Weight of evidence in safety pharmacology guidance: A case for explicit probabilistic reasoning in the revision of ICH S7A.

ICH S7A (2000) defined the foundational framework for safety pharmacology studies supporting the registration of human pharmaceuticals. Overall, the guidance applied a risk-based, rational scope combined with a prescriptive framework of core battery endpoints collected under GLP guidelines that were required prior to first-in-human testing. Because of the evolving landscape of pharmaceutical modalities, nonclinical methodologies, and regulatory science over the last two decades, a critical examination of its relationship to weight-of-evidence (WoE) and risk-based reasoning is warranted. The ce

Post-immobilization phosphorus fluctuations drive selective microbial hydrolysis of sedimentary organic phosphorus and long-term P release.

In-situ immobilization has been widely utilized as an efficient and cost-effective phosphorus (P) treatment technology for lake eutrophication. However, post-immobilization P fluctuations in the overlying water may trigger the compensation of endogenous P from sediments over the long term. Microbially mediated hydrolysis of sedimentary organic phosphorus (SOP) is widely recognized as a pivotal process driving this P replenishment. However, the specific SOP hydrolysis pathways and the underlying microbial interactions remain to be systematically elucidated. Herein, this study explored the post-

Crystal structure and biochemical analysis of the polyester-degrading carboxylesterase SM0281 from the symbiotic legume-associated rhizobacterium Sinorhizobium (Ensifer) meliloti.

The microbial degradation of synthetic polyesters such as polyethylene terephthalate (PET) is mediated by diverse α/β-hydrolases, many of which remain poorly characterized. In this study, we investigated SM0281, an uncharacterized α/β-hydrolase from the symbiotic legume-associated rhizobacterium Sinorhizobium meliloti. Sequence analysis revealed that SM0281 shares low similarity with known polyester-degrading enzymes. Biochemical characterization of purified SM0281 demonstrated that it is a carboxylesterase with preference for medium-chain length monoester substrates (C4-C8) and displays broad

Management of Minor Superior Sagittal Sinus Laceration in Parasagittal Meningioma Surgery: Operative Video.

Superior sagittal sinus (SSS) injury represents a potentially serious intraoperative complication during parasagittal meningioma surgery. Although primary venous repair with direct suturing remains the preferred reconstructive strategy when technically feasible, selected minor sinus wall injuries may require less manipulative hemostatic strategies aimed at preserving venous patency while minimizing sinus manipulation. We report the case of a 44-year-old man with multiple intracranial meningiomas who underwent resection of a right frontal parasagittal lesion located at the junction between the

Counseling and use of surrogacy among cancer survivors (CUSACS).

PURPOSE: International guidelines recommend fertility counseling at diagnosis for all women with cancer of child-bearing potential, yet whether gestational surrogacy (GS) is included in such counseling is not clear, nor is whether and what proportion of survivors are aware of GS. We assessed the counseling and use of GS, and predictors of oncologist-provided GS counseling among female cancer survivors. METHODS: A survey on fertility and GS counseling was distributed via national advocacy organizations (April-November 2024). Descriptive statistics and logistic regression evaluated predictors o

Utilization of the Community Engagement Studio model to facilitate participatory design of a multicenter randomized controlled trial of the efficacy of scoliosis specific exercise rehabilitation.

INTRODUCTION: Engaging individuals with lived experience enhances the rigor and real-world relevance of clinical research. In adolescent idiopathic scoliosis (AIS), where non-surgical evidence remains limited, involving patients and caregivers informs design, recruitment, and retention for future trials. METHODS: Using the Community Engagement (CE) Studio model, we conducted a 1.5-hour virtual session with children aged 10-16 years with AIS and their caregivers residing in North America. Participants (no spinal surgery; with/without brace) were recruited through engagement databases, advocacy

Recommendations for the Evaluation and Management of Inflammatory Bowel Disease With Irritable Bowel Syndrome-Like Symptoms: A Joint Rome Foundation and International Organization for the Study of IBD (IOIBD) Consensus.

BACKGROUND & AIMS: A substantial proportion of persons with inflammatory bowel disease (IBD) in remission continue to experience abdominal pain, altered bowel habits, and bloating that resemble irritable bowel syndrome (IBS). Lack of standardized definitions and evidence-based management strategies leads to diagnostic ambiguity and potentially unnecessary escalation of IBD therapy. A joint Rome Foundation/International Organization for the Study of Inflammatory Bowel Disease Working Team developed consensus recommendations on nomenclature, evaluation, and treatment of IBD with IBS-like symptom

Attention maps reveal stimulus-dependent retinal population codes.

INTRODUCTION: Understanding how deep learning models map neural population activity to stimuli requires both high predictive accuracy and interpretable internal mechanisms. METHODS: In this work, we employ the POYO framework, a scalable transformer architecture based on spike tokenization and latent modeling, to decode large-scale retinal ganglion cell recordings. We ask whether the model's attention mechanisms can provide biologically meaningful insight by evaluating two contrasting conditions: a uniform flash stimulus and a spatiotemporally structured moving ball stimulus. RESULTS: We show

Spatiotemporal metabolomics of tobacco leaves in response to Pseudomonas syringae infection.

Tobacco (Nicotiana tabacum L.) is a major economic crop and a model for plant-pathogen interactions, yet the spatiotemporal dynamics of defense metabolism during infection remain poorly characterized. Here, we used MALDI-MSI-based spatial metabolomics to systematically profile tobacco leaves during Pseudomonas syringae infection. Multidimensional analysis of 1,399 annotated metabolites revealed distinct spatiotemporal regulation patterns. Temporally, early infection (12 h postinfection (hpi)) was characterized by increased organic acids and terpenoids, followed by a mid-stage shift toward phen
