Eyes Wide Open: Assessing Early Visual Behavior in Zebrafish Larvae.

Early diagnosis is critical for the effective management of neurodegenerative disorders, and retinal alterations have emerged as promising early biomarkers due to the retina's close developmental and functional link to the brain. The zebrafish (Danio rerio), with its rapid development, transparent embryos, and evolutionarily conserved visual system, represents a powerful and versatile model for studying retinal degeneration. This review discusses a range of behavioral assays-including visual adaptation, motion detection, and color discrimination-that are employed to evaluate retinal function i

Role of sex hormones in the reactivation of Toxocara canis larvae in pregnant bitches.

Based on an exhaustive review, a theory was proposed with an immunoendocrine perspective on the reactivation of dormant Toxocara canis larvae in pregnant bitches and their transmission to fetuses through the placenta, or milk and colostrum to puppies. A historical review was carried out on the reactivation of dormant larvae of T. canis in pregnant bitches which suggested that larval reactivation of T. canis is not solely the effect of a single hormone (prolactin) but is the result of a series of events triggered by progesterone, prolactin, and estrogens. In the first third of gestation, proges…

Emerging Resistance in Anopheles stephensi Larvae to the Larvicide Temephos in Chennai, India.

This study aimed to evaluate the susceptibility of immature Anopheles stephensi to temephos formulation, a primary antilarval intervention currently in operational use. The assessment was conducted against both the WHO-recommended discriminatory dosage with temephos (0.25 parts per million [ppm]) and the operational field dosage with temephos 50% emulsifiable concentrate (EC; 1 ppm). F1 generation immature An. stephensi from two areas of Chennai, India was used for the study. The larvae (F1) from both areas were fully susceptible to the WHO discriminatory dosage. Larval susceptibility tests wi…

Molecular basis of quinine-induced feeding suppression in Hyphantria cunea larvae.

BACKGROUND: Feeding behavior in herbivorous insects is profoundly influenced by chemical cues present in their environment, with some compou … gely unexplored. RESULTS: We demonstrate that quinine significantly suppresses feeding in H. cunea larvae. Using calcium imaging and intracellular calcium concentration measurements via a microplate reader, we identified a bitter gustatory receptor, HcGr148, that specifically responds to quinine. Quantitative real-time polymerase chain reaction analysi …

Myclobutanil induces cardiovascular toxicity via thyroid disruption in zebrafish larvae.

Myclobutanil (MYC), a widely used triazole fungicide, is extensively employed in agriculture. MYC residues are frequently found in ecosystems and living organisms. Nevertheless, research on its potential adverse effects on embryonic development remains limited. In this study, zebrafish embryos were exposed to MYC (0, 0.5 and 1 mg/L) between 4 and 96 h post fertilization (hpf) to evaluate the cardiovascular toxicity and molecular mechanisms. The results demonstrated that MYC decreased the survival and hatching rate, and heart rate, but increased the malformation rate and spontaneous movement. M

Cellular Uptake and Tissue Retention of Microplastics in Black Soldier Fly Larvae.

Microplastic pollution is a pressing global concern, yet its immunotoxicological impacts on invertebrates remain poorly understood. The Black Soldier Fly (Hermetia illucens) larva has gained attention for its role in waste management and potential bioremediation, making it essential to evaluate its interactions with microplastics. In this study, fluorescent carboxylate-modified polystyrene microbeads were directly injected into the hemocoel of larvae to bypass gut-associated variables and investigate systemic immune responses. Experimental groups were analyzed at multiple time points (1 h, 6 h…

Anesthesia and humane euthanasia methods for king worm larvae (Zophobas morio).

A growing spotlight on insect welfare as research subjects, fodder for vertebrate animals, and patients for veterinarians highlights the need to establish humane euthanasia methods for these species. An insect of increasing commercial importance is the king worm (Zophobas morio), a type of darkling beetle larvae. The initial objective of this study was to determine if prolonged exposure to isoflurane would lengthen anesthetic recovery and lead to euthanasia. Larvae were exposed to isoflurane for 10 min, 40 min, 70 min, 190 min, 13 h, and 24 h (n = 10 for each treatment duration group), and tim…

Suppression of Contraction Raises Calcium Ion Levels in the Heart of Zebrafish Larvae.

Zebrafish larvae have emerged as a valuable model for studying heart physiology and pathophysiology, as well as for drug discovery, in part thanks to its transparency, which simplifies microscopy. However, in fluorescence-based optical mapping, the beating of the heart results in motion artifacts. Two approaches have been employed to eliminate heart motion during calcium or voltage mapping in zebrafish larvae: the knockdown of cardiac troponin T2A and the use of myosin inhibitors. However, these methods disrupt the mechano-electric and mechano-mechanic coupling mechanisms. We have used ratiome…

Anaphylaxis after consumption of wasp larvae in Reunion Island: a case report.

The practice of entomophagy is common in Asia, Africa, and South America and is now spreading to Europe and the United States. Entomophagy is not without risk since humans can develop allergic reactions to the ingested insects. Here we describe a case of anaphylaxis after consumption of Polistes olivaceus larvae in a 23-y-old man living in Reunion Island, a French overseas department where wasps and other insects are occasionally consumed as part of local traditions. The patient developed diffuse pruritus with facial edema, nausea, and vomiting 15 min after ingesting pan-fried wasp larvae duri…

The cluster digging behavior of larvae confers trophic benefits to fitness in insects.

Collective behaviors efficiently impart benefits to a diversity of species ranging from bacteria to humans. Fly larvae tend to cluster and form coordinated digging groups under crowded conditions, yet understanding the rules governing this behavior is in its infancy. We primarily took advantage of the Drosophila model to investigate cooperative foraging behavior. Here, we report that Drosophila-related species and the black soldier fly have evolved a conserved strategy of cluster digging in food foraging. Subsequently, we investigated relative factors, including larval stage, population densit…

A pH-powered mechanochemical engine regulates the buoyancy of Chaoborus midge larvae.

The freshwater aquatic larvae of the Chaoborus midge are the world's only truly planktonic insects, regulating their buoyancy using two pairs of internal air-filled sacs, one in the thorax and the other in the seventh abdominal segment. In 1911, August Krogh demonstrated the larvae's ability to control their buoyancy by exposing them to an increase in hydrostatic pressure.1 However, how these insects control the volume of their air-sacs has remained a mystery. Gas is not secreted into the air-sacs, as the luminal gas composition is always the same as that dissolved in the surrounding water.1,2…

Propranolol hydrochloride induces neurodevelopmental toxicity and locomotor disorders in zebrafish larvae.

Propranolol hydrochloride is the first-line drug for the clinical treatment of hypertension, arrhythmia, and other diseases. However, with the increasing use of this drug, its safety and environmental health have received more and more attention. In this study, aquatic vertebrate zebrafish were used as a model to study the toxic effects and mechanisms of propranolol hydrochloride. It was revealed that zebrafish larvae exposed to propranolol hydrochloride showed aberrant head nerve development and locomotor disorders. Additionally, exposure to propranolol hydrochloride could induce oxidative st…

Developmental disorders caused by cefixime in the otic vesicles of zebrafish embryos or larvae.

To explore the developmental toxicity of cefixime (CE) in the developmental disorder and toxicity mechanism of CE on otic vesicles, zebrafish embryos were used as an animal model. The results showed that CE increased mortality in a dose-dependent manner and decreased the hatching rate of zebrafish larva at 96 hpf. Interestingly, CE significantly reduced the area of the saccule and utricle, as well as the area of otic vesicles in zebrafish larvae (p < 0.001). Fibroblast growth factor 8a (Fgf8a) inhibitors and bone morphogenetic protein (BMP) inhibitors caused similar morphological changes. CE d…

Feeding preference of insect larvae to waste electrical and electronic equipment plastics.

Waste electrical and electronic equipment (WEEE) plastics not only pollute the environment, but are challenging to treat in an environmentally friendly manner. Biodegradation by insect larvae is potentially an eco-friendly method to treat WEEE plastics, but information about the feeding preference of insect larvae to WEEE plastics is lacking. In this study, a total of nine WEEE and pristine plastics were chosen to feed larvae of the following two insect species, i.e. Galleria mellonella and Tenebrio molitor. G. mellonella larvae significantly favor corresponding pristine plastics compared to t…

Exploration of zebrafish larvae as an alternative whole-animal model for nephrotoxicity testing.

Due to an increasing demand for testing of new and existing chemicals and legal restrictions for the use of animals, there is a strong need for alternative approaches to assess systemic toxicity. Embryonic and larval zebrafish (Danio rerio) are increasingly recognized as a promising alternative whole-animal model that may be able to overcome limitations of cell-based in vitro assays and bridge the gap between high-throughput in vitro screening and low-throughput in vivo tests in animals. Despite the relatively simple anatomical structure of the zebrafish larval kidney (pronephros) - composed o…
