Evaluation of the Long-Term Administration of Proton Pump Inhibitors (PPIs) in the Mineral Nutrient's Bioavailability.

Nutrient absorption in the human body can be influenced by factors, such as diet and medication use. The interaction between drugs and nutrients may lead to adverse effects, primarily due to reduced levels of essential elements. Therefore, evaluation of these interactions is important to prevent health complications. Proton pump inhibitors (PPIs), widely used to reduce gastric acid production, have been associated with interactions that may cause disorders. This study aims to assess the continuous use of PPIs and their effect on the bioavailability of Fe, Ca, Zn, Mg, Cu, and K in the diet. Rat

Unexpected effects of antibiotics on microbiota-mediated drug metabolism in a fistulated dog model.

The human gut microbiome plays a crucial role in the metabolism of drugs, with various bacterial species catalysing the biotransformation of pharmaceutical compounds. In vitro microbiota models, typically derived from faecal material, have been employed to study this phenomenon. However, they often do not fully capture the complex and dynamic environment of the living colon. This study aimed to evaluate whether in vitro models accurately reflect in vivo microbiota-drug interactions by comparing both approaches within the same individual animal via a fistulated dog model, with the fistula locat

[Lipophilic yeasts of the genus Malassezia and skin diseases. I. Seborrhoeic dermatitis].

Recent technological advances have revived the interest in Malassezia yeasts and their clinical role, which has long been a matter of controversy because of their fastidious nature in vitro and relative difficulty in isolation, cultivation and identification. Lipophilic yeasts of the genus Malassezia form a part of normal microbial flora of healthy human (and animal) skin, but they also have been associated with several dermatological diseases, like seborrhoeic dermatitis and atopic dermatitis. Our understanding of the interactions between Malassezia and the host might provide new opportunitie

A review of the physiological bases of the anorexic action of phenylpropanolamine (d,1-norephedrine).

Phenylpropanolamine (PPA) is a phenethylamine that induces a variety of effects including anorexia and weight loss. The present review compares the acute anorexic effects of PPA with those of amphetamine in animals and humans, describes the persistent chronic effects of PPA on feeding as well as differences in anorexic potency between the norephedrine enantiomers (d-norephedrine less than 1-norephedrine) and summarizes the generality of PPA anorexia. The review also describes the putative mechanisms by which PPA is thought to reduce feeding behavior including alteration of gastric emptying and

Experimental Models to Investigate Viral and Cellular Dynamics in Respiratory Viral Co-Infections.

Respiratory viral co-infections by viruses such as influenza virus, SARS-CoV-2, and respiratory syncytial virus (RSV) are a significant clinical issue in high-risk populations such as children, elderly patients, and immunocompromised individuals. Sequential and simultaneous co-infections exacerbate disease severity, leading to acute respiratory distress syndrome (ARDS), prolonged hospitalization, and increased mortality. Molecular and immunological interactions are complex, context-dependent, and largely unknown. Experimental models of infection that accurately mimic human respiratory physiolo

Microbiota-drug interactions: Impact on metabolism and efficacy of therapeutics.

The microbiome not only represents a vital modifier of health and disease, but is a clinically important drug target. Therefore, study of the impact of the human microbiome on drug metabolism, toxicity and efficacy is urgently needed. This review focuses on gut and vaginal microbiomes, and the effect of those microbiomes or components thereof on the pharmacokinetics of specific chemotherapeutic agents, immunotherapies, anti-inflammatory and antimicrobial drugs. In some cases, the presence of specific bacterial species within the microbiome can alter the metabolism of certain drugs, such as che

Cellular and molecular function of mucolipins (TRPML) and polycystin 2 (TRPP2).

Mucolipins (transient receptor potential mucolipin, TRPML) and polycystin-2 proteins (transient receptor potential polycystin, TRPP) constitute two small families of cation channels with motif and sequence similarities to the transient receptor potential (TRP) class of non-selective cation channels. Genetic defects in TRPML1 and TRPML3 in humans and in animal models cause the accumulation of large vacuoles, leading to a variety of cellular phenotypes including neurological and neurosensory deficiencies. TRPML1 is a Ca(2+)-, K(+)-, and Na(+)-permeable cation channel sensitive to pH changes, and

Partial characterization and tissue distribution of the feline mu opiate receptor.

Heroin abuse is a common route of acquiring HIV-1 infection. However, the effects of opiates on lentivirus disease progression are not well understood. Feline immunodeficiency virus is recognized as a good animal model for HIV-1, but characterization of the opiate receptor system in cats is lacking. Here we report the partial sequencing of the feline mu opiate receptor (MOR) and demonstrate a homology of 92 and 93% to the published human MOR sequences. Additionally, MOR transcripts were detected in the feline brain and tonsil but not in the spleen. Also, specific receptor ligand interactions w

Platelets, prostaglandins and inflammation.

In exudates of implanted sponges in rats, made thrombocytopenic by the administration of anti-platelet serum, there are significant reductions in the platelet and leucocyte counts and of the content of prostaglandin-like activity. It is concluded that platelets migrate into the developing sponge exudates, are the source of the prostaglandins and interact with complement to cause leucocyte migration. In normal animals the administration of indomethacin and sodium salicylate cause similar effects to thrombocytopenia whereas the injection of human plasma fraction affects only the leucocyte migrat

Modeling gliomas with organoids: reconstructing the human neural microenvironment for translational neuro-oncology.

Gliomas represent some of the most lethal and biologically complex tumors of the central nervous system, with poor outcomes across both adult and pediatric populations. Beyond tumor-intrinsic genetic alterations, glioma progression is increasingly recognized to be driven by dynamic interactions with the neural microenvironment, particularly through direct tumor-neuron communication. Studies of adult and pediatric gliomas, including glioblastoma (GBM) and diffuse midline glioma (DMG), using single-cell profiling and multi-omics technologies have revealed extensive tumor intrinsic and extrinsic

Identification of the Single Immunodominant Region of the Native Human CC Chemokine Receptor 6 Recognized by Mouse Monoclonal Antibodies.

Chemokines and their receptors play an important role in cell trafficking and recruitment. The CCR6 chemokine receptor, selectively expressed on leukocyte populations, has been shown to play a deleterious role in the pathogenesis of various chronic inflammatory diseases and, as such, may constitute a prime target in the development of immunotherapeutic treatment. However, to date no neutralizing mouse monoclonal antibodies (mAbs) specific for this chemokine receptor have been reported, whereas information on small molecules capable of interfering with the interaction of CCR6 and its ligands is

Aberrant salience is selectively associated with ventral midbrain connectivity to the dorsal but not ventral striatum.

The dorsal midbrain-striatal dopaminergic stream encodes salience in animals, while it's unknown in humans. The midbrain-striatal dopaminergic hyperactivity is postulated to attribute aberrantly-heightened salience to ordinary stimuli, forming delusion and hallucination. While animal studies implicate the ventral midbrain-striatal stream, patient studies suggest the dorsal stream, leaving the precise localization of aberrant salience unclear. We investigated these issues by examining midbrain-striatal functional connectivity. Resting-state functional MRI was acquired from 116 patients with sch

Synanthropic mammals as sentinels of multi-pathogen circulation at a wildlife-urban interface.

Peri-urban forest fragments used for ecotourism concentrate interactions among wildlife, domestic animals, and humans, creating conditions that favour pathogen transmission through environmental, vector-borne, and trophic pathways. In these landscapes, synanthropic small mammals may act as integrative indicators of pathogen exposure because they interact with wildlife, domestic animals, and human-modified environments. This study conducted multi-pathogen surveillance in synanthropic small mammals to investigate infection patterns at a wildlife-urban interface in a Brazilian ecotourism area. Se

In utero exposure to endocrine disruptors and developmental neurotoxicity: Implications for behavioural and neurological disorders in adult life.

Endocrine disrupting chemicals (EDCs) are a class of environmental toxicants that interfere with the endocrine system, resulting in developmental malformations, reproductive disorders, and alterations to immune and nervous system function. The emergence of screening studies identifying these chemicals in fetal developmental matrices such as maternal blood, placenta and amniotic fluid has steered research focus towards elucidation of in utero effects of exposure to these chemicals, as their capacity to cross the placenta and reach the fetus was established. The presence of EDCs, a majority of w

Ability of rabies vaccine strains to elicit cross-neutralising antibodies.

Two European Bat Lyssaviruses (EBLV-1 and EBLV-2) have been identified (n > 750 cases) in European bat species. In addition, EBLVs have been detected as "spillover" cases in three humans, one stone marten and four sheep. A further concern is the interaction of companion animals with infected bats and the possibility of subsequent lyssavirus infection. Cat-bat interactions represent approximately 32 % (n = 398) of passive lyssavirus surveillance submissions at the Veterinary Laboratories Agency (VLA) (1987-2004). Cats therefore represent a potentially significant spillover host. This study eval

The rabies virus matrix protein (RABV M) interacts with host histone deacetylase 6 (HDAC6) to activate the MEK/ ERK signaling pathway and enhance viral replication.

Rabies virus (RABV) is the causative agent of rabies, posing a severe threat to human and animal health. The matrix (M) protein of RABV plays crucial roles during viral infection. In this study, we identified RABV M protein interacted with host histone deacetylase 6 (HDAC6) through a combination of immunoprecipitation and mass spectrometry analysis. Specifically, the catalytic domains of HDAC6 (amino acids 435-835) was shown to be critical for the interaction between HDAC6 and the RABV M protein. Overexpression of HDAC6 significantly enhanced RABV replication, whereas inhibition of HDAC6 expre

A comprehensive review on potential drug-drug interactions of proton pump inhibitors with antidiabetic drugs metformin and DPP-4 inhibitors.

A drug interaction is a condition in which two or more drugs are taken at the same time. Type 2 diabetes mellitus is a significant contributor to polypharmacy. Proton pump inhibitors (PPIs) are often prescribed in combination with metformin or DPP-4 inhibitors (sitagliptin, saxagliptin, linagliptin, and alogliptin) or a combined dose of metformin and DPP-4 inhibitor to treat gastritis in diabetic patients. This review article mainly focused on evaluating the potential drug-drug interactions (DDIs) between PPIs (i.e. esomeprazole, lansoprazole, omeprazole, pantoprazole, and rabeprazole) with me

Defining parameters for successful immunocytotherapy of persistent viral infection.

Persistent infections with viruses such as HIV, Epstein-Barr virus, cytomelagovirus, and hepatitis B and C viruses continue to be major human health problems. Immunocytotherapy for persistent viral infections has proven successful in animal models but less effective in humans. While the requirement of antigen-specific CD8(+) T cells is known, the precise role of CD4(+) T cells as regards specific priming, numbers needed, and interaction with CD8(+) T cells is less clear. To address these issues, we used a mouse model of persistent virus infection in which adoptive transfer of T cells effective

Myocardial Tissue Engineering With Cells Derived From Human-Induced Pluripotent Stem Cells and a Native-Like, High-Resolution, 3-Dimensionally Printed Scaffold.

RATIONALE: Conventional 3-dimensional (3D) printing techniques cannot produce structures of the size at which individual cells interact. OBJECTIVE: Here, we used multiphoton-excited 3D printing to generate a native-like extracellular matrix scaffold with submicron resolution and then seeded the scaffold with cardiomyocytes, smooth muscle cells, and endothelial cells that had been differentiated from human-induced pluripotent stem cells to generate a human-induced pluripotent stem cell-derived cardiac muscle patch (hCMP), which was subsequently evaluated in a murine model of myocardial infarct
