Expression of xenobiotic metabolizing cytochrome P450 genes in a spinosad-resistant Musca domestica L. strain.

BACKGROUND: Spinosad is important in pest management strategies of multiple insect pests. However, spinosad resistance is emerging in various pest species. Resistance has in some species been associated with alterations of the target-site receptor, but in others P450s seems to be involved. We test the possible importance of nine cytochrome P450 genes in the spinosad-resistant housefly strain 791spin and investigate the influence of spinosad on P450 expression in four other housefly strains. RESULTS: Significant differences in P450 expression of the nine P450 genes in the four strains after sp

Evaluation of the efficacy of bendiocarb in indoor residual spraying against pyrethroid resistant malaria vectors in Benin: results of the third campaign.

BACKGROUND: Since 2008, the National Malaria Control Program (NMCP) has been engaged in the implementation of indoor residual spraying (IRS) in Benin. The first and second round was a success with a drastic decrease of malaria transmission in areas under IRS. We present here the results of the third round. The purpose of this study was to compare the results of the third round of IRS to those achieved during the first two rounds. A second success of IRS will enable the Government of Benin to extend the strategy to other areas. METHODS: Mosquito collections were carried out in the department o

Exploring the contributions of bed nets, cattle, insecticides and excitorepellency to malaria control: a deterministic model of mosquito host-seeking behaviour and mortality.

Domestic and personal protection measures against malaria exposure either divert host-seeking vectors to other hosts or kill those attempting to feed. Here, we explicitly model mosquito host-seeking processes in the context of local host availability and elucidate the impacts and mechanisms of pyrethroid-treated bed nets in Africa. It has been suggested that excitorepellent insecticides could increase exposure of unprotected humans by concentrating mosquito biting activity on this vulnerable group. This worst-case scenario is confirmed as a possibility where vector populations lack alternative

Differential tolerance of two morphological variants of Culex tritaeniorhynchus (Diptera: Culicidae), a Japanese encephalitis vector, to pyrethroid insecticides in Mysore, India.

Five synthetic pyrethroids, deltamethrin, cypermethrin, permethrin, alpha-cypermethrin, and lambdacyhalothrin, were tested on the larvae of 2 morphological variants of Cx. tritaeniorhynchus collected from Mysore City (ground pools) and outside Mysore (paddy fields), in Karnataka State, India. The morphological characters, ie, length and width of siphon, siphonal index, comb scale numbers, pecten teeth numbers, length of anal gills and anal gill index of larvae of two Cx. tritaeniorhynchus populations were found to be significantly different (p<0.05). To elicit further detail of these two varia

Activity and residual effect of two formulations of lambdacyhalothrin sprayed on palm leaves to Rhodnius prolixus.

The insecticidal activity and residual effect of two formulations of lambdacyhalothrin were evaluated with Rhodnius prolixus; laboratory and field tests were conducted in the State of Chiapas, Mexico. The results indicate that the lethal concentrations of the active ingredient of SC (LC50 = 2.37 and LC90 = 8.5 mg, a.i./m2) were 4-8 times than those with the insecticide WP applied on R. prolixus bugs in palm leaves, a common building material for thatched roofs. Other investigators in South America recommended applying 30 mg a.i./m2 in porous materials; we obtained that the products WP and SC w

CYP4C64 Directly Metabolizes Nitenpyram to IM-1-4 Conferring Resistance in the Whitefly, Bemisia tabaci.

Neonicotinoid insecticides are widely used to control sap-feeding pests, but their effectiveness is increasingly limited by cytochrome P450-mediated metabolic resistance. Here, we investigated the role of CYP4C64 in nitenpyram (NIT) detoxification in Bemisia tabaci through expression analyses, bioassays, heterologous expression, and in vitro metabolism assays. CYP4C64 was functionally expressed in insect cells using a Bac-to-Bac baculovirus system. UPLC-MS analysis showed that CYP4C64 catalyzed the conversion of NIT to IM-1-4, the major metabolite identified under the experimental conditions,

Gel Carriers for Plant Extracts and Synthetic Pesticides in Rodent and Arthropod Pest Control: An Overview.

Insecticides and rodenticides form the basis of integrated pest management systems worldwide. As pest resistance continues to increase and entire groups of chemical active ingredients are restricted or banned, manufacturers are looking for new options for more effective formulations and safer application methods for the remaining pesticide ingredients. In addition to new technological adaptations of mainstream formulations in the form of sprays, fumigants, and dusts, the use of gel formulations is becoming increasingly explored and employed. This article summarizes information on the current a

Development and application of a PCR-RFLP assay revealing widespread distribution of the pyrethroid resistance-associated VGSC V1016G mutation in Aedes albopictus from Guangyuan City, Sichuan Province of China.

BACKGROUND: Aedes albopictus is a primary vector of multiple arboviruses, including dengue, chikungunya, yellow fever, and Zika virus. Its control relies heavily on pyrethroid insecticides. The V1016G mutation in the voltage-gated sodium channel (VGSC) is a well-documented mechanism conferring pyrethroid resistance in Ae. albopictus, which directly challenges the efficacy of pyrethroid-based control. Understanding of the status of insecticide resistance will offer insights to inform evidence-based vector management. However, current phenotypic monitoring is laborious and time-consuming, highli…

Biocontrol of mosquito vectors through herbal-derived silver nanoparticles: prospects and challenges.

Mosquitoes spread several life-threatening diseases such as malaria, filaria, dengue, Japanese encephalitis, West Nile fever, chikungunya, and yellow fever and are associated with millions of deaths every year across the world. However, insecticides of synthetic origin are conventionally used for controlling various vector-borne diseases but they have various associated drawbacks like impact on non-targeted species, negative effects on the environment, and development of resistance in vector species by alteration of the target site. Plant extracts, phytochemicals, and their nanoformulations ca

The Cry4B toxin of Bacillus thuringiensis subsp. israelensis kills Permethrin-resistant Anopheles gambiae, the principal vector of malaria.

Resurgence of malaria has been attributed, in part, to the development of resistance by Anopheles gambiae, a principal vector of the disease, to various insecticidal compounds such as Permethrin. Permethrin, a neurotoxicant, is widely used to impregnate mosquito nets. An alternative strategy to control mosquitoes is the use of Bacillus thuringiensis subsp. israelensis (Bti) because there is no observable resistance in the field to the bacterium. Bti kills mosquitoes by targeting cadherin molecules residing in the midgut epithelium of larvae of the insect. Cry proteins (Cry4A, Cry4B, Cry10A and

Some emerging issues on the malaria problem in Kenya.

Malaria in Kenya has been characterised by marked variability in its epidemiology, partly reflecting the obstacles and progress made in the control strategies. The impact of anti-vector activities in the 1970s and before have been observed for variable lengths of time afterwards. Malaria has re-emerged in areas previously with little or no transmission. The recovery of infective Anopheles gambiae vector in higher altitudes affirms the potential for transmission in areas where epidemics have been known to occur. Morbidity and mortality patterns in the otherwise endemic lowlands have become incr

These efforts are crucial since a resurgence of malaria due to drug and/or insecticide resistance, program cuts, lack of political support and donor fatigue could set back malaria control success in the sub-region and threaten malaria control and elimination if resistance spreads to other regions.

The ecdysone receptor regulates several key physiological factors in Anopheles funestus.

BACKGROUND: Malaria is a devastating disease, transmitted by female Anopheles mosquitoes infected with Plasmodium parasites. Current insecticide-based strategies exist to control the spread of malaria by targeting vectors. However, the increase in insecticide resistance in vector populations hinder the efficacy of these methods. It is, therefore, essential to develop novel vector control methods that efficiently target transmission reducing factors such as vector density and competence. A possible vector control candidate gene, the ecdysone receptor, regulates longevity, reproduction, immunity…

The antennal transcriptome of Triatoma infestans reveals substantial expression changes triggered by a blood meal.

BACKGROUND: Triatoma infestans is the main vector of Chagas disease in the Americas, currently transmitting it in Argentina, Paraguay, and Bolivia. Many T. infestans populations present insecticide resistance, reducing the efficiency of control campaigns. Alternative vector control methods are needed, and molecular targets mediating fundamental physiological processes can be a promising option to manipulate kissing bug behavior. Therefore, it is necessary to characterize the main sensory targets, as well as to determine whether they are modulated by physiological factors. In order to identify …

Tick prevention at a crossroad: new and renewed solutions.

Ticks have major economic impact through diseases they transmit, direct losses due to their detrimental effect and the efforts invested in prevention measures directed against them. Chemical acaricides represent the main line of anti-tick defense in both humans and domestic animals, but increasing concerns regarding development of acaricide resistance, especially in the cattle tick Rhipicephalus microplus, and environmental safety issues indicate the need for other, less aggressive but equally efficient methods. This paper aims to evaluate the potential, the scientific and economical limitatio

The genetic architecture of a complex trait: Resistance to multiple toxins produced by Bacillus thuringiensis israelensis in the dengue and yellow fever vector, the mosquito Aedes aegypti.

The bacterial insecticide Bacillus thuringiensis subsp. israelensis (Bti) is an increasingly popular alternative to chemical insecticides for controlling mosquito populations. Because Bti toxicity relies on the action of four main toxins, resistance to Bti is very likely a complex phenotype involving several genes simultaneously. Dissecting the underlying genetic basis thus requires associating a quantitative measure of resistance to genetic variation at many loci in a segregating population. Here, we undertake this task using the dengue and yellow fever vector, the mosquito Aedes aegypti, as
