Psoriasis is a chronic autoimmune skin disorder with no cure, posing challenges in long-term therapy and economic burden. Sweroside (SOS), an iridoid compound from Gentiana, shows promise for treatment due to its anti-inflammatory properties. In this study, SOS significantly reduced erythema, thickening, and scaling in IMQ-induced psoriasiform mice, lowered serum TNF-α and IL-1β levels, and suppressed inflammatory marker expression. Molecular docking revealed strong binding to IL-1β and NLRP3 proteins. Western blot and RT-PCR confirmed that SOS inhibited NLRP3, Cleaved-Caspase-1, ASC, and IL-1

In 2022, the American Heart Association updated "Life's Simple 7" to "Life's Essential 8" (LE8). In June 2023, the Delphi consensus proposed that metabolic dysfunction-associated steatotic liver disease (MASLD) should be the new successor of the term nonalcoholic fatty liver disease. We aimed to explore the association of 4 health behaviors defined by LE8 with MASLD and to systematically investigate the mediating role of inflammatory markers and oxidative stress. This study investigated cross-sectional data from 3691 adults in the 2017 to 2018 National Health and Nutrition Examination Survey.

Atopic dermatitis (AD) is a chronic inflammatory skin disorder characterized by barrier dysfunction, immune dysregulation, and oxidative stress. Effective therapy requires strategies that improve local drug retention while addressing both inflammation and barrier repair. In this study, hyaluronic acid-butyrate (HAB) conjugates with different molecular weights were synthesized and evaluated as dual-functional therapeutics for AD. IVPT using normal and AD-like skin demonstrated that HAB, particularly 5 k-HAB, achieved superior penetration and markedly enhanced skin retention (6.47-fold) compared

Blood and airway eosinophilia represent markers for the endotype-driven treatment of allergic asthma. Little is known on mechanisms that link eosinophils and airway epithelial cells before and after these cells are infiltrated by eosinophils during allergic response. Given that innate immune mechanisms, mainly mediated by epithelial-derived cytokines (interleukin [IL]-33, IL-25, TSLP [thymic stromal lymphopoietin]), induce eosinophil-maturing/attractive substances, we thought to evaluate the crosstalk between eosinophils and airway epithelial cells in the context of IL-33-mediated allergic inf

BACKGROUND: Emerging evidence links macrophage overactivation to sepsis-associated acute lung injury (ALI), yet the role of lung tissue-derived extracellular vesicles (Ti-EVs) in this process remains unclear. This study combines transcriptomic profiling and functional validation to reveal how Lung Ti-EVs mediate macrophage polarization through miRNA-dependent NLRP3 inflammasome activation.
METHODS: We established a sepsis mouse model, extracted and characterized lung tissue-derived EVs, performed high-throughput transcriptome sequencing and bioinformatics analysis. Intratracheal administratio

Sepsis is a life-threatening condition characterized by organ dysfunction, with the liver being particularly vulnerable due to inflammation triggered by Kupffer cell activation. Ferroptosis, an iron-dependent form of regulated cell death associated with macrophages, has emerged as a key pathogenic mechanism. This study aimed to investigate the protective effects of coptisine (COP), a natural alkaloid, against sepsis-induced hepatic ferroptosis and injury using in vivo and in vitro models. Sepsis was induced in mice via cecal ligation and puncture (CLP) or lipopolysaccharide (LPS) challenge, fo

Ulcerative colitis (UC) is a chronic, immune-mediated disorder with limited treatment efficacy due to drug resistance. As key immune effectors, Macrophages drive UC pathogenesis: The M1 polarization promoted through P-selectin/PSGL-1 binding between platelets and macrophages exacerbates inflammation. Rutin-PEG-PLGA nanoparticles (P@Rut) were engineered by encapsulating rutin in PEG-PLGA cores. Biomimetic platelet membrane nanoparticles (PP@Rut) were synthesized by extracting platelet membranes and coating P@Rut. The blockade of platelet-macrophage interactions by PP@Rut was assessed in vitro a

BACKGROUND: While platelet hyperreactivity constitutes an independent risk factor for major adverse cardiovascular events (MACEs) in coronary artery disease, its molecular underpinnings remain poorly characterized. Recent advances in transcriptomic profiling have revealed potential associations with specific RNA signatures. Through systematic bioinformatics analysis of differential gene expression patterns and pathway activation in CHD patients, this study aims to elucidate key molecular regulators of platelet hyperactivity, establishing a theoretical framework for developing precision therape

INTRODUCTION: Preterm hypoxic-ischemic encephalopathy (pHIE) is a complex brain injury that contributes to chronic neural inflammation and neurological disorders. The signs and symptoms of in utero pHIE can often be overlooked, untreated or lumped into more generic conditions such as encephalopathy of prematurity (EOP). Clinical interventions like hypothermia and erythropoietin do not improve pHIE. We characterized a murine model for pHIE, which includes hypoxia and maternal factors as a cost-effective alternative to large animal models of HIE.
METHODS: We injected pregnant mouse dams with LP

Older adults with established cardiovascular diseases (CVD) are at elevated risk of heart failure (HF). Frailty, a hallmark of multi-system aging, may contribute to HF development through inflammation. However, population-based evidence remains scarce. Leveraging data from 49,530 CVD patients in the UK Biobank, frailty was measured by five components: weight loss, exhaustion, low physical activity, slow walking speed, and low grip strength. We employed Cox regression models to assess the association between frailty and incident HF, and conducted mediation analyses to evaluate mediating roles o

PURPOSE: Adenosine deaminase 2 Deficiency (DADA2) is an autoinflammatory disease characterized by systemic vasculopathy, strokes and mild immunodeficiency. Recently NETosis has been implicated in the pathogenesis of Deficiency of Adenosine Deaminase 2. To deep investigate the possible effects of NETs on the immune system we characterized proteomic profile of NETs from DADA2 as compared to HD and Polyarteritis Nodosa (PAN) patients. To determine if NETs contain possibly immunogenic antigens we study functional aspects on Dendritic Cells after in vitro stimulation with NETs.

BACKGROUND: Vernonia anthelmintica (L.) Willd., containing isorhamnetin (ISO) as an active compound, is a conventional treatment for vitiligo within the Xinjiang region. We previously demonstrated the protective role of ISO in mitigating oxidative damage within HaCaT cells, suggesting its therapeutic potential in treating vitiligo. However, its mechanism and effect on melanocytes immune damage remain unclear.
AIM: Here, we examined the effect of ISO on inflammation and apoptosis in melanocytes. Moreover, we comprehensively analyzed key genes in vitiligo treatment.
METHODOLOGY: We employed fl

Matrix metalloproteinases (MMPs) are a major group of proteases known to regulate the turnover of the extracellular matrix (ECM). We observed that induced MMP-12 promotes eosinophilic inflammation-related epithelial cell mesenchymal transition (EMT), bronchial fibrosis, and airway obstruction in an allergen-exposed mouse model of chronic airway diseases in allergen-exposed mice and in airway-specific CC10-IL-13-overexpressed mice. Our histological analysis showed that the parabronchial and perivascular accumulation of eosinophils, fibroblasts, and collagen is significantly decreased in MMP-12-

Eosinophils are the predominant immune cells implicated in the pathogenesis of asthma, highlighting the need for strategies to mitigate their effects. While previous studies have implicated the importance of lysosomes in eosinophil function, the precise role of lysosomes in eosinophil activation during asthma remains insufficiently understood. In this study, we demonstrate that lysosomal acidity and cathepsin L activity in eosinophils are elevated in asthmatic patients and mouse models. Genetic deletion or pharmacological inhibition of cathepsin L in eosinophils attenuates allergic airway infl

INTRODUCTION: Intestinal inflammation is a chronic, relapsing disorder of the gastrointestinal tract characterized by dysregulated immune responses, microbial dysbiosis, and environmental influence. Pathogen clearance is related to the severity of intestinal diseases and macrophage apoptosis. Escherichia coli Nissle 1917 (EcN) alleviates the intestinal inflammation caused by Salmonella enterica serotype Salmonella Typhimurium (Salmonella Typhimurium, S. Tm) infection.
METHODS: Through gene editing, we found that the regulatory gene arcA largely affects arginine production in EcN via the argin

Dysregulated tissue-resident macrophages (TRMs) contribute to the pathogenesis of inflammatory bowel disease (IBD) and multiple sclerosis (MS). Uncovering molecular regulators of the divergent role of TRMs in inflammation can advance therapeutic strategies for autoimmune disorders. Here, a significant downregulation of SMAD-specific E3 ubiquitin protein ligase 2 (SMURF2) is reported in TRMs within inflamed intestinal tissues from both IBD patients and mouse models. Notably, TRM-specific deficiency of Smurf2 significantly exacerbates TRM proliferation in dextran sulfate sodium (DSS)-induced col

BACKGROUND: Volatile organic compounds (VOCs) are ubiquitous environmental pollutants which have been suggested to have adverse effects on human health. While the influence of environmental pollutant exposures on periodontitis has attracted elevating attention in recent years, the epidemiological evidence on the association between VOCs exposure and periodontitis was scarce. This study aimed to investigate the potential mediating role of systemic inflammation factors in the complex association between VOCs exposure and periodontitis.
METHODS: Utilizing data from the National Health and Nutrit

BACKGROUND: Thoracic aortic dissection (TAD) is an irreversible cardiovascular disorder with high mortality and morbidity. However, the molecular mechanisms remain elusive. Thus, identifying an effective therapeutic target to prevent TAD is especially critical. The purpose of this study is to elucidate the potential mechanism of inflammation and vascular smooth muscle cell (VSMCs) phenotypic switch in β-aminopropionitrile fumarate (BAPN)-induced TAD.
METHODS: A mouse model of TAD induced by BAPN and IL-1β -stimulated HVSMCs in vivo and in vitro models, respectively. ACE2 Knockdown mice treate

PURPOSE: Our team identified a new cardiac glycoside, Toxicarioside H (ToxH), in a tropical plant. Previous research has indicated the potential of cardenolides in mitigating inflammation, particularly in the context of NETosis. Therefore, this study sought to examine the potential of ToxH in attenuating allergic airway inflammation by influencing the immune microenvironment.
METHODS: An OVA-induced airway inflammation model was established in BALB/c mice. After the experiment was completed, serum, bronchoalveolar lavage fluid (BALF), and lung tissue samples were collected and further examine
