Despite effective first-line regimens, some patients with diffuse large B-cell lymphoma (DLBCL) still experience relapse or resistance, emphasizing the urgent need for innovative treatment approaches. Cytidine triphosphate synthase 1 (CTPS1) is a key regulatory and rate-limiting enzyme for de novo nucleotide synthesis pathway. However, the role of CTPS1 in DLBCL and its potential therapeutic value remain unknown. We found that high levels of CTPS1 were associated with poor prognosis in patients with DLBCL. The single-cell RNA sequencing (scRNA-seq) revealed that phospholipid metabolism and mit

UNLABELLED: Diffuse large B-cell lymphomas (DLBCL) are genetically and phenotypically heterogeneous, making diagnosis and treatment challenging. Current models suggest DLBCLs derive from follicular B cells engaged in adaptive immune responses. By studying cooccurring truncating mutations in SPEN and NOTCH2 in the BN2-DLBCL subtype, our data suggest a previously unrecognized extrafollicular trajectory. Using animal models and human specimens, we find that this cooperative mutational axis supports expansion of putative clonal precursors with features of marginal zone, memory, and a distinct, aut

Diffuse large B-cell lymphoma (DLBCL) most commonly arises in nodal regions but may also present extranodally. We report two cases of large B-cell lymphoma developing in postpneumonectomy cavities more than a decade after surgery for primary lung carcinoma. In both patients, no history of systemic immunodeficiency, organ transplantation, or chronic immunosuppressive therapy was present, and HIV, hepatitis B, and hepatitis C serologies were negative. In Case 1, the lymphoma showed an activated postgerminal center phenotype and was negative for EBER and HHV-8. In Case 2, the tumor was CD20-negat

INTRODUCTION: Diffuse large B-cell lymphoma (DLBCL) is the most common and highly heterogeneous subtype of non-Hodgkin lymphoma, and accurate staging is essential for treatment planning and prognosis. This study explored the value of 2-[18F]fluoro-2-deoxy-D-glucose positron emission tomography/computed tomography (18F-FDG PET/CT) metabolic network connectivity for the clinical staging of DLBCL.
METHODS: Forty-two previously untreated patients (23 males, 19 females; aged 30-82 years) diagnosed as de novo DLBCL, not otherwise specified, were retrospectively analyzed (stage I, n=3; II, n=5; III,

We present the case of primary testicular diffuse large B-cell lymphoma diagnosed at the stage of pectoral skin metastasis in a 40-year-old male patient. He presented with a painless mass in the left scrotum that was complicated by ulcerated gynecomastia. An orchiectomy combined with a pectoral tumorectomy led to this diagnosis. The tumor was of the non-centro-germinative B subtype and classified as pT3NxM1b, Ann-Arbor stage IV E. The patient died while undergoing chemotherapy. This case underscores the poor prognosis associated with this histological type and the value of immunohistochemistry

Adult intussusception is an uncommon clinical entity, representing approximately 5% of all intussusception cases and 1%-5% of intestinal obstructions in adults. (1) In contrast to the pediatric population, adult intussusception is most often secondary to an underlying organic lesion, frequently a neoplasm. Malignant lesions account for nearly 30%-50% of small bowel cases. (2) Among these, primary small intestinal lymphoma is a rare cause, with diffuse large B-cell lymphoma (DLBCL) being particularly uncommon. We herein report a case of chronic intermittent intussusception revealing primary DLB

INTRODUCTION: Primary gastrointestinal lymphoma is a rare form of extranodal non-Hodgkin lymphoma, accounting for approximately 1%-4% of all gastrointestinal malignancies. Among primary gastrointestinal lymphomas, involvement of the small intestine accounts for approximately 20%-30% of cases. CD5-positive diffuse large B-cell lymphoma (DLBCL) is a rare variant, representing approximately 5%-10% of all DLBCL cases and is associated with advanced age, extranodal involvement, and poorer prognosis. We report a rare case of ileocecal CD5-positive DLBCL presenting with acute obstructive symptoms.

RATIONALE: Primary breast diffuse large B-cell lymphoma (PB-DLBCL) is a rare extranodal manifestation of non-Hodgkin lymphoma (NHL) that can closely resemble breast carcinoma clinically and radiologically. This overlap may delay accurate diagnosis and appropriate treatment. We report a case of PB-DLBCL in a postmenopausal woman whose initial multimodal imaging findings were suspicious for breast carcinoma, but whose final diagnosis was established by core needle biopsy with immunohistochemical and molecular evaluation.
PATIENT CONCERNS: A 59-year-old postmenopausal woman presented with a pain

Diffuse large B-cell lymphoma (DLBCL) is characterized by molecular complexity and heterogeneity. The SET oncoprotein is overexpressed in B-cell chronic lymphocytic leukemia and non-Hodgkin lymphoma. Acting as an intrinsic inhibitor of the tumor suppressor protein phosphatase 2A (PP2A), targeting SET has shown anti-cancer activity. In this study, we investigated the molecular mechanism of SET antagonism in DLBCL. Overexpression of SET resulted in decreased phosphatase activities of PP2A and SHP-1, while activating Lyn. Conversely, SET inhibition, either through antagonists or siRNA, led to the

Diffuse large B cell lymphoma (DLBCL) is the most common type of non-Hodgkin lymphoma, and the activated B cell-like subtype (ABC-DLBCL) is associated with particularly poor outcome. Many ABC-DLBCLs harbor gain-of-function mutations that cause inappropriate assembly of the CARMA1-BCL10-MALT1 (CBM) signalosome, a cytoplasmic complex that drives downstream NF-κB signaling. MALT1 is the effector protein of the CBM signalosome such that its recruitment to the signalosome via interaction with BCL10 allows it to exert both protease and scaffolding activities that together synergize in driving NF-κB.

Next Generation Sequencing-based subtyping and interim- and end of treatment positron emission tomography (i/eot-PET) monitoring have high potential for upfront and on-treatment risk assessment of diffuse large B-cell lymphoma patients. We performed Dana Farber Cancer Institute (DFCI) and LymphGen genetic subtyping for the HOVON84 (n = 208, EudraCT-2006-005174-42) and PETAL (n = 204, EudraCT-2006-001641-33) trials retrospectively combined with DFCI genetic data (n = 304). For all R-CHOP treated patients (n = 592), C5/MCD- and C2/A53-subtypes show significantly worse outcome independent of the

Relapsed or refractory diffuse large B-cell lymphoma (R/R DLBCL) has a poor prognosis, particularly in patients ineligible for autologous stem cell transplantation or cellular immunotherapy. This study evaluated the effectiveness of polatuzumab vedotin combined with rituximab and bendamustine (PBR) therapy in comparison to conventional chemotherapy in patients with R/R DLBCL. Of 86 patients with first relapse or primary refractory DLBCL, 32 received PBR in any subsequent line, while 54 received conventional chemotherapy alone. For comparison, 32 baseline-matched patients were selected from the

BACKGROUND: A significant proportion of patients fail to respond adequately to the standard R-CHOP regimen for diffuse large B-cell lymphoma (DLBCL). Rapid proliferation requires energy and the interaction between H+ ions and mitochondria suggests that regulating acid secretion in tumor cells may be a therapeutic strategy for DLBCL. This study constructed a macrophage membrane-coated liposomal formulation (MM-Lipid@Vpz) for the targeted delivery of vonoprazan to combat DLBCL, which enables the evasion of the reticuloendothelial system, ensuring enhanced specificity in tumor targeting.

It is recognized that autoimmune diseases such as primary Sjogren's syndrome and rheumatoid arthritis have an increased risk of developing lymphoma, particularly non-Hodgkin's lymphoma (NHL). The progression of primary biliary cholangitis (PBC), a chronic autoimmune liver disease, to lymphoma on this basis has rarely been reported. This article describes a 67-year-old female patient who developed diffuse large B-cell lymphoma (DLBCL) one year after the diagnosis of PBC. She had experienced a transient improvement after receiving treatment but died as a result of infections and other related pr

Klippel-Feil syndrome (KFS) is a rare congenital disorder characterized by the fusion of cervical vertebrae, with a clinical presentation that can vary widely due to genetic and phenotypic diversity. While KFS can occur as an isolated anomaly, it is often associated with other congenital conditions, such as Sprengel deformity, which may present with or without an omovertebral bone, complicating diagnosis and management. This particular case also involves diffuse large B-cell lymphoma (DLBCL), the most common subtype of non-Hodgkin lymphoma. We hereby present a complex case of a 60-year-old mal

Thrombosis is a common complication in cancer patients, with a substantial impact on morbidity and mortality. Diffuse large B-cell lymphoma (DLBCL) and other aggressive lymphomas carry a high venous thromboembolism (VTE) risk. Extracellular vesicles (EVs) have gained attention in recent research as a new potential biomarker for VTE development. To determine the profile and association of EVs with VTE in patients with DLBCL, we conducted a prospective cohort study on 62 patients diagnosed with DLBCL. A total of 11 patients (17.7%) developed VTE. The concentrations of platelet-derived EVs (PEVs)

The B-cell lymphoma 6 (BCL6) transcription factor plays a key role in the establishment of germinal center (GC) formation. Diffuse large B-cell lymphoma (DLBCL) originates from the GC reaction due to dysregulation of BCL6. Disrupting BCL6 and its corepressors' interaction has become the foundation for rationally designing lymphoma therapies. However, BCL6 inhibitors with good activities in vitro and in vivo are rare, and there are no clinically approved BCL6 inhibitors. In this study, we discovered and developed a novel range of [1,2,4] triazolo[1,5-a] pyrimidine derivatives targeting BCL6/SMR

OBJECTIVES: Diffuse large B-cell lymphoma (DLBCL) constitutes 30-40% of non-Hodgkin lymphoma cases. Despite therapeutic advances, persistence of relapsed cases has been linked to the complex tumor microenvironment (TME) and its interactions with lymphoma cells. In particular, characterising T-cell subsets, including rare cell types, and their interplay with the remaining TME is crucial for unravelling DLBCL pathogenesis and refining therapeutic strategies.
METHODS: Using flow and spectral cytometry with unsupervised analysis, we investigated T-cell subpopulations across DLBCL biopsies and con
