From the fat body to the hemolymph: Profiling tick immune and storage proteins through transcriptomics and proteomics.

Ticks are blood-feeding arachnids that are known to transmit various pathogenic microorganisms to their hosts. During blood feeding, ticks activate their metabolism and immune system to efficiently utilise nutrients from the host's blood and complete the feeding process. In contrast to insects, in which the fat body is known to be a central organ that controls essential metabolic processes and immune defense mechanisms, the function of the fat body in tick physiology is still relatively unexplored. To fill this gap, we sought to uncover the repertoire of genes expressed in the fat body associa

Prevalence and Potential Risk Factors for Bartonella Infection in Tunisian Stray Dogs.

Bartonellae are blood-borne and vector-transmitted pathogens, some are zoonotic, which have been reported in several Mediterranean countries. Transmission from dogs to humans is suspected, but has not been clearly demonstrated. Our objectives were to determine the seroprevalence of Bartonella henselae, Bartonella vinsonii subsp. berkhoffii, Bartonella clarridgeiae, and Bartonella bovis (as a proxy for Candidatus Bartonella merieuxii) in stray dogs from Tunisia, identify the Bartonella species infecting the dogs and evaluate potential risk factors for canine infection. Blood samples were collec

Rickettsia infection in dogs and Rickettsia parkeri in Amblyomma tigrinum ticks, Cochabamba Department, Bolivia.

Only few published data are available on ticks and tick-borne zoonotic pathogens in Bolivia. To evaluate rickettsial seroprevalence and infection in dogs and ticks, during February-April 2007, we collected whole blood, sera, and ticks from dogs living in the rural, peri-urban, and urban areas of Cochabamba, Bolivia. Dog sera were subjected to enzyme-linked immunosorbent assay test to detect IgG antibodies against Rickettsia rickettsii and 68.2% of samples were found to be positive (n = 30; 95% confidence interval [CI]: 52.4-81.4). Blood samples and ticks were tested using polymerase chain reac

[Equine granulocytic anaplasmosis (EGA): Case description and overview of the epidemiological situation with focus on Germany].

Equine granulocytic anaplasmosis (EGA) is a tick-borne disease caused by Anaplasma (A.) phagocytophilum. In Central, Northern, Western, and Eastern Europe, ticks of the Ixodes (I.) ricinus/I. persulcatus complex, in Germany mainly I. ricinus, are considered as vectors. Ixodes ricinus ticks show peaks of activity during the spring and summer months, particularly April to July. Changing climatic conditions, however, have meanwhile led to a year-round risk of tick exposure and thus infections with A. phagocytophilum. The presented case report underlines the seasonal peak phase of the infection ri

Closing the sepsis gap: from molecular mechanisms to scalable bedside care.

Recent estimates point to roughly 49 million cases and 11-13 million deaths annually due to sepsis, with a disproportionate burden borne by low- and middle-income countries. Three decades of progress, summarised in the recently updated Surviving Sepsis Campaign guidelines, have refined how clinicians screen for, resuscitate, and treat patients with sepsis. Yet, persistent uncertainty surrounds early identification, pathogen detection, hemodynamic targets, adjunctive therapies, and the long-term burden carried by survivors. This Collection, Sepsis: Treatment, intervention, mortality, brings tog

Neglected vector-borne bacterial diseases and arboviruses in the Mediterranean area.

Arthropod vectors can transmit pathogenic microorganisms from one vertebrate to another during their blood meal. Although some vector-borne diseases have been eradicated in the Mediterranean area, such as malaria and dengue, recent endemic microorganisms (Toscana virus, Rickettsia spp.) remain neglected even though they cause many more cases. New diagnostic tools and innovative tools for the identification and characterization of vector species and microorganisms have been developed at IHU Méditerranée Infection, either internally or through collaborative and integrated projects. We have detec

Characterization of Culicoides and mosquito fauna at the National Zoological Garden of Rabat, Morocco.

Zoos are considering to be essential places for the conservation of wild animal species. It is essential to prevent their infection by pathogens especially for those belonging to threatened or extinct species. Zoo captive animals are susceptible to several Culicoides and mosquito borne-viruses. In order to further evaluate the risk of pathogen transmission in zoos, it is essential to identify the presence of potential vector species, as well as the animals bitten by those vectors. For this purpose, Culicoides and mosquito species composition was investigated in ten sites from March to June 202

Vector-borne and zoonotic diseases of dogs in North-west New South Wales and the Northern Territory, Australia.

BACKGROUND: Vector-borne diseases of dogs in Australian Aboriginal communities are relatively unexplored. These dogs represent a unique group with variable ecto- and endo-parasitic burdens, nutritional stresses and a general lack of veterinary intervention. We investigated haemoprotozoal and bacterial pathogen prevalences in relation to erythrocyte and platelet numbers in dogs from North-West New South Wales (N-W NSW) and the Northern Territory (NT; Central Australia). METHODS: Real-time PCR (qPCR) amplification of Anaplasma platys, Babesia vogeli, Mycoplasma haemocanis, Candidatus Mycoplasma

Developmental regulation and extracellular release of a VSG expression-site-associated gene product from Trypanosoma brucei bloodstream forms.

Trypanosomes evade host immunity by exchanging variant surface glycoprotein (VSG) coats. VSG genes are transcribed from telomeric expression sites, which contain a diverse family of expression-site-associated genes (ESAGs). We have discovered that the mRNAs for one ESAG family, ESAG9, are strongly developmentally regulated, being enriched in stumpy forms, a life-cycle stage in the mammalian bloodstream that is important for the maintenance of chronic parasite infections and for tsetse transmission. ESAG9 gene sequences are highly diverse in the genome and encode proteins with weak similarity t

Human immunodeficiency virus transmission and surgeons: update.

Concerns of patients and health care providers regarding disease transmission in the health care setting are not unique to the current human immunodeficiency virus (HIV) epidemic. The data show that the risk to health care workers (HCWs) from patients is far greater than the risk to patients from HCWs. Despite the high frequency of contact with potentially infected fluids, surgeons appear to have a low occupational risk of acquiring HIV infection. Further, the actual risk of surgeon-to-patient transmission appears to be extremely small. Recommendations that HIV-infected surgeons restrict their

Intrapartum colonization with Streptococcus pneumoniae, early-onset sepsis and deficient specific neonatal immune responses.

BACKGROUND: Intrapartum colonization with Streptococcus pneumoniae (S. pneumoniae) is a rare but important risk factor for severe courses of early-onset sepsis (EOS) in the newborn, as underlined in the case of a preterm infant born after 32 weeks of gestation described here. One potential explanation could be an immature immune response of the neonate to S. pneumoniae, however, immunological data in term and preterm infants are scarce. METHODS: To determine the neonatal immune responses to S. pneumoniae, flow-cytometry analysis of the cytokine production by CD14+ cells was performed after fu

Molecular analysis of Ixodes rugicollis, Candidatus Neoehrlichia sp. (FU98) and a novel Babesia genotype from a European badger (Meles meles).

The European badger (Meles meles) is a widespread mammal in most countries of the European continent, with increasingly recognized veterinary/medical importance owing to its preferred habitats (including pastures and urban environments), broad spectrum of food items, and role as a game hunting target. However, ticks and tick-borne pathogens associated with badgers are only partly known, and most of them have not yet been analysed with molecular biological methods The aim of this study was to perform molecular taxonomic analysis of ticks collected from a road-killed European badger, as well as

An integrated IoT microfluidic platform for vector-borne pathogen detection via closed-loop valve control and dual-mode fluid handling.

Vector-borne diseases, including arboviral infections and malaria, pose a major global health threat and underscore the need for rapid, accurate, and multiplex nucleic acid detection technologies for clinical diagnosis and surveillance. However, previous studies on reagent-preloaded and switching valve-based microfluidic cartridge platforms have insufficiently addressed several key engineering challenges, particularly the reliable control of multichannel switching valves and the inherently critical requirement for uniform reagent distribution across multiple reaction zones. These limitations a

Dynamic Changes in Host Immune Response During Crimean-Congo Hemorrhagic Fever and Severe Fever with Thrombocytopenia Syndrome in Mice.

Crimean-Congo hemorrhagic fever virus (CCHFV) and severe fever with thrombocytopenia syndrome virus (SFTSV) are tick-borne pathogens that cause severe illness and high mortality. Early diagnosis is critical, particularly in resource-limited settings, to enable timely intervention. Host gene expression profiling offers a promising approach to identify potential biomarkers for early detection, disease staging, and logical treatment decision-making. Using a transient IFN-α/β receptor-suppressed mouse model, we performed targeted transcriptomic analysis on blood samples collected at 2, 3, and 4 da
