Analysis of trace levels of reactive carbonyl compounds (RCCs), including formaldehyde, acetaldehyde, acrolein, malonaldehyde, glyoxal, and methyl glyoxal, is extremely difficult because they are highly reactive, water soluble, and volatile. Determination of these RCCs in trace levels is important because they are major products of lipid peroxidation, which is strongly associated with various diseases such as cancer, Alzheimer's disease, aging, and atherosclerosis. This review covers the development and application of various derivatives for RCC analysis. Among the many derivatives which have

We investigate the nature of valency and bonding in the highly unusual O(2)PR(3) "phosphadioxirane" species recently isolated as an intermediate in the reaction of (1)O(2) with organic phosphines PR(3). Commonly, this species is depicted as a Lewis structure with five bonds at the phosphorus center, suggesting hypervalent involvement of extra-valence d-orbitals in the hybridization. However, nonhypervalent bonding patterns, such as open zwitterionic peroxides or R(2)PO(2)(+)R(-) ion pairs, could also achieve the observed hypercoordination. In the present work, we employ ab initio and hybrid de…

BACKGROUND: It was reported that elevation of the intracellular concentration of free Ca2+ ([Ca2+]i) by a calcium ionophore increased the release of herpes simplex virus type 1 (HSV-1). Freely diffusible hydrogen peroxide (H2O2) is implied to alter Ca2+ homeostasis, which further enhances abnormal cellular activity, causing changes in signal transduction, and cellular dysfunction. Whether H2O2 could affect [Ca2+]i in HSV-1-infected cells had not been investigated.
RESULTS: H2O2 treatment increased the amount of cell-free virus and decreased the proportion of viable cells. After the treatment,

The neonatal brain responds differently to hypoxic-ischemic injury and may be more vulnerable than the mature brain due to a greater susceptibility to oxidative stress. As a measure of oxidative stress, the immature brain should accumulate more hydrogen peroxide (H2O2) than the mature brain after a similar hypoxic-ischemic insult. To test this hypothesis, H2O2 accumulation was measured in postnatal day 7 (P7, neonatal) and P42 (adult) CD1 mouse brain regionally after inducing HI by carotid ligation followed by systemic hypoxia. H2O2 accumulation was quantified at 2, 12, 24, and 120 h after HI

There is accumulating evidence of altered antioxidant enzyme activities and increased levels of lipid peroxidation in schizophrenia. Free radical-mediated abnormalities may contribute to specific aspects of schizophrenic symptomatology and complications of its treatment. However, few studies have evaluated both antioxidant enzymes and lipid peroxidation in the same schizophrenic patient groups treated with typical or atypical antipsychotics. Plasma malondialdehyde (MDA) levels and superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) and catalase (CAT) activities were analyzed using esta

We used animal models to study connection between oxidating system and sphingomyelin signaling cascade, because this models are more close related to people disease. Activation of n-sphingomyelinase (n-SMase) in mice liver and brain is coincided in time with increased level of peroxide products (conjugated dienes) after injection of tumor necrosis factor alpha (TNF-alpha). We found that ceramide can induce peroxide oxidation and lead to accumulation of TNF-alpha in animal organs. Nitric oxide (NO) donors (S-nitrosoglutathione and dinitrosyl iron complex) reversibly inhibited activity of n-SMas

A detailed study is presented on the use of an easily accessible probe (the europium-tetracycline 3:1 complex; referred to as Eu3Tc) for determination of hydrogen peroxide (HP). Eu3Tc undergoes a 15-fold increase in luminescence intensity on exposure to an excess of HP. Data are given on the time dependence of the reaction, on the pH dependence of the absorption and emission spectra of both the probe and its complex with HP, and on the effect of stoichiometry between Eu3+ and Tc on selectivity and signal change. HP can be quantified in aqueous solution of pH 6.9 over a 2-400 microM concentrati

Despite its importance for understanding the behaviour of antimony in the environment, the oxidation kinetics of Sb(III) with natural oxidants is still not well understood. We have studied the oxidation of Sb(III) by hydrogen peroxide on a time scale of hours in the presence of some trace metals, Cu(II), Mn(II), Zn(II) and Pb(II), under pH and concentration conditions close to natural ones. The effects that these trace metals have on Sb(iii) oxidation by hydrogen peroxide vary. Zn(II) had no catalytic effect at all, but Cu(II), Mn(II) and Pb(II) did, though their effects were not uniform. Cu(I

Reactive oxygen species (ROS) are reported to play a role in inducing the proteinuria of nephrotic syndrome (NS). This study investigated paraoxonase (PON), total antioxidant response (TAR), and oxidant total peroxide together with serum proteins and lipoproteins in children with steroid-sensitive NS. The study included 40 children with steroid-sensitive NS (21 with acute-period NS in group I, 19 nonproteinuric while receiving steroids in group II) and 22 sex- and age-matched formerly nephrotic children in remission weaned from steroids (group III). The following parameters were determined: to

Epidemiological and clinical studies have proved the beneficial effect of bioactive compounds of vegetables and medicinal plants used in phytotherapy on prevention of several diseases. Beyond the well-known natural antioxidants, such as vitamins, other natural substances can function as antioxidants. In 1992 the Saas Fee Declaration was published by leader nutritionist and biochemists underlining the importance of prevention in health maintenance based on scientific studies with natural compounds. According to the declaration the antioxidants are the main active and beneficial components. Auth

Presenilin-1 (PS1) is the gene responsible for the development of early-onset familial Alzheimer's disease. To probe the functions of PS1 on neuronal resistance to oxidative stress, we pharmacologically examined the death signals in PS1-deficient neurons induced by oxidative stress. Because the death of primarily cultured neurons lacking PS1 is caused by hydrogen peroxide in calcium-dependent manners in vitro [J Neurochem 78 (2001) 807], we tested the neuronal survival-promoting ability of inhibitors against calcium-dependent/cell death-related signaling molecules, such as ERKs, JNK, p38 MAP k

The role of reactive oxygen-induced oxidative damage to lipids (i.e., lipid peroxidation, LP) and proteins has been strongly supported in previous work. Most notably, a number of free radical scavengers and lipid antioxidants have been demonstrated to be neuroprotective in traumatic brain injury (TBI) models. However, the specific sources of reactive oxygen species (ROS), the time course of oxidative damage and its relationship to post-traumatic neurodegeneration in the injured brain have been incompletely defined. The present study was directed at an investigation of the role of the ROS, pero

The effect of iron dinitrosyl complexes, S-nitrosoglutathione, and glutathione on free radical oxidation of rat heart mitochondria induced by tert-butyl hydroperoxide and metmyoglobin or their combination with ferritin was studied. It was shown that iron dinitrosyl complexes or the combination of S-nitrosoglutathione and glutathione inhibited most effectively the peroxidation of mitochondrial membranes. It was found that ferritin stimulated the prooxidant action of metmyoglobin. Using EPR spectroscopy, it was established that, in conditions of O2*- generation, the destruction of iron dinitrosy

This study examined the relationship between oxidative stress and enzymic antioxidant status in the erythrocytes of thirty-two adult cervical cancer patients and an equal number of age-matched cervicitis patients and healthy subjects. Lipid peroxidation was significantly increased, while the activities of enzymic antioxidants (superoxide dismutase, catalase, glutathione peroxidase) and glucose-6-phosphate dehydrogenase were decreased in the erythrocytes of cervical cancer patients as compared to healthy subjects and cervicitis patients. Thus, this study has demonstrated elevated lipid peroxida
