Biological effects of blocking blue and other visible light on the mouse retina.

BACKGROUND: To elucidate the biological effects of blocking fluorescent light on the retina using specific blocking materials. METHODS: Seven- to 8-week-old BALB/c mice were divided into three groups and placed in one of the three boxes: one blocked ultraviolet and violet wavelengths of light (violet blockade), one blocked ultraviolet, violet, blue and some other visible wavelengths (blue-plus blockade), and one allowed most visible light to pass through (control). They were then exposed to a white fluorescent lamp for 1 h at 5.65E-05 mW/cm(2) /s. After treatment, the electroretinogram, retin…

Local activation of dendritic cells alters the pathogenesis of autoimmune disease in the retina.

Interest in the identities, properties, functions, and origins of local APC in CNS tissues is growing. We recently reported that dendritic cells (DC) distinct from microglia were present in quiescent retina and rapidly responded to injured neurons. In this study, the disease-promoting and regulatory contributions of these APC in experimental autoimmune uveoretinitis (EAU) were examined. Local delivery of purified, exogenous DC or monocytes from bone marrow substantially increased the incidence and severity of EAU induced by adoptive transfer of activated, autoreactive CD4 or CD8 T cells that w…

Neuronal clustering and fasciculation phenotype in Dscam- and Bax-deficient mouse retinas.

Individual types of retinal neurons are distributed to minimize proximity to neighboring cells. Many of these same cell types extend dendrites to provide coverage of the retinal surface. These two cardinal features of retinal mosaics are disrupted, for certain cell types, in mice deficient for the Down syndrome cell adhesion molecule, Dscam, exhibiting an aberrant clustering of somata and fasciculation of dendrites. The Dscam mutant mouse retina also exhibits excess numbers of these same cell types. The present study compared these two features in Dscam mutant retinas with the Bax knockout ret…

Rax : developmental and daily expression patterns in the rat pineal gland and retina.

Retina and anterior neural fold homeobox (Rax) gene encodes a transcription factor essential for vertebrate eye development. Recent microarray studies indicate that Rax is expressed in the adult rat pineal gland and retina. The present study reveals that Rax expression levels in the rat change significantly during retinal development with a peak occurring at embryonic day 18, whereas Rax expression in the pineal is relatively delayed and not detectable until embryonic day 20. In both tissues, Rax is expressed throughout postnatal development into adulthood. In the mature rat pineal gland, the …

Gene expression and protein distribution of orexins and orexin receptors in rat retina.

Orexins, composed of orexin A and orexin B, are identified as endogenous ligands of two orphan G-protein-coupled receptors: orexin 1 and orexin 2 receptors (OX1R and OX2R). Orexins are implicated in regulating wake/sleep states, feeding behaviors, etc. Using reverse transcription-polymerase chain reactive (RT-PCR) analysis and immunofluorescence double labeling, we investigated the distributions of orexin A, orexin B, OX1R and OX2R in rat retina. RT-PCR analysis revealed the presence of mRNAs of prepro-orexin, OX1R and OX2R in rat retina. Immunostaining for orexin A and orexin B was observed i…

Biplexiform ganglion cells contact photoreceptors in the retina of the greenling Hexagrammos octogrammus.

The biplexiform cell (bpxRGC) is a relatively and recently discovered type of retinal ganglion cells. Like "ordinary" ganglion cells, bpxRGCs have dendrites arborizing within the inner plexiform layer. However, as distinct from other ganglion cells, they have dendrites ascending to the outer plexiform layer. To date, bpxRGCs have been found in mammals, amphibians, and teleost fishes (Cook et al. [1996] Vis Neurosci 13:517-528). The mammalian and amphibian bpxRGCs form direct contacts with photoreceptors and may participate in rapid signal transmission to the brain (Mariani [1982] Science 216:1…

Detection of the temporal arcade in fundus images of the retina using the Hough transform.

Quantitative analysis of the vascular architecture of the retina can help in monitoring the effects of retinopathy on the visual system. Retinopathy affects the blood vessels in the retina through modification of the shape, width, tortuosity, and the angle of insertion of the temporal arcade. Monitoring the openness of the temporal arcade and changes with treatment can facilitate improved diagnosis and optimized treatment. We propose methods for the detection and parametric modeling of the temporal arcade, including gradient operators and Gabor functions to detect retinal vessels, and the Houg…

Photoperiod-dependent changes in melatonin synthesis in the turkey pineal gland and retina.

The effect of photoperiod on melatonin content and the activity of the melatonin-synthesizing enzymes, namely, serotonin N-acetyltransferase (AANAT) and hydroxyindole-O-methyltransferase, were investigated in the pineal gland and retina of turkeys. The birds were adapted to 3 different lighting conditions: 16L:8D (long photoperiod), 12L:12D (regular photoperiod), and 8L:16D (short photoperiod). Pineal, retinal, and plasma melatonin concentrations oscillated with a robust diurnal rhythm, with high values during darkness. The duration of elevated nocturnal melatonin levels in the turkey pineal g…

Mechanisms of endothelial cell guidance and vascular patterning in the developing mouse retina.

The appropriate guidance and patterning of vessels during vascular development is critical for proper tissue function. The loss of these guidance mechanisms can lead to abnormal vascularization and a number of pathological conditions. The molecular basis of endothelial cell guidance and subsequent tissue specific vascular patterning remains largely unknown in spite of its clinical relevance and biological importance. In this regard, retinal vascular development offers many advantages for studying endothelial cell guidance and the mechanisms by which characteristic vascular patterns are formed.…

The retinal vasculature and function of the neural retina in a rat model of retinopathy of prematurity.

PURPOSE: In a rat model of retinopathy of prematurity (ROP), the retinal vasculature and function of the neural retina were studied longitudinally. Vascular and neural parameters were evaluated for significant relationships. METHODS: Retinopathy was induced by exposing newborn rats to alternating 50% and 10% oxygen until age 14 days. To evaluate the function of the neural retina, electroretinographic (ERG) responses to full-field stimuli were recorded from dark-adapted rats at ages 18 and 31 days. Sensitivity and saturated amplitude of photoreceptor and postreceptor activity were derived from…

Diurnal and circadian rhythms in melatonin synthesis in the turkey pineal gland and retina.

The pineal gland and retina of the turkey rhythmically produce melatonin. In birds kept under a daily light-dark (LD) illumination cycle melatonin concentrations in the pineal gland and retina were low during the light phase and high during the dark phase. A similar melatonin rhythm with high night-time values was also observed in the plasma. The pineal and retinal melatonin rhythms mirror oscillations in the activity of serotonin N-acetyltransferase (AANAT; the penultimate enzyme in the melatonin biosynthetic pathway). In contrast, in both the pineal gland and retina the activity of the enzym…

Microglial responses in the avascular quail retina following transection of the optic nerve.

This study was undertaken to investigate microglial responses in the avascular central nervous system using the quail retina that is known to be devoid of blood vessels. Following intraorbital optic nerve transection (ONT), the quail retina was examined immunohistochemically at various times up to 6 months. A few days after transection, microglia in the inner retinal layers revealed features of activation. Activated cells displayed an amoeboid shape and enhanced QH1-immunoreactivity. The numbers of these amoeboid cells were rapidly increased, first in the inner plexiform layer (IPL), and then …
