A model to test how ticks and louping ill virus can be controlled by treating red grouse with acaricide.

Ticks are the most important vectors of disease-causing pathogens in Europe. In the U.K., Ixodes ricinus L. (Ixodida: Ixodidae) transmits louping ill virus (LIV; Flaviviridae), which kills livestock and red grouse, Lagopus lagopus scoticus Lath. (Galliformes: Phasianidae), a valuable game bird. Tick burdens on grouse have been increasing. One novel method to reduce ticks and LIV in grouse may be acaricide treatment. Here, we use a mathematical model parameterized with empirical data to investigate how the acaricide treatment of grouse might theoretically control ticks and LIV in grouse. Assumi…

The importance of the aggregation of ticks on small mammal hosts for the establishment and persistence of tick-borne pathogens: an investigation using the R(0) model.

Aggregation of parasites amongst hosts is important for the epidemiology of vector-borne diseases because hosts that support the majority of the vector population are responsible for the majority of pathogen transmission. Ixodes ricinus ticks transmit numerous pathogens of medical importance including Borrelia burgdorferi s.l. and tick-borne encephalitis virus. One transmission route involved is 'co-feeding transmission', where larvae become infected via feeding alongside infected nymphs. The aggregation of ticks on hosts leads to an increase in the number of larvae feeding alongside nymphs, i…

Cultivation of an ovine strain of Ehrlichia phagocytophila in tick cell cultures.

Ehrlichia phagocytophila (previously known as Cytoecetes phagocytophila) which causes tick-borne fever (TBF) in sheep and pasture fever in cattle in the UK and mainland Europe is transmitted by the temperate hard tick Ixodes ricinus. The disease in sheep is characterized by fever, leucopenia and immunosuppression. Studies on the pathogenesis and other aspects of the disease have been hampered because the organism has not been cultivated in continuous or primary cell culture systems. This paper describes the first successful cultivation of a European isolate of E. phagocytophila in two continuo…

Incidence from coincidence: patterns of tick infestations on rodents facilitate transmission of tick-borne encephalitis virus.

Tick-borne encephalitis (TBE) virus has a highly focal distribution through Eurasia. Endemic cycles appear to depend on the transmission of non-systemic infections between ticks co-feeding on the same rodent hosts. The particular features of seasonal dynamics and infestation patterns of larval and nymphal Ixodes ricinus, but not Dermacentor reticulatus, from 4 regions within TBE foci in Slovakia, are such as to promote TBE virus transmission. The distributions of larvae and nymphs on their principal rodent hosts are highly aggregated and, rather than being independent, the distributions of eac…

First characterization of Borrelia burgdorferi sensu lato from ticks and skin biopsy in Bulgaria.

Lyme borreliosis is the most frequent tick-borne disease in the Northern hemisphere. Here we describe the first isolation of Borrelia burgdorferi sensu lato in Bulgaria: the midguts of 47 Ixodes ricinus obtained by flagging from the Central park in Sofia, Bulgaria were cultivated for borreliae in BSK medium. The eight isolates obtained from the ticks and one skin isolate from a Bulgarian patient with erythema migrans were subjected to phenotypic [outer surface protein A (OspA) serotyping] and genotypic analysis (pulsed-field gel electrophoresis typing followed by large restriction fragment pat…

Detection of the emerging tick-borne pathogen, Tamdy virus, in Dermacentor marginatus ticks, southeastern Türkiye.

Tamdy virus (TAMV) is an emerging tick-borne virus in the family Nairoviridae, identified as a human pathogen causing febrile illness after tick exposure. This study aimed to detect and characterise TAMV in ticks collected from non-endemic regions of Türkiye. A total of 529 ticks were sampled from six provinces and identified as Haemaphysalis parva (27%), Rhipicephalus bursa (26%), Haemaphysalis punctata (20.7%), Dermacentor marginatus (17.2%), and Ixodes ricinus (8.8%). Screening with a single-step generic PCR revealed one TAMV-positive pool out of 96. For characterisation, the S segment of T…

Immunomodulatory Effects of a Tick Salivary Serpin on Psoriasis-like Inflammation.

Psoriasis is a chronic inflammatory disease with a complex pathogenesis, and it is mainly driven by a dysregulation in immune responses. Therapeutic strategies constantly require novel compounds targeting immune modulation to substitute the current traditional drugs characterized by side effects and limited efficacy. In this study, we used a mannan-induced psoriasis-like inflammation mouse model to investigate the immunomodulatory potential of Iripin-3, a salivary serpin from the Ixodes ricinus ticks. Mice treated with Iripin-3 showed improvements in the severity of psoriasis-like lesions, as …

Days of Future Passed: Best Calibration Does Not Guarantee Reliable Forecasts of Future Tick Distributions.

Forecasting species distributions under future climate scenarios has become a standard application of Species Distribution Models (SDMs), yet their temporal transferability is rarely evaluated. We assessed the reliability of future projections using four widespread tick species (Amblyomma americanum, Dermacentor variabilis, Ixodes ricinus, and Ixodes scapularis) with occurrence records spanning from 1961-2025. Models were calibrated sequentially using decade-specific records and projected onto subsequent decades, for which independent observations were available. Climate predictors derived fro…

Epidemiological, Clinical and Analytical Features in Lyme Borreliosis Patients Seropositive for Babesia divergens/venatorum.

Lyme borreliosis (LB), caused by Borrelia burgdorferi sensu lato (s.l.) and babesiosis, caused by Babesia divergens and Babesia venatorum, are both transmitted by the ixodid tick Ixodes ricinus. Although these diseases coexist in Spain and other European regions, no studies have been carried out to assess the impact on the health of patients exposed to both causative pathogens. This retrospective study, conducted in Asturias in northwestern Spain between 2015 and 2017, analyzed the possible complications arising from exposure to these pathogens. To this end, the epidemiological, clinical and a…

A new hotspot of tick-borne encephalitis virus (TBEV) in the Autonomous Province of Trento, Italy.

The tick-borne encephalitis virus (TBEV) has expanded its distributional range in Europe over recent decades. Italy is considered a low tick-borne encephalitis (TBE) incidence country, nonetheless, human cases have recently increased, reaching their maximum so far in 2022. This study reports on a new TBE hotspot in the Autonomous Province of Trento (Italy), along with a description of TBEV genetic variants at the provincial level. Tick sampling was performed where several TBE human cases were reported in 2022. As a result, 458 Ixodes ricinus ticks were collected (38 females, 49 males, 371 nymp…

Global risk dynamics of Borrelia miyamotoi in the context of climate change.

The impact of Borrelia miyamotoi on human health, facilitated by the expanding geographical distribution and increasing population of Ixodes ticks, remains obscure in the context of global climate change. We employed multiple models to evaluate the effect of global climate change on the risk of B. miyamotoi worldwide across various scenarios. The habitat suitability index of four primary vector tick species for B. miyamotoi, including Ixodes persulcatus, Ixodes ricinus, Ixodes pacificus and Ixodes scapularis, was projected using a boosted regression tree model, considering multiple shared soci…

The Stable Matching Problem in TBEV Enzootic Circulation: How Important Is the Perfect Tick-Virus Match?

Tick-borne encephalitis virus (TBEV), like other arthropod-transmitted viruses, depends on specific vectors to complete its enzootic cycle. It has been long known that Ixodes ricinus ticks constitute the main vector for TBEV in Europe. In contrast to the wide distribution of the TBEV vector, the occurrence of TBEV transmission is focal and often restricted to a small parcel of land, whereas surrounding areas with seemingly similar habitat parameters are free of TBEV. Thus, the question arises which factors shape this focal distribution of TBEV in the natural habitat. To shed light on factors d…
