MELAS mitochondrial DNA mutation A3243G reduces glutamate transport in cybrids cell lines.

MELAS (mitochondrial myopathy, encephalopathy, lactic acidosis and stroke-like episodes) is commonly associated with the A3243G mitochondrial DNA (mtDNA) mutation encoding the transfer RNA of leucine (UUR) (tRNA (Leu(UUR))). The pathogenetic mechanisms of this mutation are not completely understood. Neuronal functions are particularly vulnerable to alterations in oxidative phosphorylation, which may affect the function of the neurotransmitter glutamate, leading to excitotoxicity. In order to investigate the possible effects of A3243G upon glutamate homeostasis, we assessed glutamate uptake in

Glutamate-like immunoreactivity marks compartments of the mushroom bodies in the brain of the cricket.

In the mushroom bodies of the brain of the cricket Gryllus bimaculatus, the distribution of glutamate-like immunoreactivity is shown by using several immunocytochemical staining protocols and confocal and conventional microscopy. Glutamate-like staining of intrinsic cells of mushroom bodies (Kenyon cells), their axons and projections, is demonstrated for the first time. Two types of Kenyon cells constituting distinct, separated populations within the perikaryal layer and in prominent neuropilar subcompartments exhibit strong (type III cells) or medium (type II cells) glutamate-like immunoreact

Non-opioid antitussives inhibit endogenous glutamate release from rabbit hippocampal slices.

The non-opioid antitussive drugs, dextromethorphan, caramiphen and carbetapentane, are also anticonvulsant. The effects of these antitussives on potassium-stimulated release of endogenous amino acids from rabbit hippocampal slices was tested. All 3 drugs significantly reduced the release of glutamate, with carbetapentane being the most potent (IC50 approximately 40 microM). We suggest that the anticonvulsant action may be due to their ability to decrease glutamate release.

Biochemical development of the human brain. III. Benzodiazepine receptors, free gamma-aminobutyrate (GABA) and other amino acids.

Benzodiazepine receptor binding and the concentration of the free tissue GABA, glutamate, glutamine, aspartate, and 13 other amino acids were estimated in specimens of cerebral cortex and cerebellum taken from normal foetuses and from perinatal and postnatal infants and adults postmortem. Compared with the ontogenetic increase, previously reported, in [3H]muscimol binding, that of [3H]flunitrazepam binding developed more rapidly in the cerebellum and less rapidly in the cerebral cortex. The concentration of GABA in both brain parts increased with age more rapidly than did that of glutamate dec

Hypoxic Adaptation of Mitochondrial Metabolism in Rat Cerebellum Decreases in Pregnancy.

Function of brain amino acids as neurotransmitters or their precursors implies changes in the amino acid levels and/or metabolism in response to physiological and environmental challenges. Modelling such challenges by pregnancy and/or hypoxia, we characterize the amino acid pool in the rat cerebellum, quantifying the levels and correlations of 15 amino acids and activity of 2-oxoglutarate dehydrogenase complex (OGDHC). The parameters are systemic indicators of metabolism because OGDHC limits the flux through mitochondrial TCA cycle, where amino acids are degraded and their precursors synthesiz

Preparation and preliminary quality evaluation of aspirin/L-glutamate compound pellets.

L-glutamate is an important component of protein. It can prevent gastrointestinal damage caused by NSAIDs. We constructed two-phase enteric-coated granules of aspirin and L-glutamate compound by extrusion spheronization method and fluidized bed coating. The subliminal effective dose of L-glutamate is 100 mg/kg tested by model of gastric ulcer of rats induced by aspirin and drug administration. HPLC-UV and UV-Vis methods were adopted to determine content and cumulative release of aspirin and L-glutamate as quality analysis method indexes. The prescription and process optimization were carried o

T-Cells and excitotoxicity: HIV-1 and other neurodegenerative disorders.

Until recently the central nervous system (CNS) was considered an immune-privileged site, however, technological and immunological advances have resulted in the CNS being reclassified as an "immune-specialized site." The immune cells, particularly T-cells, continuously patrol the brain and are involved in neuroimmune responses. As such, any changes in the brain microenvironment could affect the physiological functioning of T-cells. Particularly, neurotransmission- associated abnormalities, such as excitotoxicity associated with hypersecretion of glutamate, could severely affect the neuroimmune

Wernicke's encephalopathy: an excitotoxicity hypothesis.

Thiamine deficiency is a recognized cause of Wernicke's encephalopathy (WE), a condition in which small necrotic lesions are found in close proximity to the third and fourth ventricles and the Sylvian aqueduct. Although the neuropathology of WE is well-established, the pathogenic mechanisms that determine the formation and distribution of brain lesions identified with this illness are not understood. It is proposed here that glutamate neurotoxicity causes the brain lesions in WE. Glutamic acid decarboxylase (GAD), an enzyme mainly confined to the central nervous system, protects most regions o…

Chronic methionine sulfoximine administration reduces synaptosomal aspartate and glutamate in rat striatum.

Methionine sulfoxime (MS) is an inhibitor of glutamine synthetase, an astroglial enzyme believed to be involved in the maintenance of glutamine, a major precursor for neurotransmitter pools of the excitatory amino acids aspartate and glutamate in striatal afferent axon terminals. MS was infused for 7 days (24 micrograms/day) into the lateral cerebral ventricle of rats. On the side of MS infusion, there was a decrease of striatal synaptosomal aspartate (61%), glutamine (63%), and glutamate (48%), while taurine and gamma-aminobutyrate were unaltered when compared to vehicle-treated rats. The res

Interactions between glutamate, GABA, acetylcholine and histamine in the periaqueductal gray's control of vocalization in the squirrel monkey.

In the squirrel monkey (Saimiri sciureus), vocalization was elicited by periaqueductal injections of the glutamate agonist homocysteic acid, the acetylcholine agonist carbachol, the GABA antagonist bicuculline and the monoaminergic transmitter histamine. All chemically induced vocalizations could be blocked completely by prior periaqueductal injections of the non-specific glutamate antagonist kynurenic acid and the GABA agonist muscimol. The acetylcholine antagonist scopolamine and the histamine antagonist diphenhydramine, in contrast, blocked their respective transmitter agonists' vocalizatio

Specificity of GABA as a stimulator of dopamine release in rat substantia nigra.

The effects of amino acids and the benzodiazepine, flurazepam, on spontaneous and potassium-stimulated [3H]dopamine (DA) release were studied in slices of rat substantia nigra. None of the amino acids tested liberated [3H]DA directly. Flurazepam increased [3H]DA output by 51% and this effect was partially blocked by bicuculline but not by picrotoxin. GABA, L-glutamate and flurazepam, but none of the other compounds, potentiated the release of [3H]DA evoked by 20 mM KCl. The facilitation produced by flurazepam, but not by GABA or L-glutamate, was weakly antagonized by bicuculline and not picrot

Evidence of adaptation of maternofetal transport of glutamine relative to placental size in normal mice, and in those with fetal growth restriction.

KEY POINTS: Fetal growth restriction (FGR) is a major risk factor for stillbirth and has significant impact upon lifelong health. A small, poorly functioning placenta, as evidenced by reduced transport of nutrients to the baby, underpins FGR. It remains unclear how a small but normal placenta differs from the small FGR placenta in terms of ability to transfer nutrients to the fetus. Placental transport of glutamine and glutamate, key amino acids for fetal growth, was assessed in normal mice and those with FGR. Glutamine and glutamate transport was greater in the lightest versus heaviest placen

Thrombin potentiates D-aspartate efflux from cultured astrocytes under conditions of K+ homeostasis disruption.

Thrombin levels increase in brain during ischemia and hemorrhagic episodes, and may contribute to excitotoxic neural damage. This study examined the effect of thrombin on glutamate efflux from rat cortical cultured astrocytes using 3H-D-aspartate as radiotracer. The glutamate efflux was initiated by addition of 100 mM K+ plus 1 mM ouabain (K/O) to replicate extracellular and intracellular ionic changes that occur during cerebral ischemia. Upon exposure to K/O, astrocytes swelled slowly and progressively with no evidence of volume regulation. The K/O-induced swelling was inhibited by 65% with b

Activation of the Pancreatic "Metabolic Synapse" Aggravates Type 2 Diabetes Mellitus by Inducing PANoptosis in β-Cells.

UNLABELLED: Pancreatic β-cells play a central role in type 2 diabetes mellitus (T2DM), yet the interactions between β-cells and stromal components within the islet microenvironment remain poorly defined. We investigated the contribution of pancreatic fibroblasts to β-cell dysfunction and T2DM progression. We used single-cell sequencing technology and in vitro experiments to investigate the mechanisms by which bariatric surgery ameliorates T2DM. We introduce the novel concept of a "metabolic synapse" to describe the interaction between pancreatic fibroblasts and β-cells. Our findings reveal tha

Nitric oxide dependence of glutamate-mediated modulation at a vertebrate neuromuscular junction.

Recent evidence has revealed a contribution of glutamate in the stereotyped cholinergic neuromuscular transmission. Indeed, receptors, transporters and glutamate itself are present at the neuromuscular junction (NMJ) while glutamate activation of metabotropic receptors (mGluRs) decreases synaptic transmission and mediates depression through presynaptic mechanisms. However, we have shown that the mGluRs are located postsynaptically, inconsistent with the presynaptic action of glutamate. In the present study, we tested whether nitric oxide (NO) serves as a retrograde messenger mediating the dist

The safety evaluation of monosodium glutamate.

L-Glutamic acid and its ammonium, calcium, monosodium and potassium salts were evaluated by the Joint FAO/WHO Expert Committee on Food Additives (JECFA) in 1988. The Committee noted that intestinal and hepatic metabolism results in elevation of levels in systemic circulation only after extremely high doses given by gavage (>30mg/kg body weight). Ingestion of monosodium glutamate (MSG) was not associated with elevated levels in maternal milk, and glutamate did not readily pass the placental barrier. Human infants metabolized glutamate similarly to adults. Conventional toxicity studies using die…
