Bisphenol S exacerbates high-fat diet-induced obesity and insulin resistance in offspring: insights from a multi-tissue systemic lipidomic and proteomic atlas.

Bisphenol S (BPS) is a widely used substitute for bisphenol A in food-contact materials; but its role in perinatal programming of obesity and insulin resistance remains unclear. Here, we constructed a multi-tissue lipidomic-proteomic atlas to examine the effects of perinatal BPS exposure in high-fat diet-fed offspring mice. Non-targeted lipidomics revealed extensive disturbances in lipid metabolism across the heart, liver, pancreas, white adipose tissue (WAT), and brown adipose tissue (BAT), identifying BAT as the primary target. Males displayed extensive lipidomic alterations, whereas females…

Improving the intestinal lipidome coverage in a gnotobiotic mouse model using UHPLC-MS-based approach through optimization of mobile phase modifiers and column selection.

Lipidomics is focusing on the screening of lipid species in complex mixtures using mass spectrometry-based approaches. In this work, we aim to enhance the intestinal lipidome coverage within the Oligo-Mouse-Microbiota (OMM12) colonized mouse model by testing eight mobile phase conditions on five reversed-phase columns. Our selected mobile phase modifiers included two ammonium salts, two concentrations, and the addition of respective acids at 0.1 %. We compared two columns with hybrid surface technology, two with ethylene bridged hybrid technology and one with core-shell particles. Best perform…

Circulating Metabolomic and Lipidomic Signatures Identify a Type 2 Diabetes Risk Profile in Low-Birth-Weight Men with Non-Alcoholic Fatty Liver Disease.

The extent to which increased liver fat content influences differences in circulating metabolites and/or lipids between low-birth-weight (LBW) individuals, at increased risk of type 2 diabetes (T2D), and normal-birth-weight (NBW) controls is unknown. The objective of the study was to perform untargeted serum metabolomics and lipidomics analyses in 26 healthy, non-obese early-middle-aged LBW men, including five men with screen-detected and previously unrecognized non-alcoholic fatty liver disease (NAFLD), compared with 22 age- and BMI-matched NBW men (controls). While four metabolites (out of 6…

Improved Peripheral and Hepatic Insulin Sensitivity after Lifestyle Interventions in Type 2 Diabetes Is Associated with Specific Metabolomic and Lipidomic Signatures in Skeletal Muscle and Plasma.

Lifestyle interventions with weight loss can improve insulin sensitivity in type 2 diabetes (T2D), but mechanisms are unclear. We explored circulating and skeletal muscle metabolite signatures of altered peripheral (pIS) and hepatic insulin sensitivity (hIS) in overweight and obese T2D individuals that were randomly assigned a 12-week Paleolithic-type diet with (diet-ex, n = 13) or without (diet, n = 13) supervised exercise. Baseline and post-intervention measures included: mass spectrometry-based metabolomics and lipidomics of skeletal muscle and plasma; pIS and hIS; ectopic lipid deposits in…

Vascular Lipidomic Profiling of Potential Endogenous Fatty Acid PPAR Ligands Reveals the Coronary Artery as Major Producer of CYP450-Derived Epoxy Fatty Acids.

A number of oxylipins have been described as endogenous PPAR ligands. The very short biological half-lives of oxylipins suggest roles as autocrine or paracrine signaling molecules. While coronary arterial atherosclerosis is the root of myocardial infarction, aortic atherosclerotic plaque formation is a common readout of in vivo atherosclerosis studies in mice. Improved understanding of the compartmentalized sources of oxylipin PPAR ligands will increase our knowledge of the roles of PPAR signaling in diverse vascular tissues. Here, we performed a targeted lipidomic analysis of ex vivo-generate

Daily mycoprotein consumption for 1 week does not affect insulin sensitivity or glycaemic control but modulates the plasma lipidome in healthy adults: a randomised controlled trial.

Mycoprotein consumption has been shown to improve acute postprandial glycaemic control and decrease circulating cholesterol concentrations. We investigated the impact of incorporating mycoprotein into the diet on insulin sensitivity (IS), glycaemic control and plasma lipoprotein composition. Twenty healthy adults participated in a randomised, parallel-group trial in which they consumed a 7 d fully controlled diet where lunch and dinner contained either meat/fish (control group, CON) or mycoprotein (MYC) as the primary source of dietary protein. Oral glucose tolerance tests were performed pre-

Phosphatidylserine-Related Lipid Alterations Are Associated with the Shift of Oily Skin Towards an Acne-Associated Unhealthy State in Young Women: A Skin Surface Lipidomic Study.

BACKGROUND: Oily skin is not synonymous with acne. However, the skin surface lipid differences between healthy and acne-associated states within an oily-skin population remain insufficiently characterised. OBJECTIVES: To characterise compositional and structural differences in skin surface lipids (SSLs) between healthy and acne-associated unhealthy states among young women with oily skin. METHODS: Sixty-six women aged 18-24 years with oily skin were enrolled, including 33 with healthy oily skin and 33 with mild-to-moderate acne-associated unhealthy oily skin. SSLs were analysed by ultra-perf

Mapping of Lipidome Profile in Drug-Resistant Clinical Isolates of Mycobacterium tuberculosis Through Quali-Quantitative Liquid Chromatography-Mass Spectrometry Identifies Signature Lipids.

The complex lipid composition of Mycobacterium tuberculosis (MTB) plays a pivotal role in pathogenesis, immune evasion, and antimicrobial resistance. The continued surge in drug-resistant strains underscores the need for a deeper understanding of the molecular architecture underlying MTB pathogenesis and drug resistance. In this study, a comparative lipidomics approach was applied to explore the resistance-associated lipid alterations in drug-sensitive (DS), drug-resistant (DR), multidrug-resistant (MDR), and pre-extensively drug-resistant (PXDR) MTB clinical isolates. Lipids derived from whol…

Effects of manual acupuncture on fatty acid metabolism in a rat model of diarrhea-predominant irritable bowel syndrome: a multi-tissue metabolomic and plasma lipidomic investigation.

OBJECTIVE: Diarrhea-predominant irritable bowel syndrome (IBS-D) is a subtype of IBS. Many patients are dissatisfied with the efficacy of drug treatment and seek complementary and alternative medical therapies, especially acupuncture treatment. In this study, we evaluated the effect of manual acupuncture (MA) on IBS-D using a rat model and investigated the underlying mechanism of action by analyzing the changes in untargeted metabolomics of plasma/liver/colon and targeted plasma lipidomics. METHODS: Thirty male Sprague-Dawley rats were divided into Control, IBS-D and MA groups (n = 10 each). …

Lipidomic profiling of serum and liver tissue reveals hepatoprotective mechanism of taxifolin in rats with CCl4-induced subacute hepatic injury based on LC-MS/MS.

Currently, the hepatoprotective activity of taxifolin, a flavonoid isolated from Pseudotsuga taxifolia, has been reported in many animal models. However, whether the protective effect of taxifolin on the liver is related to its effect on lipidomics is unclear. Based on the significant therapeutic effect of taxifolin on CCl4 induced subacute hepatic injury, we observed the intervention of taxifolin by lipidomics. The results demonstrate that taxifolin can effectively reverse the damage caused by CCl4, which including hepatocyte vacuolization and necrosis. Lipomic profiling based on liquid chrom…

Direct Water-Assisted Laser Desorption/Ionization Mass Spectrometry Lipidomic Analysis and Classification of Formalin-Fixed Paraffin-Embedded Sarcoma Tissues without Dewaxing.

BACKGROUND: Formalin-fixed paraffin-embedded (FFPE) tissue has been the gold standard for routine pathology for general and cancer postoperative diagnostics. Despite robust histopathology, immunohistochemistry, and molecular methods, accurate diagnosis remains difficult for certain cases. Overall, the entire process can be time consuming, labor intensive, and does not reach over 90% diagnostic sensitivity and specificity. There is a growing need in onco-pathology for adjunct novel rapid, accurate, reliable, diagnostically sensitive, and specific methods for high-throughput biomolecular identif

A Lipidomics Approach to Assess the Association Between Plasma Sphingolipids and Verbal Memory Performance in Coronary Artery Disease Patients Undertaking Cardiac Rehabilitation: A C18:0 Signature for Cognitive Response to Exercise.

BACKGROUND: Early subtle deficits in verbal memory, which may indicate early neural risk, are common in patients with coronary artery disease (CAD). While exercise can improve cognition, cognitive response to exercise is heterogeneous. Sphingolipids have been associated with the development and progression of CAD, and impairments in sphingolipid metabolism may play roles in neurodegeneration and in the neural adaptation response to exercise. OBJECTIVE: In this study, change in plasma concentrations of sphingolipids was assessed in relation to change in verbal memory performance and in other c

Transcriptomic and Lipidomic Analyses Reveal a Novel Antiapoptotic Mechanism of Peptide IRW in the Arteries of Spontaneously Hypertensive Rats through the Reduction of Ceramide Levels and Arachidonic Acid Release.

IRW, an antihypertensive peptide derived from ovotransferrin, has been shown to lower blood pressure in spontaneously hypertensive rats (SHRs) by upregulating angiotensin-converting enzyme 2 (ACE2). ACE2 is cardioprotective and a well-documented inhibitor of apoptosis. This study aims to investigate the anti-apoptotic effects of IRW and its underlying mechanism in SHRs' vasculature. IRW was orally administered to SHRs at 15 mg/kg body weight. Transcriptomic analysis of mesenteric arteries revealed enrichment of genes involved in apoptosis suppression by IRW; proteins associated with pro-apopto

Lipidome of Endemic Fish Comephorus dybowskii (Scorpaeniformes, Comephoridae) in Lake Baikal Plays an Essential Role in Maintaining Organism Homeostasis Due to Biomembrane Modifications and Metabolic Consistency.

The lipid and fatty acid composition of the little Baikal oilfish (Comephorus dybowskii Korotneff, 1905) was studied to identify the features of its biochemical adaptation to the environmental conditions of Lake Baikal. Multivariate analysis of lipid classes and FA profiles revealed reproducible clustering patterns in both muscle tissue and whole-body samples, indicating the presence of distinct physiological states differing in membrane organization and lipid metabolism. Lipid-class variability was primarily associated with sphingomyelin, cholesterol esters, phosphatidylinositol, and lysophos
