Huntington's disease (HD) is a neurodegenerative disorder characterized by accumulation of mutant huntingtin protein and significant loss of neurons in striatum and cortex. Citrullinemia type 1 (CTLN1) is a rare inherited disorder caused by mutations in the argininosuccinate synthetase 1 (ASS1) gene, disrupting the urea cycle and leading to toxic ammonia accumulation. Carbonic anhydrase IX (CAIX) is known to be overexpressed in various tumors and plays a significant role in tumor development and progression. A series of 3-sulfonamide benzoate derivatives with a 7-membered azepane ring were synthesized and evaluated for their CAIX inhibitory activities.
