BACKGROUND: The intracerebroventricular injection of beta-amyloid (Aβ) in mice allows the investigation of acute effects on cognitive function and cellular pathology. The aim of this investigation was to further characterize the time course of Aβ-induced cognitive and behavioural changes and to detect potential molecular mechanisms.
METHODS: Cannulas were implanted in the lateral cerebral ventricle. 14days after surgery the mice were injected with Aβ1-42 or phosphate buffered saline (PBS). Starting 2, 4 or 8 (PBS only 4) days after injection we evaluated cognitive and behavioural performance

BACKGROUND: Frontal Lobe Epilepsy (FLE) is the second most frequent type of partial epilepsy and its onset is generally in childhood. Though cognitive and behavioural impairments have been described as co-morbid disorders in epilepsy, their extent in FLE, particularly in children, remains unknown.
AIMS: In this study, we assess cognitive skills and behaviour in a cohort of paediatric FLE patients.
METHODS: We measured the performance of 71 children with cryptogenic FLE on intelligence tests, neuropsychological tests, and behavioural questionnaires. Age-dependent normative values were used fo

OBJECTIVE: Accelerated long-term forgetting (ALF) occurs when newly learned information decays faster than normal over extended delays. It has been recognised most frequently in temporal lobe epilepsy, including Transient Epileptic Amnesia (TEA), but can also be drug-induced. Little is known about the evolution of ALF over time and its impacts upon other memory functions, such as autobiographical memory (ABM). Here we investigate the long-term outcome of ALF and ABM in a group of patients with TEA and a single case of baclofen-induced ALF.
METHODS: Study 1 involved a longitudinal follow-up of

Memory decline is a central cognitive symptom in Parkinson's Disease (PD). While task-fMRI studies link hippocampal activity (AHA) to poorer memory and olfactory performance, this relationship during rest remains understudied. The objectives of this study are to examine differences in resting-state hippocampal networks, explore the occurrence of reduced AHA within these networks, and investigate its impact on memory and olfaction in PD. Thirty-nine PD patients awaiting evaluation for device-aided Parkinson therapy and 46 healthy controls (HC) underwent resting-state fMRI (rs-fMRI). PD patients

BACKGROUND AND PURPOSE: Previous studies have shown the hippocampus and basal ganglia to be highly sensitive to hypoxic-ischemic insult. The authors' aim was to evaluate the long-term effects of perinatal asphyxia (PA) on the hippocampus and caudate nucleus in a group of participants born at term and who met the criteria for hypoxic-ischemic encephalopathy (HIE). Additionally, the authors looked for damage in other brain regions using voxel-based morphometry (VBM).
METHODS: The sample consisted of 13 participants (8 boys and 5 girls) with a mean age at study of 16.23 years (+/- 2.89) with ant

INTRODUCTION: The term amnesic stroke is used to describe a condition in which the dominant symptom is a relatively persistent acute amnesia of a vascular origin. It may appear in cases of lesions in the posterior cerebral artery (PCA), thalamic arteries and anterior choroidal artery, and clinical reports describing this condition are scarce.
CASE REPORT: We describe the case of a 77-year-old male with vascular risk factors who presented sudden onset mnemonic deficit that affected basic activities of daily living, and apathy. Examination of the patient revealed an anterograde episodic amnesia

AIMS: Critically ill old patients sometimes require repeated surgical interventions, and thus it is important to determine the influence of repeated exposure to anesthetics on learning and memory. Sevoflurane, a widely used inhalation anesthetic, has few neurological adverse effects and offers a rapid return to consciousness. But the long-term influence of sevoflurane exposure and the effect of repeated sevoflurane exposure on cognition have rarely been reported, and available studies are contradictory.
MATERIALS AND METHODS: In the present study, the Morris water maze test was employed to in

Memory B cells underpin the capacity of the immune system to confer durable protective immunity by mounting rapid and enhanced secondary antibody responses. Hence, they are central to vaccine efficacy and to limiting long-term tissue damage after reinfection. Despite their importance, the cellular origins, molecular programs, and tissue-specific specialization of memory B cells remain incompletely understood. Here, we review recent advances in memory B cell development and regulation, with a particular focus on tissue-resident memory B cells. We discuss how memory B cells arise from distinct d

Loss-of-function mutations or abnormal expression of the X-linked gene encoding methyl CpG binding protein 2 (MeCP2) cause a spectrum of postnatal neurodevelopmental disorders including Rett syndrome (RTT), nonsyndromic mental retardation, learning disability, and autism. Mice expressing a truncated allele of Mecp2 (Mecp2(308)) reproduce the motor and social behavior abnormalities of RTT; however, it is not known whether learning deficits are present in these animals. We investigated learning and memory, neuronal morphology, and synaptic function in Mecp2(308) mice. Hippocampus-dependent spati

BACKGROUND: Declines in physical function, brain structure, and cognition are common effects of aging in later life. Here we investigate whether changes in physical function and gray matter volumes over a seven-year span predict cognitive performance in older adults.
METHODS: We analyzed five assessments of physical function and MRI-derived gray matter volumes in 219 older adults (mean age 73.7, SD = 2.5). We applied Bayesian linear regression models and model averaging to examine their associations with cognitive performance. We deemed associations meaningful if the inclusion Bayes factor (B

Alzheimer's disease (AD) is a progressive neurodegenerative disorder characterized by cognitive dysfunction and impaired synaptic plasticity. This study evaluated whether chronic L-carnitine (LC) attenuates amyloid-beta 1-42 (Aβ₁-₄₂)-induced spatial learning and memory deficits following intracerebroventricular (ICV) Aβ₁-₄₂ infusion. Forty adult male Wistar rats were randomly assigned to four groups (n = 10/group): Sham + Vehicle, Sham + LC, AD + Vehicle, and AD + LC. Following ICV Aβ₁-₄₂ infusion, LC (100 mg/kg/day, i.p.) or vehicle was administered for 28 days. Spatial learning and memory, h

Emotionally salient experiences are better remembered than events that have little emotional context. Several lines of evidence indicate that the amygdala plays an important role in this emotional enhancement of memory. Visual recognition memory relies on synaptic plasticity in the perirhinal cortex, but little is known about the mechanisms involved in emotional enhancement of this form of memory. The results of the present study, performed in rat brain slices, show for the first time that the amygdala input to the perirhinal cortex undergoes synaptic plasticity. Stimulation in the amygdala re

Numerous studies have shown the deleterious effects of sleep deprivation (SD) on memory. However, SD in various durations may induce different effects. Studies have reported that short-term or acute SD can improve cognitive functions. In addition, streptozotocin (STZ) significantly impairs learning and memory, and induces inflammation and oxidative stress. In this study, we aimed to investigate the effect of two types of SD (short term: 6 h; long term: 24 h) on STZ-induced spatial memory impairment in rats, with respect to the serum level of catalase (CAT), malondialdehyde (MDA), glutathione (

Subconcussive head injuries are connected to both short-term cognitive changes and long-term neurodegeneration. Further study is required to understand what types of subconcussive impacts might prove detrimental to cognition. We studied cadets at the US Air Force Academy engaged in boxing and physical development, measuring head impact motions during exercise with accelerometers. These head impact measures were compared with post-exercise memory performance. Investigators explored multiple techniques for characterizing the magnitude of head impacts. Boxers received more head impacts and achiev

The intracisternal injection of 20 micronl of saline into a rat within an hour after training it in an active avoidance response was found to induce retrograde amnesia. The results obtained by combining this procedure with the intracisternal injection of 2,6 Diaminopurine (DAP), which inhibits RNA synthesis and prevents long-term memory formation when administered at the proper time and in the correct dosage, suggest the existence of a medium term memory (MTM) mechanism. MTM was normally evident up to 75 min after training but was demonstrable up to 210 min when LTM formation was prevented by

Early-life episodic experiences, despite seeming rapidly forgotten, are stored long-term in a latent form. The contribution of this unique hidden memory storage is not understood. Here, we show that contextual memories formed in infant mice are recovered in adulthood following weak behavioral reminders (savings) of the infantile experience. Savings in adults also facilitates new congruent learning but fails to influence other types of hippocampus-dependent learning, and this facilitation is a developmental prerogative. Both infant memory reinstatement and facilitation of new learning in adults

Vaccination has become a widely used method to induce immune protection against microbial pathogens, including viral and bacterial microorganisms. Both humoral and cellular immunity serve a critical role in neutralizing and eliminating these pathogens. An effective vaccine should be able to induce a long-lasting immune memory response. Recent investigations on different subsets of T cells have identified a new subset of T cells using multi-parameter flow cytometry, which possess stem cell-like properties and the ability to mount a rapid immune response upon re-exposure to antigens known as ste

Neural plasticity-the ability to alter a neuronal response to environmental stimuli-is an important factor in learning and memory. Short-term synaptic plasticity and long-term synaptic plasticity, including long-term potentiation and long-term depression, are the most-characterized models of learning and memory at the molecular and cellular level. These processes are often disrupted by neurodegeneration-induced dementias. Alzheimer's disease (AD) accounts for 50% of cases of dementia. Vascular dementia (VaD), Parkinson's disease dementia (PDD), dementia with Lewy bodies (DLB), and frontotempor
