremes in trait anxiety, which are based on selective breeding for low vs. normal vs. high open-arm exploration on the elevated plus-maze. Genetically selected low anxiety-related behavior (LAB) coincided with hyperactivity in the home cage. Given the fact that several psychiatric disorders such as schizophrenia, mania, and attention deficit hyperactivity disorder (ADHD) share hyperactivity symptom, we systematically examined LAB mice with respect to unique and overlapping endophenotypes of the three diseases. To this end Venn diagrams were used as an instrumen

Dexmedetomidine elicits a prolonged anxiolytic effect by inhibiting adrenergic neurons in the locus coeruleus in mice.

Anxiety disorders impose a substantial global burden, yet current pharmacotherapies often lack sustained efficacy and carry risks of depende … lytic effects lasting ≥3 days, evidenced by improved performance in open-field, light-dark box, and elevated plus maze tests. In vivo fiber photometry and chemogenetic approaches demonstrated that Dex suppresses LC-NE neuronal hyperactivity and normalizes mPFC NE levels. Crucially, neuron-specific knockdown of α2 receptors in the LC abolished Dex's anxiolytic effects, co …

Brain Monoamine Dysfunction in Response to Predator Scent Stress Accompanies Stress-Susceptibility in Female Rats.

Post-traumatic stress disorder (PTSD) is prevalent in women; however, preclinical research on PTSD has predominantly been conducted in male animals. Using a predator scent stress (PSS) rodent model of PTSD, we sought to determine if stress-susceptible female rats show altered monoamine concentrations in brain regions associated with PTSD: the medial prefrontal cortex (mPFC), nucleus accumbens (NAc), and dorsal (dHIPP) and ventral (vHIPP) hippocampus. Female Sprague-Dawley rats were exposed to a single, 10-min PSS exposure and tested for persistent anhedonia, fear, and anxiety-like behavior ove

A mild stressor induces short-term anxiety and long-term phenotypic changes in trauma-related behavior in female rats.

INTRODUCTION: Anxiety and anxiety-influenced disorders are sexually dimorphic with women being disproportionately affected compared to men. Given the increased prevalence in women and the documented differences in anxiety and trauma behavior between male and female rats this paper sought to examine the link between stress, anxiety, and fear learning and extinction in female rats. We tested the hypothesis that a mild stressor will induce short-and long-term increases in anxiety and produce long term effects on subsequent fear learning and extinction behavior. METHODS: We induced anxiety in fem

Microglia depletion alleviates the disruptions in circadian rhythms and anxiety caused by bmal1 deficiency.

Microglia, the intrinsic immune cells of the brain, play a crucial role in maintaining neuronal health and circadian rhythms. BMAL1 is a core transcriptional regulator of the molecular circadian clock that coordinates daily rhythms in physiology and behavior. In this study, we investigated the impact of microglial modulation on circadian disruptions and behavioral abnormalities observed in bmal-/- mice. Our results revealed disruptions in circadian rhythms and microglial activation in bmal-/- mice, characterized by altered activity patterns and increased microglia numbers. Inhibiting microglia
