A two-step, two-sample Mendelian randomization analysis investigating the interplay between gut microbiota, immune cells, and melanoma skin cancer.

This study aims to rigorously explore the potential causal relationships among gut microbiota (GM), immune cells, and melanoma skin cancer among participants from Europe, where this disease exhibits significant prevalence and profound societal impact. Using the genome-wide association analysis database, a double-sample Mendelian randomization (MR) analysis was drawn upon to investigate GM, immune cells, and melanoma skin cancer. The inverse variance weighted approach was applied to estimate the causal connections among these variables. A two-step MR analysis was employed to quantitatively gaug

The Unified Human Virome Database: A toolkit for expanded human virome analysis.

Current approaches for computationally analyzing viruses within human microbiomes often rely on databases largely composed of fragmented viral genomes from gastrointestinal samples, limiting identification of viruses exclusively found outside the gastrointestinal tract and analyses requiring high-quality genomes. To address these issues, we created the Unified Human Virome Database (UHVDB), comprising 575,497 high-quality, annotated viral genomes from human gastrointestinal, airway, skin, and urogenital sample metagenomes. We developed an associated toolkit that uses UHVDB to characterize viru

Overlapping features of atopic dermatitis and alopecia areata: from pathogenesis to treatment.

Atopic dermatitis (AD) and alopecia areata (AA) have traditionally been regarded as inflammatory dermatoses with independent pathogenic mechanisms, with the former mostly categorized as a type 2 inflammatory disease and the latter as a type 1 inflammatory disease. However, immunologic studies have shown that the immunologic properties of AD and AA do not strictly follow the traditional classification. Both diseases are associated with systemic Th1, Th2, Th17, and Th22 cytokine imbalances, shared genetic susceptibility loci, overlapping immune pathways, and microbiome-mediated modulation of ski

Antifungal Monotherapy in Acne: Topical Ketoconazole 2% Cream Versus Adapalene Gel in Treating Acne Vulgaris: A Comparative Study.

BACKGROUND: Acne is a widely prevalent disorder, with a largely negative impact on the patient's quality of life. Bacterial etiology and the role of Cutibacterium acne have always been incriminated in in acne pathogenesis. However, in the last decade, deeper understanding of skin microbiome postulates a possible role of other pathogens like Malassezia spp. ketoconazole has shown efficacy to both pathogens. OBJECTIVES: To study the efficacy of topical ketoconazole 2% in the treatment of acne vulgaris (AV) compared to the standard treatment with adapalene. METHODS: Fifty-eight acne patients in…

Chronic wound microenvironment mediates selection of biofilm-forming multi drug resistant Staphylococcus epidermidis with capability to impair healing.

Venous leg ulcers (VLU) are the most common chronic wounds characterized by bacterial biofilms and perturbed microbiome. Staphylococcus epidermidis is primarily known as skin commensal beneficial for the host, however, some strains can form biofilms and cause infections. By employing shotgun metagenomic sequencing we show that genetic signatures of antimicrobial resistance, adhesion and biofilm formation in VLU isolates correlate with in vitro bacterial traits. We demonstrate that the capability of chronic wound isolates to form biofilms and elicit IL-8 and IL-1β expression in human ex vivo wo

The Role of Vitamin C and Vitamin D in the Pathogenesis and Therapy of Periodontitis-Narrative Review.

Periodontitis is a common disorder affecting the bone and soft tissues of the periodontal complex. When untreated, it may lead to severe mobility or even loss of teeth. The pathogenesis of periodontitis is complex, with crucial factors being chronic inflammation in gingival and periodontal tissues and oral microbiome alterations. However, recent studies highlight the alleged role of vitamins, such as vitamin C (VitC) and vitamin D (VitD), in the development of the disease. VitC regulates numerous biochemical reactions, but foremost, it is involved in synthesizing collagen. It was reported that

Post-Mortem Microbiology: Retrospective Analysis of Infections Caused by Enterococcus Strains.

Post-mortem microbiological tests are one of the basic methods for diagnosing the etiology of infections in forensic pathology. One of the major groups of microorganisms abundant in various parts of the human body during life and after death is Enterococcus spp. Depending on the area of the body involved and the patient's condition, enterococci can be considered to be a microbiome, transient flora or a pathogen responsible for infection. The data used for the analysis were 12 medico-legal autopsy and microbiological reports. Enterococcus spp. was isolated in 10 out of 12 cultures of blood samp

Effect of Cigarette Smoke on Gut Microbiota: State of Knowledge.

Cigarette smoke is a representative source of toxic chemical exposures to humans, and the adverse consequences of cigarette smoking are mediated by its effect on both neuronal and immune-inflammatory systems. Cigarette smoking also is a major risk factor for intestinal disorders, such as Crohn's disease and peptic ulcer. On the other hand, cigarette smoking is protective against developing ulcerative colitis. The effects of cigarette smoking on intestinal disorders include changes in intestinal irrigation and microbiome, increases in permeability of the mucosa, and impaired mucosal immune resp

Exploring the Diverse Immune and Genetic Landscape of Psoriatic Arthritis.

Psoriatic arthritis (PsA) is characterized by delays in diagnosis and modest effect of treatment in terms of joint response. An understanding of molecular pathomechanisms may aid in developing diagnostic and prognostic models. Genetic susceptibility (e.g., HLA class I genes, IL-23-related genes) can be responsible for the pattern of psoriatic manifestations and affinity for tissue involvement. Gene expression analysis indicates an inflammatory profile that is distinct for PsA, but disparate across tissues. This has clinical implications, as for example, dual blockade of IL-17A and IL-17F can l

Vitamin D Deficiency in the Gulf Cooperation Council: Exploring the Triad of Genetic Predisposition, the Gut Microbiome and the Immune System.

Vitamin D is a fat soluble secosteroid that is primarily synthesized in the skin upon exposure to Ultraviolet B (UVB) sun rays. Vitamin D is essential for the growth and development of bones and helps in reducing inflammation by strengthening muscles and the immune system. Despite the endless supply of sunlight in the Gulf Cooperation Council (GCC) countries which includes United Arab Emirates, Qatar, Kuwait, Bahrain, Saudi Arabia, and Oman, Vitamin D deficiency in the (GCC) general population at various age groups remains alarmingly high. In parallel runs the increasing prevalence of acute an

A Microenvironment-Responsive Graphdiyne-Iron Nanozyme Hydrogel with Antibacterial and Anti-Inflammatory Effect for Periodontitis Treatment.

Periodontitis is a chronic inflammatory disease caused by dental plaque, which leads to tooth loosening and shifting or even tooth loss. Current treatments, including mechanical debridement and antibiotics, often fail to eradicate recalcitrant biofilms and mitigate excessive inflammation. Moreover, these interventions can disrupt the oral microbiome, potentially compromising long-term treatment outcomes. To address these limitations, an injectable nanoenzyme hydrogel composed of a dopamine (DA)-modified hyaluronic acid (HA) scaffold and a graphdiyne-iron (GDY-Fe) complex, named GDY-Fe@HA-DA, e

Gastrointestinal Conditions: Inflammatory Bowel Disease.

Inflammatory bowel diseases (IBDs) comprise a group of conditions thought to be caused by a dysregulated host immune response to the gut microbiome. IBDs, which affect 1.3% of Americans, include Crohn disease (CD), ulcerative colitis (UC), and microscopic colitis. UC and microscopic colitis are limited to the colon, but CD can occur anywhere along the gastrointestinal tract. Gastrointestinal symptoms typically predominate, including diarrhea and abdominal pain, along with systemic symptoms of weight loss, fatigue, night sweats, and fever. However, many patients have extraintestinal symptoms, o

Most self-touches are with the nondominant hand.

Self-touch may promote the transfer of microorganisms between body parts or surfaces to mucosa. In overt videography of a post-graduate office, students spent 9% of their time touching their own hair, face, neck, and shoulders (HFNS). These data were collected from 274,000 s of surveillance video in a Chinese graduate student office. The non-dominant hand contributed to 66.1% of HFNS-touches. Most importantly, mucous membranes were touched, on average, 34.3 (SE = 2.4) times per hour, which the non-dominant hand contributed to 240% more than the dominant hand. Gender had no significant effect o

Diseases Transmitted by Cats.

Humans and cats have shared a close relationship since ancient times. Millions of cats are kept as household pets, and 34% of households have cats. There are numerous diseases that may be transmitted from cats to humans. General modes of transmission, with some overlapping features, can occur through inhalation (e.g., bordetellosis); vector-borne spread (e.g., ehrlichiosis); fecal-oral route (e.g., campylobacteriosis); bite, scratch, or puncture (e.g., rabies); soil-borne spread (e.g., histoplasmosis); and direct contact (e.g., scabies). It is also likely that the domestic cat can potentially

Beyond the dichotomy: understanding the overlap between atopic dermatitis and psoriasis.

Atopic dermatitis and psoriasis have traditionally been considered distinct inflammatory skin diseases with unique pathogenic mechanisms. However, accumulating evidence suggests significant overlap in their immunological pathways, metabolic features, and microbiome characteristics, challenging this conventional dichotomy. This review comprehensively examines the complex relationship between psoriasis and atopic dermatitis, with particular emphasis on their shared and distinct pathogenic mechanisms. We analyze the immunological networks, metabolic pathways, and microbial factors contributing to
