Comprehensive Toxicity Assessment of PEGylated Magnetic Nanoparticles for in vivo applications.

Magnetic nanoparticles (MNPs) represent one of the greatest promises for the development of a new generation of diagnostic agents for magnetic resonance imaging, with improved specificity and safety. Indeed, during the last decade the number of studies published in this field has grown exponentially. However, the clinical translation achieved so far has been very limited. This situation is likely related to the fact that most studies are focused on the in vitro characterization of these new nanomaterials, and very few provide an exhaustive in vivo characterization, where key aspects, such as p

Superparamagnetic iron oxide nanoparticle targeting of adipose tissue-derived stem cells in diabetes-associated erectile dysfunction.

Erectile dysfunction (ED) is a major complication of diabetes, and many diabetic men with ED are refractory to common ED therapies. Adipose tissue-derived stem cells (ADSCs) have been shown to improve erectile function in diabetic animal models. However, inadequate cell homing to damaged sites has limited their efficacy. Therefore, we explored the effect of ADSCs labeled with superparamagnetic iron oxide nanoparticles (SPIONs) on improving the erectile function of streptozotocin-induced diabetic rats with an external magnetic field. We found that SPIONs effectively incorporated into ADSCs and

Investigation of antibacterial properties of novel papain immobilized on tannic acid modified Ag/CuFe2O4 magnetic nanoparticles.

The non-toxic CuFe2O4 magnetic nanoparticles have been solvothermally synthesized, characterized and used as an efficient magnetic precursor for papain immobilization and then investigated antibacterial properties of them. For increasing the antibacterial activity, the silver nanoparticles were doped on CuFe2O4 magnetic nanoparticles. All prepared samples exhibited stronger antibacterial properties against Gram-positive bacteria (Staphyloccocus aureus) than Gram-negative bacteria (Escherichia coli). However, it was determined that the Ag/CuFe2O4 had a more vigorous antibacterial property again

Red blood cell membrane camouflaged magnetic nanoclusters for imaging-guided photothermal therapy.

Along with intrinsic magnetic resonance imaging (MRI) advantages, iron oxide nanomaterials capable of photothermal conversion have been reported very recently and have again raised great interest in their designs among biomedical researchers. However, like other inorganic nanomaterials, high macrophage uptake, short blood retention time and unfavorable biodistributions have strongly hampered their applications in vivo. To solve these problems, a rational design of red blood cell (RBC) membrane camouflaged iron oxide magnetic clusters (MNC@RBCs) is presented in this paper. Our data show that by

Simultaneous determination of ascorbic acid, acetaminophen and codeine based on multi-walled carbon nanotubes modified with magnetic nanoparticles paste electrode.

Based on incorporating ZnCrFeO4 into multi-walled carbon nanotubes paste matrix (MWCNTs/ZnCrFeO4/CPE), a chemically modified electrode was prepared for the simultaneous determination of ascorbic acid (AA), acetaminophen (AC) and codeine (CO). The prepared electrode, MWCNTs/ZnCrFeO4/CPE, was characterized by scanning electron microscopy (SEM) and electrochemical impedance spectroscopy (EIS). The MWCNTs/ZnCrFeO4/CPE showed an efficient electrocatalytic activity for the oxidation of AA, AC, and CO. The separations of the oxidation peak potentials for AA-AC and AC-CO were about 250mV and 630mV, re

Evaluation of magnetic nanoparticles coated by 5-fluorouracil imprinted polymer for controlled drug delivery in mouse breast cancer model.

Nanoparticles (NPs) have been extensively investigated to improve delivery efficiency of therapeutic and diagnostic agents. In this study, magnetic molecularly imprinted polymer (MIP) was synthesized by using polydopamine. Synthesized MIP was used for controlled 5-fluorouracil (5-FU) delivery in a spontaneous model of breast adenocarcinoma in Balb/c mice in the presence of an external magnetic field. Antitumor effectiveness of 5-FU imprinted polymer (5-FU-IP) was evaluated in terms of tumor-growth delay, tumor-doubling time, inhibition ratio, and histopathology. Results showed higher efficacy

The Central-European SentiMag study: sentinel lymph node biopsy with superparamagnetic iron oxide (SPIO) vs. radioisotope.

Sentinel lymph node biopsy (SLNB) is the standard surgical procedure for the axilla in early node-negative breast cancer. To date, the "gold standard" to localize the sentinel lymph node (SLN) is the radiotracer (99m)Tc with or without blue dye. The aim of this study was to evaluate potential equivalency of the new SentiMag(®) technique in comparison to the "gold standard". Within this prospective, multicentric and multinational non-inferiority study including 150 patients (99m)Tc was compared with the magnetic technique, using superparamagnetic iron oxide particles (SPIOs, Sienna+(®)) for loc

Does quantification of USPIO uptake-related signal loss allow differentiation of benign and malignant normal-sized pelvic lymph nodes?

Ultrasmall superparamagnetic iron oxide (USPIO) particles are promising contrast media, especially for molecular and cellular imaging besides lymph node staging owing to their superior NMR efficacy, macrophage uptake and lymphotropic properties. The goal of the present prospective clinical work was to validate quantification of signal decrease on high-resolution T(2)-weighted MR sequences before and 24-36 h after USPIO administration for accurate differentiation between benign and malignant normal-sized pelvic lymph nodes. Fifty-eight patients with bladder or prostate cancer were examined on a

Sialic acid-engineered mesoporous polydopamine dual loaded with ferritin gene and SPIO for achieving endogenous and exogenous synergistic T2-weighted magnetic resonance imaging of HCC.

BACKGROUND: Hepatocellular carcinoma (HCC) is a common malignant tumor with poor prognosis. Magnetic resonance imaging (MRI) is one of the most effective imaging methods for the early diagnosis of HCC. However, the current MR contrast agents are still facing challenges in the early diagnosis of HCC due to their relatively low sensitivity and biosafety. Thus, the development of effective MR agents is highly needed for the early diagnosis of HCC. RESULTS: Herein, we fabricated an HCC-targeted nanocomplexes containing SPIO-loaded mesoporous polydopamine (MPDA@SPIO), sialic acid (SA)-modified pol

Residence Time-Extended Nanoparticles by Magnetic Field Improve the Eradication Efficiency of Helicobacter pylori.

Helicobacter pylori infection is one of the leading causes of several gastroduodenal diseases, such as gastritis, peptic ulcer, and gastric cancer. In fact, H. pylori eradication provides a preventive effect against the incidence of gastric cancer. Amoxicillin is a commonly used antibiotic for H. pylori eradication. However, due to its easy degradation by gastric acid, it is necessary to administer it in a large dosage and to combine it with other antibiotics. This complexity and the strong side effects of H. pylori eradication therapy often lead to treatment failure. In this study, the chitos

Factors regulating macrophage endocytosis of nanoparticles: implications for targeted magnetic resonance plaque imaging.

BACKGROUND: The presence of activated macrophages (Mphi) is an early and consistent marker of the inflammatory nature of atherosclerotic disease. Dextran-coated superparamagnetic iron oxide particles (SPIO) are avidly endocytosed. These particles have a strong effect on magnetic resonance signal and have been proposed as a non-invasive probe for the presence of early non-occlusive atherosclerotic disease. We describe the extent to which endogenous and exogenous factors regulate Mphi uptake of SPIO particles. METHODS AND RESULTS: Cultured murine Mphi-like cells (J744A.1) incubated with SPIO (0

Nanomagnetic System for Rapid Diagnosis of Acute Infection.

Pathogen-activated antibody-secreting cells (ASCs) produce and secrete antigen-specific antibodies. ASCs are detectable in the peripheral blood as early as 3 days after antigen exposure, which makes ASCs a potential biomarker for early disease detection. Here, we present a magnetic capture and detection (MCD) assay for sensitive, on-site detection of ASCs. In this approach, ASCs are enriched through magnetic capture, and secreted antibodies are magnetically detected by a miniaturized nuclear magnetic resonance (μNMR) system. This approach is based entirely on magnetics, which supports high con
