A grape seed extract increases active glucagon-like peptide-1 levels after an oral glucose load in rats.

We have previously reported that procyanidins, a class of flavonoids, improve glycemia and exert an incretin-like effect, which was linked to their proven inhibitory effect on the dipeptidyl-peptidase 4 (DPP4) activity. However, their actual effect on incretin levels has not been reported yet. Therefore, in the present study we have evaluated whether a grape seed extract enriched in procyanidins (GSPE) modulates plasma incretin levels and attempted to determine the mechanisms involved. An acute GSPE treatment in healthy Wistar female rats prior to an oral glucose load induced an increase in pl

Toll-like receptor 4-mediated regulation of spontaneous Helicobacter-dependent colitis in IL-10-deficient mice.

BACKGROUND & AIMS: The commensal microbiota is believed to have an important role in regulating immune responsiveness and preventing intestinal inflammation. Intestinal microbes produce signals that regulate inflammation via Toll-like receptor (TLR) signaling, but the mechanisms of this process are poorly understood. We investigated the role of the anti-inflammatory cytokine interleukin (IL)-10 in this signaling pathway using a mouse model of colitis. METHODS: Clinical, histopathologic, and functional parameters of intestinal inflammation were evaluated in TLR4(-/-), IL-10(-/-), and TLR4(-/-)

Relative allergenicity of cow's milk and cow's milk-based formulas in an animal model.

Human infants fed cow's milk or cow's milk-based formula may become intolerant to milk proteins partially because of allergic reactions. However, the relative allergenicity of these diets has not been clearly determined at the intestinal level. In this study, the allergenicity of cow's milk and milk-based formula was evaluated by examining intestinal anaphylaxis caused by challenge with a milk protein fraction, beta-lactoglobulin (beta LG), in guinea pigs orally sensitized to these diets. Colonic segments were removed and challenged with beta LG. An antigen-induced, anaphylactically mediated e

Gut-Derived Exosomes Mediate Memory Impairment After Intestinal Ischemia/Reperfusion via Activating Microglia.

Intestinal ischemia/reperfusion is a grave condition with high morbidity and mortality in perioperative and critical care settings and causes multiple organ injuries beyond the intestine, including brain injury. Exosomes act as intercellular communication carriers by the transmission of their cargo to recipient cells. Here, we investigate whether exosomes derived from the intestine contribute to brain injury after intestinal ischemia/reperfusion via interacting with microglia in the brain. Intestinal ischemia/reperfusion was established in male C57/BL mice by clamping the superior mesenteric a

Antihyperglycemic potential of incretins orally delivered via nano and microsystems and subsequent glucoregulatory effects.

Type 2 diabetes mellitus (T2DM) is mainly associated with impaired insulin secretion by the pancreatic β-cells, insulin resistance and elevated hepatic gluconeogenesis. Incretin based treatments for T2DM are now widely investigated and used. The incretin based therapies mainly include incretin hormones which are glucose-dependent insulinotropic peptides (GIP) and glucagon like peptide-1 (GLP-1) released from the endocrinal cells in the small intestine in response to food intake. The main function of GLP-1 is to induce insulin secretion and suppress glucagon secretion. This review describes the

Mineral absorption and secretion patterns in the alimentary tract of the roe deer (Capreolus capreolus).

Concentrations of macrominerals; Na, K, Ca, Mg, P, and Cl were measured in different sections of the alimentary tract of five roe deer, Capreolus capreolus, kept in captivity and fed a diet of grass pellets and oats. By means of the non-absorbed marker-slaughter technique (using 51CrEDTA as marker), sites of secretion and absorption of minerals in the alimentary tract were determined. Large amounts of P, Na and K were secreted into the rumen, whereas Cl was secreted into the abomasum. The larger amounts of these minerals were absorbed from the distal small intestine and caecum/proximal colon.

Substance K and salivary secretion in the rat.

Intravenous injections of substance K (SK), a novel member of the family of tachykinins, evoked secretion from the three major salivary glands of the rat in the presence of muscarinic, alpha-adrenergic and beta-adrenergic receptor blockade; the submaxillary glands contributed most and the sublingual glands least to the total volume secreted. SK was less potent than substance P (SP) in evoking fluid and amylase secretion. However, the amylase concentration in parotid saliva evoked by SK was twice that evoked by SP, a finding which indicates that in the glands there are more than just one type o

The role of gastric secretion in post-diverted pancreatic hypersecretion in conscious rats.

1. In rats prepared with chronic external pancreatic fistulae, gastric fistulae and choledocho-enterostomy, the volume and protein secretion of pancreatic juice was measured. 2. The pancreatic secretion of water and protein after juice diversion was significantly lower when the gastric fistula was open than when closed. Secretion was abolished after atropine, with the fistula open and in animals with ligated pylori. 3. Intraduodenal HCl significantly raised juice volume and protein in animals with diverted juice when the gastric fistula was open. 4. Soybean trypsin inhibitor produced a signifi

RiboTRAP-seq identifies spatially distinct functions for the anterior and posterior intestine in immune and metabolic regulation in Caenorhabditis elegans.

The intestine integrates food-derived cues to coordinate organismal physiology, yet the molecular specialization of discrete regions along the intestinal epithelium remains unclear. Here, we generate cell type-specific translatomes of the Caenorhabditis elegans intestine during fasting and refeeding using discrete promoters for the anterior quartet of intestinal cells (INT1) and the remaining eight pairs of intestinal cells (INT2-9). We found that the anterior-most INT1 cells are a translationally distinct intestinal subcompartment that is particularly enriched for immune- and stress response

Metabolic sensor AMPK licenses intestinal CD103+ DCs to induce Treg responses.

Intestinal dendritic cells (DCs) play a central role in maintaining gut tolerance through priming of peripheral regulatory T cells (pTreg cells). DC tolerogenicity has been linked to catabolic metabolism, but the role of AMP-activated kinase (AMPK), a key regulator of catabolic metabolism, in regulating intestinal tolerance remains unclear. We found high AMPK activation in intestinal DCs, and loss of AMPKα1 in CD11c-expressing cells (CD11cΔAMPKα1) led to reduced frequencies of intestinal RALDH+ CD103+ cDC2s. This was associated with reduced pTreg cell induction and consequently increased type

Histology and mucin histochemistry of the digestive tract of yellow catfish, Pelteobagrus fulvidraco.

The histology and characteristics of mucins secreted by epithelial mucous cells of the digestive tract in yellow catfish, Pelteobagrus fulvidraco were investigated using light microscope and transmission electron microscope. The digestive tract was divided into a pharynx, oesophagus, U-shaped stomach (with a cardiac, fundic and pyloric part) and intestine, composed of anterior intestine, middle intestine and posterior intestine, which consisted of a mucosa (epithelial layer), lamina propria-submucosa, muscularis and serosa. A large number of isolated longitudinal striated muscular bundles were

MyD88-mediated signals induce the bactericidal lectin RegIII gamma and protect mice against intestinal Listeria monocytogenes infection.

Listeria monocytogenes is a food-borne bacterial pathogen that causes systemic infection by traversing the intestinal mucosa. Although MyD88-mediated signals are essential for defense against systemic L. monocytogenes infection, the role of Toll-like receptor and MyD88 signaling in intestinal immunity against this pathogen has not been defined. We show that clearance of L. monocytogenes from the lumen of the distal small intestine is impaired in MyD88(-/-) mice. The distal ileum of wild-type (wt) mice expresses high levels of RegIII gamma, which is a bactericidal lectin that is secreted into t

Galnt3, an enzyme engaged in protein glycosylation modification, is essential for the maintaining of intestinal health in zebrafish.

Intestinal inflammation significantly impairs intestinal function and is closely associated with various health complications. Understanding its molecular mechanisms is crucial for developing effective therapeutic strategies. Galnt3, a member of the polypeptide N-acetylgalactosaminyltransferase family, participates in multiple biological processes, yet its specific role in intestinal inflammation remains poorly understood. In this study, we observed a significant downregulation of zebrafish galnt3 in response to GCRV virus or poly(I:C) infection. Galnt3 knockout (galnt3-/-) zebrafish exhibited

Transcriptome analysis reveals intestinal gene expression and metabolic pathways in yaks.

The yak, a ruminant species native to high-altitude plateaus, serves as a vulnerable model for studying adaptation to extreme environments. Exposed to hypoxia, low temperatures, and strong ultraviolet radiation, yaks have evolved unique physiological mechanisms to thrive under such conditions. Among its organs, the intestine is a crucial metabolic organ and plays a key role in environmental adaptation. Therefore, this study systematically compared the transcriptional regulatory mechanisms of the small and large intestine between yaks and cattle, thereby revealing the metabolic mechanisms under

Crosstalks Between Gut Microbiota and Vibrio Cholerae.

Vibrio cholerae, the causative agent of cholera, could proliferate in aquatic environment and infect humans through contaminated food and water. Enormous microorganisms residing in human gastrointestinal tract establish a special microecological system, which immediately responds to the invasion of V. cholerae, through "colonization resistance" mechanisms, such as antimicrobial peptide production, nutrients competition, and intestinal barrier maintenances. Meanwhile, V. cholerae could quickly sense those signals and modulate the expression of relevant genes to circumvent those stresses during

Diversion of bile and pancreatic secretion in the rat and its effect on cysteamine-induced duodenal and peptic ulcer development under maximal acid secretion.

In this investigation we assess whether diversion of bile and pancreatic secretion protects rats from ulceration under maximal acid secretion as it does in cysteamine-induced ulcers. The rats were divided into a cysteamine-group (Cy) comprising 16 rats and a secretagogue group (PC) with 18 white male rats. Half of the animals of each group were submitted to the diversion-operation (division of the duodenum distally from the pylorus, jejuno-duodenostomy and entero-y-anastomosis), Cy2 and PC2 respectively. The other animals were not operated (Cy1 and PC1). In the cysteamine group the rats were g

Measurement of duodeno-gastric reflux in dogs.

For the elaboration of a formula estimating the reflux volume of duodenal secretion into the stomach, gastric and duodenal secretion have been studied during pentagastrin and secretin stimulation. In the stimulated gastric secretion the concentration of sodium varied with the secretion rate, whereas the concentration of chloride was nearly constant. The concentration of sodium in duodenal juice was constant, but the chloride concentration dropped significantly during secretin stimulation. Secretin induces duodenal reflux. When duodenal reflux occurs, a substantial amount of the sodium in gastr

CEA levels in fluids bathing gastrointestinal tumors.

A controlled prospective study was undertaken to determine if fluids which bathe malignancies may contain carcinoembryonic antigen (CEA) earlier in the course of gastrointestinal cancer than does plasma of the same patient and may offer a better means for diagnosis. CEA titers were normal (less than 2.5 ng per ml) in the plasma of 42 healthy volunteers. Normal CEA levels were also found in the plasma and in the colonic mucus of 14, the gastric juice of 18, duodenal drainage of 10, and bile of 11 normal control subjects. The colonic mucus of 3 patients with ulcerative colitis, gastric secretion
