Plant growth and development are intricately regulated by molecular mechanisms, with the mitogen-activated protein kinase (MAPK) signaling cascade and its associated modules being pivotal. In this study, we identified and characterized a member of the MAPKKK family, MtMAP3Kδ6, from Medicago truncatula. This gene, classified within the B3 subgroup of the MAPKKK family, was expressed across various tissues during plant growth. The knockout mutant of MtMAP3Kδ6 displayed dwarfism, characterized by reduced branching and smaller leaf size, whereas overexpression of MtMAP3Kδ6 in Medicago truncatula l

WRKY transcription factors have been implicated in the regulation of disease resistance associated with plant immune responses, which has crucial implications for defense responses against stress in plants. The role played by the PsnWRKY70 gene of Populus (Populus simonii × P. nigra) in triggering the mechanism between the phyllosphere microbiome and plant defense against foliar pathogens remains unclear. Molecular ecological network analysis demonstrated that the stability and complexity of the phyllosphere bacterial community of Populus were influenced by Alternaria alternata infection. Spec

Metal homeostasis in giant leucaena (Leucaena leucocephala subsp. glabrata) is of interest due to the plant's production of mimosine, an iron-chelating secondary metabolite. Real-time PCR performed on root and foliar tissue showed the upregulation of 19 genes following exogenous application of iron. Notable genes affected include glutathione synthase (20-fold increase in leaf), ferric chelate reductase (15-fold increase in root), mimosinase (20-fold increase in leaf) and nicotianamine synthase (30-fold increase in root). Transcriptome sequence data and 5'-RLM-RACE methods identified the comple

ERF transcription factors play a pivotal role in orchestrating plant growth and development processes. In this study, PagERF113 was isolated from the 84 K poplar (Populus alba × P. glandulosa) and its function was detected using molecular and genetic methods. Subcellular localization and trans-activation assays indicated that PagERF113 lacks transcriptional activation capability, residing in both the cell membrane and nucleus. Phenotypic analyses revealed that transgenic poplar lines overexpressing PagERF113 (OE) exhibited weedy growth compared to wild-type (WT), characterized by small leaves,

Upon pathogen attack, plants very quickly undergo rather complex physico-chemical changes, such as the production of new chemicals or alterations in membrane and cell wall properties, to reduce disease damages. An underestimated threat is represented by root parasitic nematodes. In Vitis vinifera L., the nematode Xiphinema index is the unique vector of Grapevine fanleaf virus, responsible for fanleaf degeneration, one of the most widespread and economically damaging diseases worldwide. The aim of this study was to investigate changes in the emission of biogenic volatile organic compounds (BVOC

BACKGROUND: Constitutive expression of Vitis vinifera polygalacturonase-inhibiting protein 1 (Vvpgip1) has been shown to protect tobacco plants against Botrytis cinerea. Evidence points to additional roles for VvPGIP1, beyond the classical endopolygalacturonase (ePG) inhibition mechanism, in providing protection against fungal infection. Gene expression and biochemical datasets previously obtained, in the absence of infection, point to the cell wall, and particularly the xyloglucan component of transgenic VvPGIP1 lines as playing a role in fungal resistance.
RESULTS: To elucidate the role of

Alternative splicing (AS) is a pivotal post-transcriptional mechanism for plant thermal adaptation, yet its dynamics in heat-tolerant woody species remain poorly understood. Here, we map the temperature-resolved AS landscape of the highly thermotolerant woody plant Rhododendron hainanense by combining Iso-Seq with RNA-seq. Transcriptomic profiling revealed a graded regulatory architecture: moderate heat (37 °C) elicited a targeted proteostatic response, whereas severe heat (42 °C) provoked a massive transcriptome expansion. Notably, this global AS surge coincided with a widespread downregulati

Circular RNAs (circRNAs) are emerging post-transcriptional regulators, yet their landscape and functional roles in rice under combined abiotic stress remain largely unexplored. Here, we systematically reanalyzed strand-specific RNA-seq data to characterize circRNAs responsive to simultaneous heat and drought stress. Following quality control, read mapping, and dual-algorithm prediction using CIRI2 and CIRCexplorer2, we identified 208 high-confidence circRNAs distributed across all 12 chromosomes. Comparative profiling revealed 83 circRNAs uniquely expressed in control samples, 51 in stressed s

The cysteine-rich polycomb-like protein (CPP) gene family is a class of transcription factors containing conserved cysteine-rich CRC structural domains that is involved in the regulation of plant growth and stress tolerance to adversity. Relative to other gene families, the CPP gene family has not received sufficient attention. In this study, six SlCPPs were identified for the first time using the most recent genome-wide identification data of tomato. Subsequently, a phylogenetic analysis classified SlCPPs into four subfamilies. The analysis of cis-acting elements in the promoter indicates tha

To gain insight into the response of mulberry to phytoplasma-infection, the expression profiles of mRNAs and proteins in mulberry phloem sap were examined. A total of 955 unigenes and 136 proteins were found to be differentially expressed between the healthy and infected phloem sap. These differentially expressed mRNAs and proteins are involved in signaling, hormone metabolism, stress responses, etc. Interestingly, we found that both the mRNA and protein levels of the major latex protein-like 329 (MuMLPL329) gene were increased in the infected phloem saps. Expression of the MuMLPL329 gene was

PhC2H2-ZFP45 may act as a transcriptional activator to up-regulate the expression of PhPAL2 and participate in the formation of floral fragrance. C2H2-zinc finger proteins (C2H2-ZFPs) are involved in the regulation of plant development and stress resistance. However, there are few studies on the effect of C2H2-ZFPs on the regulation of floral fragrance. Petunia hybrida has become an ideal model plant for studying floral volatile benzenoids/phenylpropanoids (FVBP) metabolism. To gain insight into the participation of C2H2-ZFPs in the regulation of floral fragrance in petunia, we performed a gen

Arbuscular mycorrhizal fungi (AMF) form symbiotic associations with plant roots, profoundly shaping root system architecture (RSA) and influencing nutrient acquisition in crops. This modulation begins during the early pre-symbiotic stage, when plants and fungi interact without physical contact. Root formation is orchestrated by signaling molecules, including hormones such as auxin (IAA) and strigolactones (SLs), as well as reactive nitrogen species such as nitric oxide (NO). However, the mechanisms by which AMF spores modulate these pathways to influence root development in rice remain largely

BACKGROUND: Fruit ripening is a coordinated process that leads to an increase in sugars, decrease in acids and accumulation of pigments. Blueberry fruit exhibit an atypical climacteric ripening behavior. These fruit display an increase in respiration and ethylene production during ripening, however ethylene synthesis is developmentally regulated. In this study, the effect of ethylene on blueberry fruit ripening was investigated via preharvest applications of ethylene-releasing plant growth regulators (PGRs), ethephon and 1-aminocyclopropane 1-carboxylic acid (ACC), in one southern highbush cul

Lettuce (Lactuca sativa) is a major crop and a member of the large, highly successful Compositae family of flowering plants. Here we present a reference assembly for the species and family. This was generated using whole-genome shotgun Illumina reads plus in vitro proximity ligation data to create large superscaffolds; it was validated genetically and superscaffolds were oriented in genetic bins ordered along nine chromosomal pseudomolecules. We identify several genomic features that may have contributed to the success of the family, including genes encoding Cycloidea-like transcription factor

The non-expressor of pathogenesis-related genes 1 (NPR1) is an essential positive regulator of salicylic acid (SA)-induced pathogenesis-related (PR) gene expression and systemic acquired resistance (SAR). Two novel full length NPR1-like genes; MNPR1A and MNPR1B, were isolated from banana by application of the PCR and rapid amplification of cDNA ends (RACE) techniques. The two identified MNPR1 sequences differed greatly in their expression profile using quantitative real time (qRT)-PCR following either elicitor or Fusarium oxysporum Schlecht f. sp. cubense (Smith) Snyd (Foc) treatment. Plants activate induced defenses through the recognition of molecular patterns. Like pathogen-associated molecular patterns (PAMPs), herbivore-associated molecular patterns (HAMPs) can be recognized by cell surface pattern recognition receptors leading to defensive transcriptional changes in host plants. Herbivore-induced defensive outputs are regulated by the circadian clock, but the underlying molecular mechanisms remain unknown. To investigate how the plant circadian clock regulates transcriptional reprogramming of a specific HAMP-induced pathway, we characterized the daytime and nighttime

Plant GASA/Snakin proteins (GASA: Gibberellic Acid-Stimulated Arabidopsis) constitute a family of secreted cysteine-rich peptides. The family is defined by 12 highly conserved cysteine residues forming up to six disulfide bonds within a thermostable helical scaffold. Phylogenetically restricted to vascular plants, the family ranges from 10 members in rice to approximately 40 in polyploid species such as peanut (Arachis hypogaea), with paralogous expansion driven by whole-genome, segmental, and tandem duplication events, while the 12-cysteine spacing pattern is maintained under purifying select

The phytohormone salicylic acid (SA) has a crucial role in plant physiology. Its role is best described in the context of plant response to pathogen attack. During infection, SA is rapidly accumulated throughout the green tissues and is important for both local and systemic defences. However, some genetic/metabolic variations can also result in SA overaccumulation in plants, even in basal conditions. To date, more than forty Arabidopsis thaliana mutants have been described as having enhanced endogenous SA levels or constitutively activated SA signalling pathways. In this study, we established

Poplar trees synthesize flavan-3-ols (catechin and proanthocyanidins) as a defense against foliar rust fungi, but the regulation of this defense response is poorly understood. Here, we investigated the role of hormones in regulating flavan-3-ol accumulation in poplar during rust infection. We profiled levels of defense hormones, signaling genes, and flavan-3-ol metabolites in black poplar leaves at different stages of rust infection. Hormone levels were manipulated by external sprays, genetic engineering, and drought to reveal their role in rust fungal defenses. Levels of salicylic acid (SA),

Agrobacterium tumefaciens infects and causes crown galls in dicot plants by transferring T-DNA from the Ti plasmid to the host plant via a type IV secretion system. This process requires appropriate environmental conditions, certain plant secretions, and bacterial regulators. In our previous work, a member of the LysR family of transcriptional regulators (LsrB) in Sinorhizobium meliloti was found to modulate its symbiotic interactions with the host plant alfalfa. However, the function of its homolog in A. tumefaciens remains unclear. In this study, we show that the LsrB protein of A. tumefacie
