Survival of Vibrio cholerae in nutrient-poor environments is associated with a novel "persister" phenotype.

In response to antibiotic and/or environmental stress, some species of bacteria shift to a "persister" phenotype. Although toxigenic Vibrio cholerae, responsible for the disease cholera, can be found in nutrient-poor aquatic environments in endemic areas, the underlying mechanism(s) by which culturable cells persist in these environmental reservoirs is largely unknown. Here we report that introduction of V. cholerae into a nutrient-poor filter sterilized lake water (FSLW) microcosm promoted a shift to what we have defined as a "persister" phenotype (PP) which was culturable for >700 days. Dire

Reversible hyperinsulinemic hypoglycemia after gastric bypass: a consequence of altered nutrient delivery.

CONTEXT: Severe hypoglycemia after Roux-en-Y gastric bypass surgery (RYGB) is an increasingly recognized condition, characterized by neuroglycopenia and inappropriately elevated insulin concentrations that occur primarily in the postprandial state. Both pathophysiology and treatment of this disorder remain elusive, but it has been postulated that hyperplasia and/or hypertrophy of beta-cells due to morbid obesity and/or postsurgical nesidioblastosis may contribute. OBJECTIVE: The objective of this study was to elucidate the pathophysiology of this condition; specifically, we hypothesized that

Role of vegetation in a constructed wetland on nutrient-pesticide mixture toxicity to Hyalella azteca.

The toxicity of a nutrient-pesticide mixture in nonvegetated and vegetated sections of a constructed wetland (882 m² each) was assessed using Hyalella azteca 48-h aqueous whole-effluent toxicity bioassays. Both sections were amended with a mixture of sodium nitrate, triple superphosphate, diazinon, and permethrin simulating storm-event agricultural runoff. Aqueous samples were collected at inflow, middle, and outflow points within each section 5 h, 24 h, 72 h, 7 days, 14 days, and 21 days postamendment. Nutrients and pesticides were detected throughout both wetland sections with concentrations…

Nutrient-regulated transcriptional responses in the brown tide-forming alga Aureococcus anophagefferens.

Long-SAGE (serial analysis of gene expression) was used to profile the transcriptome of the brown tide-forming alga, Aureococcus anophagefferens, under nutrient replete (control), and nitrogen (N) and phosphorus (P) deficiency to understand how this organism responds at the transcriptional level to varying nutrient conditions. This approach has aided A. anophagefferens genome annotation efforts and identified a suite of genes upregulated by N and P deficiency, some of which have known roles in nutrient metabolism. Genes upregulated under N deficiency include an ammonium transporter, an acetami

Recovery of struvite from animal wastewater and its nutrient leaching loss in soil.

Contaminants in swine wastewater were recovered in the form of struvite, a crystal of magnesium ammonium phosphate (MAP), using a newly designed process, and the leaching loss of MAP in soil was examined. The continuous flow process was operated under optimal conditions: 1.0 molar ratio of magnesium (Mg) addition with respect to orthophosphate (OP) and an aeration rate of 0.73 L/L min. Five treatments were performed with three replications for soil pH and nutrient leaching loss tests. It was found that 93% of the OP in the swine wastewater was crystallized, and the MAP crystal formation was ve

Regulation of endothelin-1 expression and function by nutrient stress in mouse colon epithelia.

OBJECTIVE: The endothelin (ET) system is influenced by a variety of stress conditions in many tissues. However, the effects of nutrient stress conditions on ET expression and its function are not well understood in the intestinal tract, while ET-1 gene expression and peptide were found in the intestinal tract. The aim of this study was to investigate the effect of feeding and fasting on the expression of ET-1 and short-circuit current (Isc) induced by ET-1 in mouse colon. MATERIAL AND METHODS: Mice were fed freely, fasted for 48 h, and re-fed after fasting, respectively. ET-1 mRNA levels and

Gene-nutrient interactions in G6PD-deficient subjects--implications for cardiovascular disease susceptibility.

BACKGROUND/AIMS: The enzyme glucose-6-phosphate dehydrogenase (G6PD) is the principal source of reducing equivalents, necessary for regenerating reduced glutathione through NADPH in order to protect cells from oxidative damage, and whichin erythrocytes produces hemolysis. When fava beans are ingested by G6PD-deficient subjects (gene-nutrient interaction), or some oxidant drugs are assumed (gene-drug interactions), a life-threatening hemolysis can occur. However, the same defect results in lower cardiovascular disease (CVD) risk. METHODS: Physiopathological, clinical and mortality studies of C

Nutrient load generated by storm event runoff from a golf course watershed.

Turf, including home lawns, roadsides, golf courses, parks, etc., is often the most intensively managed land use in the urban landscape. Substantial inputs of fertilizers and water to maintain turf systems have led to a perception that turf systems are a major contributor to nonpoint source water pollution. The primary objective of this study was to quantify nutrient (NO(3)-N, NH(4)-N, and PO(4)-P) transport in storm-generated surface runoff from a golf course. Storm event samples were collected for 5 yr (1 Apr. 1998-31 Mar. 2003) from the Morris Williams Municipal Golf Course in Austin, TX. I

Effects of dietary genistein on nutrient use and mineral status in heat-stressed quails.

Genistein is a powerful antioxidant and plays a role in calcium and bone metabolism. We evaluated the efficacy of dietary supplementation with genistein on the nutrient use and mineral concentrations in tibia and serum of quails reared at high environmental temperature (34 degrees C). Two hundred and forty Japanese quails (10 days old) were randomly assigned to 8 treatment groups consisting of 10 replicates of 3 birds. The birds were kept in a temperature-controlled room at 22 degrees C (Thermoneutral, TN groups) or 34 degrees C (for 8 h/d; 09.00 am-05.00 pm; Heat stress, HS groups). Birds wer

Nutrient regulation of pancreatic beta-cell function in diabetes: problems and potential solutions.

Increasing prevalence of obesity combined with longevity will produce an epidemic of Type 2 (non-insulin-dependent) diabetes in the next 20 years. This disease is associated with defects in insulin secretion, specifically abnormalities of insulin secretory kinetics and pancreatic beta-cell glucose responsiveness. Mechanisms underlying beta-cell dysfunction include glucose toxicity, lipotoxicity and beta-cell hyperactivity. Defects at various sites in beta-cell signal transduction pathways contribute, but no single lesion can account for the common form of Type 2 diabetes. Recent studies highli

Adequate nutrient intake can reduce cardiovascular disease risk in African Americans.

Cardiovascular disease kills nearly as many Americans each year as the next seven leading causes of death combined. The prevalence of cardiovascular disease and most of its associated risk factors is markedly higher and increasing more rapidly among African Americans than in any other racial or ethnic group. Improving these statistics may be simply a matter of improving diet quality. In recent years, a substantial and growing body of evidence has revealed that dietary patterns complete in all food groups, including nutrient-rich dairy products, are essential for preventing and reducing cardiov
