A Multi-Component High-Fibre Meal Replacement Containing Pyrus bretschneideri Rehd. Pomace Alleviates Diet-Induced Obesity by Modulating the Gut Microbiota, Bile Acid Metabolism and Hepatic AMPK Signalling.

This study employed an integrated multi-omics approach to investigate the regulatory effects and molecular mechanisms of a multi-component, high-dietary-fibre meal replacement powder containing Pyrus bretschneideri Rehd. pomace (PDMR) in high-fat-diet-induced obese mice. The results showed that PDMR significantly reduced body weight gain, Lee's index, serum lipid and inflammatory cytokine levels, and improved adipocyte morphology. Mechanistically, PDMR activated the AMPK signalling pathway, suppressed lipogenesis via the ACC-SREBP1c-FAS axis, and promoted fatty acid β-oxidation and thermogenes…

Adult Stem Cell-Derived Intestinal Organoids as In Vitro Models of High-Fat Diet-Related Intestinal Diseases.

The dysregulation of intestinal lipid metabolism induced by a high-fat diet (HFD) is associated with metabolic diseases; however, the validi … d budding frequency, lipid droplet accumulation, enriched lipid catabolism pathways, and suppressed lipogenesis, aligning with the in vivo findings. Omics analyses of the top 87 variable genes indicated a strong congruence between FA-treated IOs and intestinal tissues from HFD mice, with control groups clustering distinctly. The differentially expressed genes …

Microalgae and Cyanobacteria in the Obesity Evidence Landscape: A PRISMA-ScR Scoping Review with Mechanistic and Safety Mapping.

Microalgae and cyanobacteria are investigated for compounds that may relate to obesity and metabolic phenotypes, yet the evidence base remai … within the anchored subset, inflammation (NF-κB), AMPK signaling, adipogenesis (PPARγ-related), and lipogenesis were the most frequently mapped nodes. Together, this evidence map clarifies where mechanistic anchoring and safety documentation are sparse and provides a clear starting point for downstream evaluation.

Sinapine Modulates Glycogen and Lipid Synthesis via IRS1-PI3K-AKT-GSK3β-GS Pathway in Insulin-Resistant Models.

In vitro, sinapine treatment improved glucose uptake and glycogen synthesis, while reducing lipogenesis, lipid accumulation, and reactive oxygen species (ROS) levels. RT-qPCR and Western blot analyses confirmed that sinapine increases glycogen synthase activity. In T2DM mice, sinapine improved glucose and lipid metabolism, enhanced insulin sensitivity …

Glycerol 3-phosphate activates lipid metabolism through GATOR2-dependent regulation of mTORC1.

Triglycerides can be formed by fatty acid esterification of glycerol 3-phosphate (G3P) and by de novo lipogenesis (DNL). We identified G3P as a stimulus that activated mTORC1, a nutrient-sensing protein complex that promotes DNL in the liver. We found that the major source of G3P in primary mouse hepatocytes was glycerol kinase (GK), which generates G3P from glycerol. Mice with a liver-specific GK deficiency showed reductions not only in hepatic triglyceride production and storage but also in mTORC1-dependent DNL. Sequentially blocking hepatic pathways for G3P metabolism and analysis of hepato…

Spatially-resolved subtype progression reveals metabolic vulnerabilities in pancreatic ductal adenocarcinoma.

BACKGROUND: Pancreatic ductal adenocarcinoma (PDAC) exhibits profound molecular heterogeneity and poor prognosis, necessitating novel tailor … tabolically ‘hot’, high-grade tumor niches characterized by concurrent enrichment of glycolysis and lipogenesis across both subtypes, nominating them as subtype-agnostic therapeutic targets. Preclinical models demonstrated that despite the basal subtype’s glycolysis dependence, both classical and basal tumors are susceptible to glycolysis inhibition. CONCLUSI …

Benzo[a]pyrene induces non-alcoholic fatty liver disease by exacerbating hepatic senescence and disrupting gut-liver axis in zebrafish.

Polycyclic aromatic hydrocarbons (PAHs), such as benzo[a]pyrene (BaP), are ubiquitous environmental contaminants that may impair liver healt … d hepatic fat accumulation and elevated TG, T-CHO, FFA, and TBA levels, associated with upregulated lipogenesis and suppressed lipid catabolism. BaP also activated the AHR signaling pathway, caused DNA damage, disrupted cell cycle regulation, and exacerbated hepatic senescence, leading to inflammation and mitochondrial dysfunction. Moreover, BaP impaired inte …

Metabolism in Tumor: Mechanisms and Therapeutic Perspectives.

Tumor metabolic reprogramming now extends far beyond the Warburg effect, spanning glycolysis, mitochondrial oxidative phosphorylation (OXPHOS), de novo lipogenesis, cholesterol biosynthesis, and fatty acid oxidation. This network is shaped by cell-intrinsic regulatory layers-transcriptional programs, histone lactylation and other posttranslational modifications, and m6A RNA modification-and by metabolic crosstalk with tumor-associated macrophages (TAMs) and cancer-associated fibroblasts (CAFs) that drives immune evasion and drug resistance. How these diverse inputs converge remains undefined: …

Mitochondria-Targeted Liposomes Boost Thermogenesis for Adipose Tissue Regulation.

Mitochondria act as central organelles regulating energy metabolism and lipogenesis in adipocytes. Mitochondrial dysfunction and oxidative stress are closely implicated in the pathogenesis of obesity. Therefore, targeting adipose mitochondrial function to reduce mitochondrial reactive oxygen (mROS) levels can enhance mitochondrial activity and promote thermogenesis, presenting a promising therapeutic strategy for obesity. In this study, we developed cationic liposomes (AE@PEP-Lip) specifically designed for adipose mitochondrial targeting. These liposomes can effectively accumulate in white adi…

Oleanolic Acid Modulates the Gut-Liver Axis to Alleviate High-Fat Diet-Induced Hepatic Lipid Deposition in Nile Tilapia (Oreochromis niloticus).

This study examined the protective mechanisms of oleanolic acid (OA) against high-fat diet (HFD)-induced hepatic steatosis and intestinal dysbiosis in Nile tilapia. Fish were allocated to four groups: normal diet (ND), HFD, and OA-supplemented HFD (50 and 250 mg/kg). After 42 days, physiological, biochemical, and histological assessments demonstrated that OA markedly reduced hepatic lipid accumulation, mitochondrial injury, and intestinal shortening. Transcriptomic analysis revealed that OA alleviated lipid dysregulation by inhibiting de novo lipogenesis and promoting lipid trafficking and β-o…
