Hepatocellular carcinoma (HCC) is a leading cause of cancer mortality worldwide; molecular biomarkers that reflect hepatitis B virus (HBV)/hepatitis C virus (HCV)-associated tumor biology and predict prognosis or therapeutic vulnerabilities are needed. The research is a secondary analysis of public data (secondary data analysis) based on Gene Expression Omnibus (GEO) and The Cancer Genome Atlas (TCGA) data to identify differentially expressed genes (DEGs) common to HBV-HCC, HCV-HCC, and non-viral HCC datasets. Gambogic acid (GA) exhibits potent antitumor effects through multiple synergistic mechanisms, making it a promising candidate for adjuvant t … orporation of a hybrid membrane derived from tumor cell and macrophage. The dual linkages allow for tumor microenvironment-specific release of both drugs under reductive and acidic conditions. Additionally, the hybrid membrane provides tumor-homing capability and helps evade immune clearance. CM@pGGH induces PANoptosis, a recently characterized form of regulated cell de …

BACKGROUND: Therapy resistance remains a major obstacle in the treatment of solid tumors and accounts for most cancer-related deaths. While tumor-intrinsic mechanisms have been well-studied, the tumor microenvironment (TME) is now recognized as a major driver of resistance through non-genetic, cell-extrinsic signaling. Stromal and immune cells-including fibroblasts, macrophages, endothelial cells, and regulatory immune cells-interact with cancer cells via cytokine signaling, direct contact, and extracellular matrix (ECM) remodeling to promote survival, immune evasion, and therapeutic adaptatio…

PURPOSE OF REVIEW: Head and neck squamous cell carcinoma (HNSCC) develops within a complex tumor microenvironment (TME) that fosters immune evasion and limits the efficacy of immune checkpoint inhibitors (ICIs). Despite advances, durable responses to ICIs remain limited, partly because of stromal barriers, myeloid cell recruitment, and metabolic constraints within the TME.
RECENT FINDINGS: Multiplex IHC/mIF, single-cell and spatial omics, and computational TME profiling reveal recurrent resistance programs, including CAF-driven immune exclusion, chemokine-dependent and hypoxia-dependent recru…

BACKGROUND: Colorectal cancer (CRC) tumor immune microenvironment (TIME) components such as programmed death-ligand 1 (PD-L1) and CD4+T/CD8+T cells play an important role in immunotherapy, which is closely related to the treatment and prognosis of patients. Due to the fact that CRC is relatively insidious and most of it has advanced and metastasis when discovered, it is important to use biopsy specimens to accurately assess the TIME before treatment. However, there are no studies investigating the association of TIME in biopsy and excision specimens and in metastatic colorectal lesions.

Pancreatic cancer exhibits extensive metabolic reprogramming that supports rapid progression and therapeutic resistance, making metabolic vulnerabilities attractive targets for intervention. Here, a tumor microenvironment-responsive nanoreactor (Pht@HMnO2-HA) is designed to induce disulfidptosis in pancreatic cancer through coordinated metabolic interference and redox catalysis. Following CD44-mediated tumor targeting and cellular internalization, the nanoreactor's responsive self-optimization within the tumor microenvironment enables localized release of phloretin, suppressing glucose uptake …

Although immune checkpoint blockade (ICB) therapy has profoundly reshaped the landscape of oncology, T-cell exhaustion (Tex) remains a core challenge in immunotherapy for solid tumors. Current research predominantly focuses on signal transduction and epigenetic regulation, whereas the adaptive alterations and dysfunction of subcellular organelles within T cells under the stress of the tumor microenvironment (TME) have not been systematically elucidated. This review proposes that suborganellar dysfunction serves as a key functional link in the progression of Tex. We systematically explore the r…

Lung cancer (LC) remains a major cause of cancer-related mortality, and current therapeutic strategies are frequently limited by treatment resistance, tumor heterogeneity, and adverse effects. Although apoptosis is a critical mechanism underlying the anticancer activity of chemotherapy and radiotherapy, accumulating evidence indicates that cancer cell fate is also regulated by interconnected processes, including oxidative stress responses, mitochondrial dysfunction, inflammation, immune modulation, and tumor microenvironment (TME) interactions. Melatonin (MLT), an endogenous indoleamine involv…

T cell exhaustion remains a significant barrier to effective adoptive cell therapy in solid tumors. Here, we demonstrate that in vitro treat … t CAL-101-treated T cells preferentially differentiate into progenitor exhausted T cells within the tumor microenvironment. These cells demonstrate enhanced tumor infiltration and upregulate the Cxcl10/Cxcr3 signaling axis. The tumor microenvironment of tumors containing CAL-101-treated T cells show reduced glycolysis, oxidative phosphorylation, and proliferation, while …

Prohibitin 1 (PHB1) and prohibitin 2 (PHB2) are highly conserved, ubiquitously expressed scaffold proteins that play central roles in cellul … tions with transcription factors (E2F1, p53, Sp1) and chromatin modifiers (MLL2, HDAC1). Within the tumor microenvironment, PHBs emerge as critical immunometabolic hubs that influence macrophage polarization, cGAS-STING activation, and sexual dimorphism in immune responses. We summarize the cancer-type-specific expression patterns of PHB1/2 and their prognostic value, a …

Adoptive T cell therapies have markedly improved outcomes in hematologic malignancies but their efficacy in solid tumors can be diminished by a hostile tumor microenvironment that impedes sustained therapeutic responses. Beyond challenges such as limited trafficking and antigen heterogeneity, engineered T cells face suppressive myeloid and stromal populations, inhibitory checkpoint ligand interactions, and metabolically hostile niches that collectively diminish effector function and persistence. To overcome these barriers, a new generation of fusion protein-based costimulatory strategies has e…

BACKGROUND: Osteosarcoma (OS) is a highly aggressive primary bone malignancy in adolescents, with poor prognosis due to limited diagnostic and therapeutic strategies. Mitochondrial dysfunction is a hallmark of cancer, and mitophagy, the selective clearance of damaged mitochondria, critically maintains cellular homeostasis. However, the specific role of mitophagy in shaping the OS tumor microenvironment (TME) at single-cell resolution remains poorly understood. This study aims to systematically characterize mitophagy patterns within the OS TME and investigate their impact on intercellular commu…

The spatial arrangement of tumor-associated macrophages (TAMs) and T cells within distinct microanatomical niches is emerging as a key regulator of clinical outcome in solid tumors. Here, we propose a comprehensive review that integrates spatial insights with functional studies of macrophage and T cell heterogeneity, to elucidate how the physical crosstalk and the localization of these cells in the tumor microenvironment (TME) can impact the prognosis and the response to immunotherapy and, vice versa, how diverse immunotherapies can dynamically reshape the TME immune geography. We first review…

The anti-tumor immune response induced by radiation therapy is typically hindered by intrinsic tumor radioresistance and the insufficient release of endogenous antigens. Pyroptosis, a highly immunogenic form of programmed cell death, is regarded as a key pathway for enhancing the immunological effects of radiotherapy. However, Gasdermin E (GSDME) is under-expressed in most tumor cells, which significantly diminishes the activation of the pyroptosis pathway and confers tumor tolerance to pyroptosis-based therapies. To address this issue, this study designed a biomimetic nanovesicle (pGSDME@TLNV

Classic Hodgkin lymphoma (cHL) is composed of rare malignant Hodgkin and Reed-Sternberg (HRS) cells within a T-cell-rich tumor microenvironment (TME). Epstein-Barr virus (EBV) is present in ∼25% of cases, but its contribution to pathogenesis and immunomodulation remains unclear due to technical barriers. Using complementary spatial proteomics and transcriptomics across multi-institutional cohorts, we systematically map key EBV-linked TME reorganization. EBV-positive cHL exhibits distinct immunological features, including memory CD8 T cell enrichment, heightened T cell dysfunction spatially cor…

Cancer is no longer viewed solely as a consequence of tumor-intrinsic genetic alterations but rather as a disease sustained by a complex and evolving tumor microenvironment (TME). The TME functions as an organized and dynamic ecosystem in which malignant cells interact continuously with immune populations, stromal elements, vascular networks, extracellular matrix components, and soluble mediators. These interactions critically regulate tumor initiation, progression, immune evasion, and metastatic dissemination. This review comprehensively examines the cellular and acellular architecture of the…

Metabolic reprogramming and immunosuppressive tumor microenvironment (TME) are hallmark features driving pancreatic ductal adenocarcinoma (PDAC) progression. Despite the therapeutic potential of targeting immunometabolism, effective strategies remain scarce in clinical practice, likely due to cell-specific metabolic heterogeneity within PDAC TME. Here, we show integration of three algorithms to estimate metabolic fluxomes and pathways using scRNA-seq data, generating a comprehensive cell type-specific metabolic atlas. Leveraging 460 PDAC samples, we establish a TME-metabolism subtyping system,…

Owing to the existence of the blood-brain barrier (BBB), achieving high-efficacy, tumor-selective delivery of therapeutic agents continues to be a pivotal unmet need in the treatment of glioblastoma. Here, based on the finding that CD93 is exclusively up-regulated on glioma-associated vascular endothelial cells (VECs), small extracellular vesicles (sEVs) were modified with insulin-like growth factor-binding protein 7 (IGFBP7), a natural ligand of CD93, to create a delivery platform that can deliver therapeutic agents to glioma-associated VECs with high efficiency. Tumor microenvironment (TME)-induced immunosuppression is a major driver of cancer treatment failure and resistance to immunotherapy. Myeloid-derived suppressor cells (MDSCs) are not only pivotal suppressors of effector immune responses but also central organizers of tumor-supportive metabolic, stromal, and cellular crosstalk. This review systematically summarizes the origin and phenotypic characteristics of MDSCs and the mechanisms governing their recruitment, expansion, and activation within the TME. This review integrates current knowledge of MDSC biology through two complementary framework…
