Several drugs targeting the GABAergic system are used in the treatment of epilepsy, but only one drug targeting glutamate receptors is on the market. This is surprising because an imbalance between excitatory and inhibitory neurotransmission lies at the core of the pathophysiology of epilepsy. One possible explanation is that drug development has been directed towards the synthesis of molecules that inhibit the activity of ionotropic glutamate receptors. These receptors mediate fast excitatory synaptic transmission in the central nervous system (CNS) and their blockade may cause severe adverse…

The neuromuscular junction is the point of contact between motor nerve and skeletal muscle, its vital role in muscle function is reliant on the precise location and function of many proteins. Congenital myasthenic syndromes (CMS) are a heterogeneous group of disorders of neuromuscular transmission with 30 or more implicated proteins. The use of animal models has been instrumental in determining the specific role of many CMS-related proteins. The mouse neuromuscular junction (NMJ) has been extensively studied in animal models of CMS due to its amenability for detailed electrophysiological and h

At the onset of puberty, ovarian hormones increase inhibitory tone in the prefrontal cortex. Inhibitory maturation is a hallmark of the initiation of developmental windows of neural plasticity; pubertal hormones may trigger the opening of an adolescent critical period for experience-dependent rewiring of circuits underlying executive function.

Epilepsy is one of the most common neurologic disorders worldwide with no distinguishable cause in 60% of patients. One-third of world's population is infected with Toxoplasma gondii (T. gondii). This intracellular parasite has high tendency to excitable cells including neurons. We assessed seizure susceptibility and involvement of dopaminergic system in male mice with acute and chronic T. gondii infection. Mice were infected by intraperitoneal injection of T. gondii cysts. Acute and chronic stages of infection were determined by quantification of SAG1/BAG1 transcripts and level of repetitive

In vertebrates, all acoustic information transmitted from the inner ear to the central auditory system is relayed by primary auditory afferents (auditory-nerve fibers; ANFs). These neurons are also the most peripheral elements to use action potentials (spikes) to encode the acoustic information. Here, we review what is known about the spiking of ANFs during spontaneous activity, when spike timing might be regarded as largely random, and during stimulation by low-frequency sounds, when spikes are phase locked to the stimulus waveform, a phenomenon generally considered a hallmark of temporal pre

Glutamatergic neural transmission is involved in both neural plasticity and neurodegeneration. This combination of roles could result in ambivalent effects in which excitotoxic neurodegeneration augments neural plasticity in parallel. Neural plasticity can be induced by exposure-based learning (EBL) that resembles timing properties of long-term potentiation (LTP) protocols (i.e., LTP-like learning). Even though it has not been demonstrated so far in animal models that perceptual effects of such stimulation protocols are mediated by typical LTP mechanisms, it has been shown that exposure-based

Nicotine has been reported to increase the intracellular Ca(2+) concentration ([Ca(2+)](i)) in sublingual acini due to neurotransmitter release from nerve terminals associated with the cell preparation (1). However, it is unclear whether or not the same reaction exists in parotid cells. Therefore, we investigated effect of nicotine on Ca(2+) mobilization in digested parotid acini from rats. After removing the parotid gland from Wistar rats, the tissues were minced and digested with collagenase. Then, the intracellular Ca(2+) indicator fura-2 was added to the preparation, and the change in [Ca(

Seizures are the result of a sudden and temporary synchronization of neuronal activity, the reason for which is not clearly understood. Astrocytes participate in the control of neurotransmitter storage and neurotransmission efficacy. They provide fuel to neurons, which need a high level of energy to sustain normal and pathological neuronal activities, such as during epilepsy. Various genetic or induced animal models have been developed and used to study epileptogenic mechanisms. Methionine sulfoximine induces both seizures and the accumulation of brain glycogen, which might be considered as a

The D2 dopamine receptors play a pivotal role in the physiopathology of schizophrenia. Their over-stimulation in the limbic area, resulting from a hyperactivity of the mesolimbic dopaminergic neurones, is linked to positive expressions of the disease, whereas their lack of stimulation in cortical regions, depending on a hypoactivity of mesocortical dopaminergic neurones, underlies the negative expressions of the disease. Concerning the extrapyramidal side effects which emanate from treatments by neuroleptic drugs, they are triggered by the blockade of nigro-striatal dopaminergic transmissions.

Variant sensory branches of the ulnar nerve were encountered during a routine dissection of the palmar aspects of a cadaver hand. The variant branches were distributed to the ulnar half of the index, ulnar half of the middle, and radial half of the ring fingers. The communicating branch between the superficial ulnar branch and the third common digital branch of the median nerve was absent. Distribution of the median nerve sensory branches was normal. The clinical significance of this variation is discussed.

In olfactory bulb slices from young rats, the field potential evoked in the glomerular layer by stimulation in the olfactory nerve layer consisted of two negative components: an early component (N1) which was blocked by bath application of the kainate/amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid (AMPA) receptor antagonist 6-cyano-7-nitroquinoxaline-2,3-dione (CNQX, 10 microM), and a late, prolonged component (N2; duration > or = 350 msec) which was unaffected by CNQX, was enhanced by reduction of Mg2+ in the medium, and was blocked by the N-methyl-D-aspartate receptor antagonist DL-2-ami

1. Cellular mechanisms underlying the enhancement by noradrenaline (NA) of inhibitory postsynaptic currents (IPSCs) were studied at inhibitory synapses in the molecular layer of the cerebellum. IPSCs were obtained from stellate cells in rat cerebellar slices using tight-seal whole-cell recording. 2. Miniature IPSCs (mIPSCs) were recorded in the presence of tetrodotoxin (TTX; 100 nM). NA (10 microM) markedly increased the frequency of mIPSCs, but did not alter their mean amplitude. Bath application of the inhibitor of adenylyl cyclase 9-(tetrahydro-2'-furyl) adenine (SQ 22,536; 300 microM), of

Pharmacologic and electrophysiologic studies over the past 20 years have shown tizanidine to be a potent, central-acting myotonolytic agent that principally affects spinal polysynaptic reflexes. This action arises from agonistic activity of the compound at noradrenergic alpha 2 receptors, resulting in both direct impairment of excitatory amino acid release from spinal interneurons and a concomitant inhibition of facilitatory coeruleospinal pathways. Similar alpha 2-receptor-mediated inhibition of interneuronal activity appears to underlie the additional antinociceptive and anticonvulsant activ

Carpal tunnel syndrome involves classic symptoms of numbness and paresthesias in the radial 3-1/2 digits, most frequently nocturnal, and pain associated with this distribution. Thenar weakness and autonomic dysfunction rarely are seen in this syndrome except in advanced cases. Provocative tests on physical examination such as the wrist flexion test and the local percussion sensitivity test over the median nerve can be extremely helpful in determining and confirming the diagnosis. Nerve conduction velocity and electromyographic studies of the median nerve and its compression can be helpful espe

Metals such as lead, zinc, copper, aluminum and manganese have been implicated in neuropsychiatric disorders. However, until fairly recently the role of iron in brain function was rather obscure, because little attention was paid to its metabolism in the brain. It is now apparent that maintenance of brain iron homoeostasis is important for the normal functioning of his organ. Most of the studies have been directed towards the cognitive and attentional deficit resulting from nutritional iron deficiency. Evidence so far suggests subsensitivity of striatal dopamine neurotransmission. By contrast

The Pulfrich effect is a simply observable visual stereo phenomenon arising when the visual latency for one eye is longer than for the another. In the paper the review of the literature devoted to the application of this phenomenon for the ophthalmological diagnosis is presented. The authors come to the conclusion that the Pulfrich effect can be successfully used in clinical practice as an indicator of many ophthalmological disorders especially of optic neuritis.
