Diencephalic seizures: responsiveness to bromocriptine and morphine.

Two patients with posttraumatic diencephalic seizures, characterized by autonomic dysfunction and extensor posturing, had partial responses to bromocriptine and complete responses to morphine. Probable synergism between the two agents was noted. These 2 cases suggest the potential effectiveness of this regimen for the treatment of diencephalic seizures, raise questions regarding the role of the dopaminergic and opioid systems in this disease entity, and support the hypothesis that diencephalic seizures represent a release phenomenon in the brain.

Noradrenergic and endogenous opioid pathways in the regulation of luteinizing hormone secretion in the male rat.

Endogenous opioid pathways (EOPs) appear to play a role in the tonic regulation of pulsatile LH secretion. The mechanism by which EOPs influence LH release is not known. However, the observation that acute chemical disruption of the noradrenergic (NE) system by alpha-receptor-blocking agents or NE synthesis inhibitors blocks the LH increase occurring in response to naloxone (NAL) suggests that the ability of EOPs to influence LH secretion may depend upon a functional NE system. We hypothesized that if EOPs required an interacting NE system to influence LH secretion, then rats that have ascendi

Inhibition of breathing after stimulation of muscle is mediated by endogenous opiates and GABA.

Stimulation of receptors in hindlimb muscles activates a supraspinal mechanism which causes a prolonged, post-stimulatory depression of breathing (Waldrop, T.G., F.L. Eldridge and D.E. Millhorn, 1982, Respir. Physiol. 50: 239-254). In the present study phrenic nerve responses to stimulation of hindlimb muscles were studied in anesthetized, paralyzed cats whose vagi and carotid sinus nerves had been cut. Body temperature and end-tidal PCO2 were kept constant with servocontrollers. The post-stimulatory depression of breathing was greatly attenuated in cats pretreated with an opiate antagonist (n

The role of endogenous opiates in the area postrema pressor pathway.

1. Intra-vertebral artery-administered angiotensin II acts at the area postrema to facilitate central sympathetic vasomotor activity. Recent evidence suggests a possible role of the opiate system in the mechanism of action of angiotensin II at the level of the brain stem. 2. In these experiments, we show that the morphine antagonist naloxone reduces significantly the magnitude of the pressor response to vertebral artery-infused angiotensin II. 3. Morphine, in contrast, doubled the peak of the vertebral response to identical doses of the peptide. Neither naloxone nor morphine affected the press

Enkephalin and substance P effects related to trigeminal pain.

Iontophoretic applications of enkephalin (20-150 nA) reduced the spontaneous firing frequency of nociceptive neurons in the trigeminal nucleus caudalis of decerebrated cats. The response evoked by noxious stimulation (tooth pulp) was gradually inhibited during the 1st minute of application of the opioid and generally remained depressed for 5 min after the current was turned off. These effects of enkephalin were blocked by intravenously or iontophoretically administered naloxone. Nonnociceptive neurons or nociceptive neurons responding to nonnoxious inputs were less frequently inhibited by enke

Endogenous opioids and opiate antagonists in autism: brief review of empirical findings and implications for clinicians.

Endogenous opioid dysfunction hypotheses for the development of autism are reviewed, along with clinical empirical studies of opiate antagonists in autism and self-injurious behaviour. There is not yet sufficient evidence to suggest the use of opiate antagonists in the treatment of autism. Further research, particularly of natrexone in severe self-injury, is warranted.

Opioid control of luteinizing hormone secretion in patients with hirsutism and hyperandrogenemia.

The present study was designed to evaluate the influence of hyperandrogenemia on the activity of the opioid system regulating LH secretion in menstruating women. Ten subjects presenting with hirsutism and hyperandrogenemia and 9 healthy normally cyclic subjects participated in the study. Naloxone or saline was administered on 2 different days both during the follicular (6-8 days after menstrual bleeding) and during the luteal phase of the menstrual cycle. Naloxone significantly increased plasma LH levels in the luteal, but not during the follicular phase of the cycle in both subject groups. It

Hormone ontogeny in the ovine fetus. XVIII. The effect of an opioid antagonist on luteinizing hormone secretion.

Endogenous opioid-like peptides influence gonadotropin release in adult animals and man; however, the role of these peptides in the regulation of fetal LH secretion is not known. We administered naloxone hydrochloride (1.3 mg/kg iv), a specific opioid receptor antagonist, to 22 chronically catheterized ovine fetuses of gestational ages 94-143 days (term = 147 days). As a control, each fetus also received the vehicle on a separate occasion, the sequence of the studies being randomized. After the administration of naloxone, LH secretion increased from 38.6 +/- 5.8 to 114 +/- 21 ng/h ml-1 (P less

Central opiate system modulation of the area postrema pressor pathway.

Angiotensin II, when given into the vertebral arteries, acts at the area postrema to augment central sympathetic vasomotor activity. The mechanism of action is unknown but recent evidence implicates an interaction with the opiate system. In dogs anesthetized with chloralose either alone or in combination with morphine, naloxone blunted the pressor response to vertebrally administered angiotensin II by 50%. Addition of morphine to dogs anesthetized with chloralose only doubled the pressor response to identical doses of angiotensin II. On the other hand, the magnitude of the pressor responses to

Differential effects of enkephalin within hippocampal areas.

The synthetic opiate D-Alanine-methionine enkephalin (D-Ala) affects neuronal activity of the fascia dentata (FD). D-Ala causes a depression of the population spike evoked by stimulation of the perforant path, which is antagonized by naloxone. In so called "Tandem" experiments it was shown that D-Ala had opposite effects on the CA1 pyramidal and FD granule cells. It is likely that in the FD the site of action of D-Ala is at the afferent synapses.

Anti-opiate (naloxone) suppression of Cushingoid degenerative changes in obese/SHR.

Young and mature, genetically obese and non-obese, spontaneously hypertensive rats (SHR) were injected with saline (controls) or naloxone for 12 weeks. Naloxone stilled the hyperphagia to a normal intake in the obese SHR (Obese/SHR) so that young Obese/SHR did not develop their usual massive obesity and mature Obese/SHR that had become massively obese were reduced to leanness. The naloxone-treated young, obese and non-obese SHR (controls) exhibited marked reduction of the weight of their pituitary and adrenal glands, whereas the pituitary and adrenal glands of naloxone-treated mature, obese an

Hormonal evidence for altered responsiveness to social stress in major depression.

In patients with major depression, abnormalities in baseline cortisol secretion and resistance to negative feedback are well established. However, it is unclear if patients with major depression have alterations in the hypothalamic-pituitary-adrenal (HPA) response to stressors. While other challenges to the HPA axis have used endocrine stimuli such as insulin-induced hypoglycemia, we now report of the response to a social stressor in patients with major depression and matched control subjects. We used the Trier Social Stress Test (TSST), a public speaking task followed by mental arithmetic cha

POMC neuropeptides and their receptors in the neuromuscular system of wobbler mice.

The pro-opiomelanocortin-derived peptides, beta-endorphin (beta-EP), ACTH and alpha-MSH act in concert to promote synaptogenesis and nerve growth in the neuromuscular system. Immunocytochemistry was used to detect the peptides in triceps muscles of normal developing and adult mice and in adult wobbler mice with motor neurone disease. Immunoreactivity for beta-EP and alpha-MSH/ACTH was present in some intramuscular motor nerves. There was a significantly higher proportion of immunoreactive nerves in the muscles of the developing and the diseased mice than in the normal adults. ACTH and beta-EP

HPA-axis hormones and prolactin responses to dextro-fenfluramine in depressed patients and healthy controls.

1. Like other authors we have established disturbances in central serotonergic neurotransmission in severely depressed patients by implementing hypothalamic pituitary adrenal (HPA)-axis hormones and prolactin responses to serotonin agonists or precursors. 2. Challenge probes with D,L fenfluramine have yielded controversial results. This substance, however, is not as serotonin-selective as previously believed. 3. Dextro(D)-fenfluramine, the dextrorotatory isomer of fenfluramine, constitutes a specific and potent serotonergic agonist. 4. In the present study the authors determined the following

Chronic kappa-opioid receptor antagonism delays the rise in blood pressure in spontaneously hypertensive rats.

A 4-week subcutaneous treatment with the kappa-opioid receptor antagonists, MR 2266 or MR 1452, delayed the age-dependent increase in systolic blood pressure in young spontaneously hypertensive rats (SHR), but not in normotensive rats. The content of beta-endorphin, [Leu5]enkephalin, and catecholamines in various brain tissues was not affected by MR 2266. Specificity of the kappa-opioid receptor antagonism was tested in SHR by injection of the kappa-opioid agonist, MR 2033, and the mu agonist, morphiceptin, after 28 days of MR 1452 or saline and 2 days after the end of treatment. The increase

[Cushing's syndrome in CRF-producing mediastinal carcinoid].

A 36-year-old patient developed marked pigmentation, marked myopathy and severe hypokalaemic alkalosis which at first pointed towards an ectopic ACTH syndrome. The dexamethasone test at a high dose indicated cortisol suppression. A mediastinal tumour was seen radiologically, but the sella was of normal size. Computed tomography provided indirect signs of a sellar space-occupying lesion which suggested an ectopic production of corticotropin-releasing factor (CRF) as cause of the Cushing's syndrome. CRF concentration in antecubital venous blood was markedly elevated to 280 ng/l. The mediastinal

[Endocrinological analysis of climacteric symptoms and gonadal dysfunction by CRF test].

Endocrinological studies on stressed climacteric women and some cases of gonadal dysfunction were carried out by analysing blood levels of adrenocorticotropic hormone (ACTH), beta-lipotropin (beta-LPH) and beta-endorphin (beta-EP) after the administration of synthetic corticotropin releasing factor (CRF, CRF test). Our results can be summarized as follows: 1. The responsiveness to CRF in perimenopausal and ovariectomized women rose and it corresponded with those of post gonadectomy in testicular feminization. 2. Responsiveness to CRF was decreased as estrogen levels decreased with the patient'

Alpha-methyldopa induces a naltrexone-insensitive antinociception and hypomotility in rats.

1. This study served to investigate whether endogenous opioid peptides play a role in the putative antinociceptive and the sedative actions of alpha-methyldopa. 2. In conscious normotensive rats, alpha-methyldopa induced hypotension, starting around 1 h and reaching a maximum 3-4 h after administration. Pretreatment with naltrexone resulted in an inhibition of alpha-methyldopa-induced hypotension. 3. alpha-Methyldopa dose-dependently increased hot plate latency which became evident after a 4 h lag period and reaching a maximum effect at 6 h. The antinociceptive effect of alpha-methyldopa was n
