Increased expression of resistin and tumour necrosis factor-alpha in pig adipose tissue as well as effect of feeding treatment on resistin and cAMP pathway.

AIM: Resistin, a novel hormone, is expressed in mouse, rat and human adipose tissue. Its resistance to insulin is controversial. We hypothesized that resistin has the similar roles like those of tumour necrosis factor-alpha (TNF-alpha), a putative mediator of insulin resistance, and they are in common involved in regulating lipid metabolism in vivo. METHODS: We demonstrated TNF-alpha and resistin expression in different growth-stage adipose tissue of genetically lean and obese pigs using semiquantitative reverse transcriptase-polymerase chain reaction and Western blotting analysis. Moreover,

Skin.

Human skin provides a barrier between the host and the physical, chemical, and biological environment. It is also a potential portal of entry for hazardous or infectious agents and a potential target of environmental toxins. Cutaneous vulnerability may take on many forms in the embryo, infant, child, and adolescent. Teratogenic agents may occasionally target skin, as appreciated in the proposed association of the antithyroid medication methimazole, with the congenital malformation known as aplasia cutis congenita. Percutaneous absorption of topically applied substances and the potential for re

Pharmacokinetics of chlorpromazine and key metabolites.

A study was carried out in 11 healthy young men to investigate the pharmacokinetics of chlorpromazine (CPZ) after a bolus intravenous (i.v.) dose (10 mg) and three single oral doses (25, 50 and 100 mg), with a washout period of two weeks between doses. Plasma levels of CPZ, CPZ N-oxide (CPZNO), CPZ sulfoxide (CPZSO) and both free and conjugated 7-hydroxy-CPZ (7-HOCPZ) were measured by extraction radioimmunoassays. CPZ exhibited multicompartmental pharmacokinetics in most subjects. There was wide between-subject variability in half life (11.05 h), volume of distribution (1215 l), volume of dist

Generation of PM2 DNA breaks in the course of reduction of chromium(VI) by glutathione.

The carcinogen chromate is efficiently taken up and reduced to chromium(III) compounds by various biological systems. To test the possible DNA damage induced in the course of chromium(VI) reduction, we used a combination of chromate with the reductant glutathione (GSH) as well as a green complex of chromium(V), which is formed in the reaction of chromate with GSH. The combination of chromate and glutathione was found to cause single-strand breaks in supercoiled circular DNA of the bacteriophage PM2. The green chromium(V) complex Na4(GSH)4Cr(V).8H2O, prepared from chromate and glutathione, also

Single-Cell Detection and Quantification of Metabolically Active Bacteria in Soil Using Heavy-Water Labeling and Raman Microspectroscopy.

Only microorganisms that are metabolically active during a defined observation window contribute directly to ongoing soil processes, yet this active fraction is difficult to quantify. Cultivation excludes uncultured taxa, deoxyribonucleic acid (DNA)-based methods primarily describe community membership and functional potential, and bulk assays obscure cell-to-cell variation. This protocol combines heavy-water (D₂O) labeling with single-cell Raman microspectroscopy, here termed Raman-deuterium isotope probing (Raman-DIP), to detect and quantify bacterial anabolic activity during labeling in soi

Robustness of interstitial photodynamic therapy treatment planning under power and positional uncertainties in light delivery.

While computer simulations can accurately model light dose delivery in interstitial photodynamic therapy (iPDT), predicting individual clinical outcomes remains difficult. In addition to biological uncertainties, inaccuracies in light delivery must be considered. Using simulations on virtual brain tumour (glioblastoma) models, we analyze two sources of uncertainty: light source power variations of [Formula: see text], [Formula: see text], and [Formula: see text], and positional deviations during source insertion, modelled as angular errors producing up to [Formula: see text] displacement. Sex-Specific Structural Vulnerability in Alzheimer's Disease: Insights from APOE ε 4-Negative Patients.

Background: The interaction between sex, APOE ε4 status, and clinical progression in Alzheimer's Disease (AD) remains a subject of debate. While females are often considered at higher risk for AD, the underlying structural neuroanatomical trajectories and how they are modulated by genotype are not fully elucidated. This study aims to evaluate how sex and the APOE ε4 genotype interact to influence longitudinal brain atrophy across three clinical groups. Methods: We analyzed longitudinal data from 2400 participants from the Alzheimer's Disease Neuroimaging Initiative (ADNI), stratified by clinic

Circadian-Related Hypothalamic Structure Differs by Chronotype in Bipolar Disorder.

BACKGROUND: Circadian rhythm disturbances represent a core feature of bipolar disorder (BD), with evening chronotype as a marker for poorer outcomes. We hypothesized that BD psychopathology combined with evening chronotype would be associated with structural alterations in circadian-related hypothalamic regions, particularly the suprachiasmatic nucleus (SCN), specific to BD relative to other psychiatric diagnoses. METHODS: We investigated structural neuroimaging data from the UK Biobank (113 BD, 205 major depressive disorder, 91 psychotic disorders, 199 healthy control). The SCN-containing an

An Overview of the Impact of the Menstrual Cycle on Nutrient Metabolism: An Integrative Perspective.

The menstrual cycle represents a dynamic infradian rhythm characterized by coordinated fluctuations in ovarian steroids that extend beyond reproductive function and influence systemic metabolism. This narrative review synthesizes current evidence on how menstrual cycle phase modulates energy balance, macronutrient metabolism, micronutrient handling, and responses to dietary bioactive compounds. Across phases, small-to-moderate but consistent differences emerge in energy intake, resting energy expenditure, substrate utilization, and protein turnover, with a tendency toward increased energy inta

Prenatal Exposure to Persistent Organic Pollutants Is Associated with Altered Lipid and Metabolomic Profiles in Mothers and Newborns.

Maternal exposure to persistent organic pollutants (POPs) has been linked with adverse pregnancy and birth outcomes, yet the underlying biological pathways involved remain poorly characterized. In this study, we examined associations between POP concentrations in maternal plasma and cord blood and metabolomic and lipidomic profiles of pregnant women and their newborns and further assessed relationships with neonatal birth weight. A total of 257 mother-infant dyads were included. Concentrations of polychlorinated biphenyls (PCBs) and organochlorine pesticides (OCs) were quantified using gas chr

Differential Effects of BMI Adjustment on Ferritin Levels in Anemic Patients.

BACKGROUND: The prevalence of obesity and anaemia is consistently increasing in India. Obesity is a chronic inflammatory state and is associated with dysregulation of iron homeostasis. Ferritin, a marker for cellular iron stores, is also an acute phase reactant that remains elevated during inflammation. Previous studies have used inflammation-adjusted serum ferritin to address this. This current study aims to calculate body mass index (BMI)-adjusted ferritin using a regression correction and evaluate its effects across gender and BMI category in anemic patients. METHODS: De-identified data we

Blue magnetophosphene perception under subthreshold magnetic stimulation in a human retinal degeneration model.

INTRODUCTION: Extremely low-frequency magnetic fields (ELF-MF; <300 Hz) can induce visual percepts known as magnetophosphenes through weak, subthreshold electric fields in neural tissue. Although magnetophosphene perception has been characterized in healthy observers, the anatomical locus of interaction (retinal vs. cortical) remains incompletely constrained, particularly under conditions of severe retinal dysfunction. METHODS: Here, we employed a controlled single-subject perturbation framework to mechanistically dissociate candidate sites of ELF-MF interaction using systematic variation of

Genome-wide association studies identify genetic determinants of synucleinopathy biomarkers.

OBJECTIVE: α-synucleinopathies are clinically and biologically heterogeneous disorders lacking reliable biomarkers to assist with early diagnosis, disease progression, patient stratification, and therapeutic targeting. Genetic variation is known to impact biomarker levels, influencing their utility and interpretation in research and clinical settings. We aimed to identify common genetic modulators of biomarker levels implicated in α-synucleinopathy pathogenesis. METHODS: Genome-wide association studies (GWASs) were conducted on 63 CSF, plasma, and urine biomarkers in 581 individuals from the

Genetic variants associated with chronic fatigue syndrome predict population-level fatigue severity and actigraphic measurements.

STUDY OBJECTIVES: The diagnosis of myalgic encephalomyelitis/chronic fatigue syndrome (CFS) is based on a constellation of symptoms which center around fatigue. However, fatigue is commonly reported in the general population by people without CFS. Does the biology underlying fatigue in patients with CFS also drive fatigue experienced by individuals without CFS? METHODS: We used UK Biobank actigraphy data to characterize differences in physical activity patterns and daily temperature rhythms between participants diagnosed with CFS compared to controls. We then tested if single nucleotide varia

A novel high-entropy TiVCrMoC3Tx assisted LDI MS for serum metabolic fingerprint in rheumatoid arthritis.

Worldwide, rheumatoid diseases account for approximately 37.57/100,000 disability-adjusted life years (DALYs). Early diagnosis of rheumatoid arthritis (RA) can effectively reduce associated risks and improve quality of life. To achieve this, accurate and rapid advanced tools are required. Herein, TiVCrMoC3, a novel high-entropy two-dimensional (2D) carbide MXene that exhibits excellent electrical conductivity, is prepared and applied to surface-assisted laser desorption ionization mass spectrometry (SALDI-MS) analysis for the first time. TiVCrMoC3 demonstrates superior performance in small-mol

Disability in medieval Poulton (Cheshire): A case of hand amputation.

OBJECTIVE: Amputations are rarely identified in the British archaeological record from the Middle Ages, with survival following hand removal particularly uncommon. This study presents a case of a healed left-hand amputation from the medieval cemetery at Poulton (Cheshire). MATERIALS: The study focuses on Skeleton 861, recovered from the Poulton Chapel burial ground, part of a broader assemblage of over 800 individuals dated to the 13th-16th centuries CE. METHODS: Standard osteological techniques were used to establish the biological profile, while radiographic imaging and macroscopic analysi

Optimal transport reveals immune perturbation and fingerprints over time in COVID-19 vaccination.

Mass cytometry enables high-throughput characterization of heterogeneous cell populations at single-cell resolution, using metal isotopes to capture cellular signals and avoiding the spectral overlap common in flow cytometry. Despite advancements, conventional data analysis often focuses on manual gating or clustering within specific samples, overlooking disparities across subjects or biological samples. To address this gap, we propose a novel framework that treats the cell-by-protein matrix as a high-dimensional distribution, using Quantized Optimal Transport (QOT) to quantify distances betwe

A single-cell cytokine dictionary of human peripheral blood.

Cytokines orchestrate immune responses, yet we still lack a comprehensive understanding of their specific effects across human immune cells due to their pleiotropy, context dependence and extensive functional redundancy. Here, we present a Human Cytokine Dictionary, created from high-resolution single-cell transcriptomes of 9,697,974 human peripheral blood mononuclear cells (PBMC) from 12 donors stimulated in vitro with 90 different cytokines. We describe donor-specific response variation and uncover robust consensus cytokine signatures across individuals. We then delineate similarities betwee
