[Study on network pharmacological mechanism of "treating different diseases with same method" of Notoginseng Radix et Rhizoma in treating diabetic nephropathy, diabetic encephalopathy and diabetic cardiomyopathy].

Pharmacology network was used to investigate the common key target and signaling pathway of Notoginseng Radix et Rhizoma in the protection a … t targets of Notoginseng Radix et Rhizoma, respectively. Enrichment analysis indicated that the AGE-RAGE signaling pathway and FoxO signaling pathway were the common pathways in the protection of Notoginseng Radix et Rhizoma against DN, DE and DCM. Network analysis of the overlapped targets showed that TNF, STAT3, IL6, VEGFA, MAPK8, CASP3 and SIRT1 wer …

A monoclonal antibody targeting the C-terminal of α-synuclein fibrils mitigates pathology in a Parkinson's disease model.

Parkinson's disease (PD) pathogenesis is driven by α-synuclein (α-syn) amyloid aggregation, with the flexible C-terminal region mediating pa … eractions between α-syn fibrils and established receptors and binding partners, including FAM171A2, RAGE, LAG3, and LC3B. Cryo-electron microscopy (cryo-EM) and cryo-electron tomography (cryo-ET) studies reveal that H21 engages α-syn fibrils in a periodic manner, inducing conformational remodeling in the fibril architecture. In a PD mouse model, H21 tr …

The Role of the HMGB1 C-Terminal Domain in Epithelial-Mesenchymal Transition and Invasion in 2D and 3D MDA-MB-231 Breast Cancer Models.

High-mobility group box 1 (HMGB1) is a multifunctional protein that operates both within the nucleus and as an extracellular signaling molec … lar plasticity and invasive potential through distinct signaling pathways not strictly dependent on RAGE (Receptor for Advanced Glycation End-product) under the tested conditions. Functional analyses indicate that this truncated form promotes epithelial-mesenchymal transition-related behaviors and activates downstream inflammatory signaling in a contex …

Lecanemab treatment improves B cell subpopulation immune homeostasis in patients with Alzheimer's disease.

INTRODUCTION: Lecanemab, the first disease-modifying therapy for Alzheimer's disease (AD), mitigates pathology primarily by clearing amyloid … onic inflammation and reversing the dysregulation of key pathways including immune response, NF-κB, RAGE, and cell adhesion. DISCUSSION: These findings uncover a novel peripheral immunomodulatory mechanism of Lecanemab, offering new insights into AD therapeutics.

We show the modularity and generality of the EPLA technology by degrading membrane-bound PD-L1, RAGE, GRP94, PSMA, ICAM-1, p32, VCAM-1 and LRP1, and extracellular amyloid-β and tau. Pseudomonas aeruginosa pulmonary infection results in S100A8/A9-dependent cardiac dysfunction.

Pseudomonas aeruginosa (P.a.) infection accounts for nearly 20% of all cases of hospital acquired pneumonia with mortality rates >30%. P.a. … ally, we found that neutrophil recruitment and release of S100A8/A9 in the lungs activates the TLR4/RAGE signaling pathways, which in turn enhance systemic inflammation and subsequent cardiac dysfunction. Paradoxically, global deletion of S100A8/A9 did not improve but aggravated cardiac dysfunction and mortality likely due to uncontrolled bacterial bur …

Potential effect of advanced glycation end products (AGEs) on spermatogenesis and sperm quality in rodents.

The aim of the present study was to assess whether dietary advanced glycation end products (AGEs) induce testicular dysfunction. Using a BALB/c mouse model, AGE intake and serum levels were found to increase in AGE diet-treated mice relative to the controls. Histopathological damage was detected in the testes and epididymides of the AGE diet-induced mice. The total number of epididymal sperm decreased, and increased abnormal sperm rate was found in the mice. Moreover, the mice testes showed an increased level of the receptor for AGEs (RAGE) and malondialdehyde (MDA). Using a Sprague-Dawley rat…

Emerging Role of High Mobility Group Box-1 in Thrombosis-Related Diseases.

High mobility group box-1 (HMGB-1), a typical damage-associated molecular pattern protein released from various cells, was first identified in 1973. It is usually stored in the nuclei of cells. Several modifications of HMGB-1 promote its translocation to the cytosol, and it is actively or passively released from cells. When outside of the cells, HMGB-1is crucial in inflammation. It exerts its biological functions via interaction with its receptors, including receptor for advanced glycation end products (RAGE) and Toll-like receptor 4(TLR4). A large number of studies showed a close link between…

Argpyrimidine-tagged rutin-encapsulated biocompatible (ethylene glycol dimers) nanoparticles: Application for targeted drug delivery in experimental diabetes (Part 2).

Diabetes mellitus is characterized by hyperglycemia and associated complications. However, long-term diabetes control is not often sustained by currently available therapeutic approaches. Research on nanoparticle-mediated drug delivery systems is in progress. Here we have tested a ligand (argpyrimidine)-tagged drug (rutin)-encapsulated biocompatible (ethylene glycol dimers) nanoparticle for targeted drug delivery in streptozotocin-induced diabetic rats. Argpyrimidine, being an advanced glycation end product (AGE), directs the nanoparticles to interact with cell surface receptors of AGEs (RAGE)…

Spectral analysis of human tremor during cognitive tasks.

Many stresses occur in our daily lives. Some of these are part of diseases of the nervous system such as ataxia and neuroses. Certain body t … ibration amplitude spectrum caused by mental arithmetic is significantly increased in the frequency rage of 0.5 to 0.7 Hz, when compared to those recorded during other tasks. The induced tremor, as well as general invisible body micro-vibration, were obtained from the three-dimensional body displacements.

HMGB1: a key molecule linking chronic inflammation to complications in type 2 diabetes mellitus and a target for exercise intervention.

The pathological process of type 2 diabetes mellitus (T2DM) is closely associated with chronic low-grade inflammation. High mobility group box 1 (HMGB1), a key damage-associated molecular pattern (DAMP), is frequently dysregulated in T2DM and is implicated in promoting insulin resistance (IR), β cell dysfunction, and the progression of multiple complications-including cardiovascular disease, nephropathy, cognitive impairment, myopathy, and dyslipidemia-primarily through activating signaling pathways such as RAGE/TLR4-NF-κB. Exercise, a cornerstone non-pharmacological intervention, effectively …

Tumor-Derived HMGB1 Orchestrates Lysosomal Metabolic Reprogramming for M1 Macrophage Polarization.

Non-small-cell lung cancer (NSCLC) is considered as a predominant contributor to cancer-related mortality with significant infiltration by T … e nucleus, initiating lysosomal metabolism. Meanwhile, in macrophages, tumor-derived HMGB1 binds to RAGE, thereby promoting polarization toward the M1 macrophage phenotype. This study uncovers a divergent metabolic program mediated by HMGB1 and demonstrates its role in lysosomal metabolic plasticity.
